Özet

Bitki kaynaklı eksozomlar, biyoteknoloji ve tıp alanlarında önemli bir araştırma konusu haline gelmiştir. Eksozomlar, hücreler arası iletişimi sağlayan, biyoaktif molekülleri taşıyan mikroskobik biyoveziküllerdir. İlk kez hayvan hücrelerinde keşfedilen eksozomlar, 2007 yılında bitki hücrelerinden de üretilebildiği gösterilerek bitkilerdeki potansiyel biyolojik işlevleri keşfedilmiştir. Bu tez çalışmasında, safran bitkisinden (Crocus sativus) izole edilen eksozomların, Fare GC-1spg spermatogoniyal hücreleri üzerinde biyolojik etkileri ve sisplatin kaynaklı hücre hasarı üzerine tedavi edici rolü araştırılmıştır. Safran ekstratından ultrasantrifüj yöntemiyle izole edilen eksozomlar taramalı elektrom mikroskopta görüntülendi ve BCA testi ile protein miktar tayini yapıldı. Hücreler 4 saat 5 µg/mL sisplatine maruz bırakıldı ve inkübasyon sonrası farklı konsantrasyonlarda eksozom (5, 10, 20, 30, 40 ve 50 ng/mL) ile tedavi edildi. Hücre proliferasyonu üzerine etkisi (3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolyum bromür, (MTT) ile değerlendirildi. Hücre döngüsü ve apoptoz üzerine etkisi flow sitometri ile araştırıldı. Hücre membran hasarı üzerine etkileri PI boyama yapılarak floresans mikroskopta görüntülendi. Nrf2 ve NF-κB düzeyleri ise Elisa ile ölçüldü. Bu çalışmanın sonuçları, safran kaynaklı eksozomların GC-1spg spermatogoniyal hücrelerinde sisplatin kaynaklı hücresel hasara karşı önemli koruyucu etkiler gösterdiğini ortaya koymaktadır. Eksozom tedavisi, hücre canlılığını artırmış, nekroz ve apoptoz oranlarını azaltmış ve hücresel antioksidan savunma mekanizmalarını iyileştirmiştir. Akış sitometrisi ve hücre döngüsü analizleri, safran eksozomlarının genotoksik stresi hafiflettiğini ve sisplatin tarafından bozulan hücre döngüsünü yeniden düzenlediğini göstermiştir. Ayrıca, safran eksozomlarının NF-κB ve Nrf2 gibi temel oksidatif stres ve enflamasyon belirteçlerinin ekspresyonunu düzenlediği, böylece oksidatif hasarı hafiflettiği ve hücresel stres tepkilerini modüle ettiği tespit edilmiştir. Bu bulgular, safran kaynaklı eksozomların genotoksik ve oksidatif hasarın azaltılmasında üreme sağlığı uygulamaları için umut vadeden bir terapötik ajan olarak potansiyelini vurgulamaktadır.

Plant-derived exosomes have become an important research topic in the fields of biotechnology and medicine. Exosomes are microscopic biovesicles that provide intercellular communication and carry bioactive molecules. Exosomes, which were first discovered in animal cells, were shown to be produced from plant cells in 2007 and their potential biological functions in plants were discovered. In this thesis, the biological effects of exosomes isolated from saffron plant (Crocus sativus) on mouse GC-1spg spermatogonial cells and their therapeutic role on cisplatin-induced cell damage were investigated. Exosomes isolated from saffron extract by ultracentrifugation were visualised under scanning electromicroscope and protein quantification was performed by BCA assay. Cells were exposed to 5 µg/mL cisplatin for 4 hours and treated with different concentrations of exosomes (5, 10, 20, 30, 40 and 50 ng/mL) after incubation. The effect on cell proliferation was evaluated by (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, (MTT). The effect on cell cycle and apoptosis was investigated by flow cytometry. The effects on cell membrane damage were visualised by PI staining under fluorescence microscope. Nrf2 and NF-κB levels were measured by Elisa. The results of this study revealed that saffron-derived exosomes showed significant protective effects against cisplatin-induced cellular damage in GC-1spg spermatogonial cells. Exosome treatment increased cell viability, decreased necrosis and apoptosis rates and improved cellular antioxidant defence mechanisms. Flow cytometry and cell cycle analyses showed that saffron exosomes alleviated genotoxic stress and re-regulated the cell cycle disrupted by cisplatin. Furthermore, saffron exosomes were found to regulate the expression of key oxidative stress and inflammation markers such as NF-κB and Nrf2, thereby alleviating oxidative damage and modulating cellular stress responses. These findings highlight the potential of saffron-derived exosomes as a promising therapeutic agent for reproductive health applications in reducing genotoxic and oxidative damage.
