Özet
Lenfomalar bağışıklık sistemi hücrelerinde gelişen kan hastalıklarıdır, vücudun patojenlerle savaşan iletişim ağı olan lenfatik sistemde gelişen kanserlerdir. Germline varyasyonlar, pediatrik kanserlerin %10'unda görülmektedir ve tedavi direnci ya da tekrarlayan malignitelere yol açabilmektedir. Hodgkin lenfoma tanısı almış olgumuzun germline örneğinde, DELTEX3L'de patojenik bir varyasyon tespit edilmiştir. Lenfomaya yatkınlıkla ilişkili olabileceğini düşündüğümüz DELTEX3L genindeki varyasyon, genin C terminal bölgesinde bulunmaktadır. DELTEX3L, E3 ubikuitin ligaz olarak görev yapmaktadır ancak bu genin C terminal bölgesinin fonksiyonu tam olarak bilinmemektedir. Bu çalışma kapsamında CRISPR-Cas9 sistemi kullanılarak DELTEX3L geninin C terminal ucu knock-out yapılarak delesyona uğratılmıştır. DTX3L-∆DTC homozigot ve heterozigot varyasyona sahip hücre modelleri oluşturulmuştur ve DTC domaininin genin fonksiyonuna olan etkisi incelenmiştir. Sanger dizileme ile DNA düzeyinde ve Western blot yöntemi ile protein düzeyindeki değişim incelenmiştir. Ayrıca, DTX3L-∆DTC hücrelerde transkriptom analizi yapılmıştır. DTX3L-DTC kaybı muhtemel NMD mekanizması üzerinden DTX3L protein anlatımının kaybolmasına neden olduğu tespit edilmiştir. DTX3L protein kaybı beraberinde PARP9 protein anlatımını da baskılamıştır. Transkriptom çalışmaları doğrultusunda, DTX3L-DTC kaybının TP53 yolağı aktivasyonu ile apoptozu indüklediği gösterilmiştir. Bu sonuçlar doğrultusunda, kanser çalışmaları için DTX3L yeni bir hedef molekül olabilir ve DTX3L-PARP9 kompleksinin baskılanması apoptozu indükleyerek proliferasyonu azaltabilir. Bununla beraber DTX3L'nin lenfomagenezle olan ilişkisini açığa çıkartmak için ilave fonksiyonel çalışmalara ihtiyaç vardır.

Lymphomas are blood disorders that are developed in immune cells, they are the cancers that grow in the body's fighting cells: the lymphatic system. Lymphomas can stem from the progenitors that govern the hematopoiesis process as well as mature lymphoid cells. They show a highly heterogeneous clinical picture and treatment response depending on the groups of cells they originate from. Germline variations are observed in 10% of pediatric cancers and can lead to treatment resistance or recurrent malignancies. A pathogenic variation in DELTEX3L was detected in the germline sample of our patient diagnosed with Hodgkin lymphoma. The variation in the DELTEX3L gene, which we think may be associated with a predisposition to lymphoma, is located in the C terminal region of the gene. DELTEX3L acts as an E3 ubiquitin ligase, but the function of the C terminal region of this gene is not fully understood. In this study, we utilized the CRISPR-Cas9 system to knock out the C-terminus of the DELTEX3L gene, leading to its deletion. We generated DTX3L-∆DTC homozygous and heterozygous variant cell models to investigate the impact of the DTC domain on gene function. Changes at the DNA level were examined by Sanger sequencing and at the protein level by Western blot. Additionally, we conducted transcriptome analysis on the DTX3L-∆DTC cells. Loss of DTX3L-DTC led to the loss of DTX3L protein expression, possibly through the NMD mechanism. We showed that DTX3L protein loss also suppressed PARP9 protein expression. Transcriptome studies revealed that DTX3L-DTC loss induced apoptosis through activation of the TP53 pathway. In conclusion, DTX3L represents a promising novel target molecule for cancer research. The inhibition of the DTX3L-PARP9 complex has the potential to induce apoptosis and reduce cell proliferation, highlighting its therapeutic potential in cancer treatment strategies. However, additional functional studies are needed to elucidate the relationship between DTX3L and lymphomagenesis.
