Özet
Amaç Diş implantlarının uzun dönem başarısında osseointegrasyonun yanı sıra peri-implant yumuşak doku entegrasyonu kritik bir öneme sahiptir. Bu çalışmada, iki boyutlu (2B) karbon esaslı malzemelerden grafen oksit (GO), indirgenmiş grafen oksit (rGO) ve azot katkılı grafen oksit (N-GO) ile kaplanmış titanyum yüzeylerinin insan dişeti fibroblast (HGF-1) hücrelerinin proliferasyonu ve adezyonuna etkileri incelenmiştir. Gereç ve Yöntem GO, Hummers yöntemiyle; rGO, askorbik asit ile GO'nun indirgenmesi yoluyla; N-GO ise kronoamperometrik yöntemle sentezlenmiştir. Her bir malzeme 0,25 mg/ml, 0,5 mg/ml ve 1 mg/ml konsantrasyonlarında hazırlanmış ve püskürtme yöntemiyle titanyum disklerin yüzeyine uygulanmıştır. HGF-1 hücrelerinin canlılığı 3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolyum bromür (MTT) testi ile, adezyon proteinlerinden biri olan vinkülinin ekspresyonu ise Western blot analiziyle değerlendirilmiştir. Örnek yüzeylerinin karakterizasyonu yapılmış ve bu özelliklerin HGF-1 hücreleriyle ilişkisi analiz edilmiştir. Bulgular 0,5 mg/ml ve 1 mg/ml konsantrasyonlarındaki N-GO ile kaplanmış titanyum yüzeylerinde, 72.saatte HGF-1 hücre proliferasyonu kontrol grubuna kıyasla istatistiksel olarak anlamlı düzeyde artmıştır (p <0,01). 0,5 mg/ml konsantrasyonundaki rGO ile kaplanmış titanyum yüzeylerinde ise HGF-1 hücre proliferasyonu kontrol grubuna göre anlamlı düzeyde azalmıştır (p <0,05). 0,5 mg/ml konsantrasyonundaki N-GO ile kaplanmış titanyum yüzeylerinde vinkülin ekspresyonu kontrol grubuna göre istatistiksel olarak anlamlı düzeyde artmıştır (p <0,0001). Sonuçlar 2B karbon esaslı malzemelerin türü ve konsantrasyonu, HGF-1 hücrelerinde farklı biyolojik yanıtlara yol açmaktadır. 0,5 mg/ml konsantrasyonunda hazırlanan N-GO'nun, titanyum implantların transmukozal yüzeylerinin kaplanması için potansiyel bir malzeme olabileceği düşünülmektedir. Elde edilen bulguların olası klinik yansımalarının ileri düzeyde in vivo çalışmalarla desteklenmesi gerekmektedir.

Objective Peri-implant soft tissue integration is as critical as osseointegration for the long-term success of dental implants. This study aimed to investigate the effects of titanium surfaces coated with two-dimensional (2D) carbon-based materials—graphene oxide (GO), reduced graphene oxide (rGO), and nitrogen-doped graphene oxide (N-GO)—on the proliferation and adhesion of human gingival fibroblast (HGF-1) cells. Materials and Methods GO was synthesized using the Hummers method; rGO was obtained by the chemical reduction of GO with ascorbic acid; and N-GO was synthesized via a chronoamperometric method. Each material was prepared at concentrations of 0.25 mg/ml, 0.5 mg/ml, and 1 mg/ml and applied to the surface of titanium discs using a spraying technique. HGF-1 cell viability was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay, while vinculin expression, as a marker of adhesion, was assessed by Western blot analysis. Surface characterization was performed, and the relationship between surface properties and HGF-1 cell behavior was analyzed. Results Titanium surfaces coated with 0.5 mg/ml and 1 mg/ml N-GO showed a statistically significant increase in HGF-1 cell proliferation at 72 hours compared to the control group (p < 0.01). In contrast, surfaces coated with 0.5 mg/ml rGO exhibited significantly reduced proliferation (p <0.05). Moreover, vinculin expression was significantly higher in HGF-1 cells cultured on 0.5 mg/ml N-GO-coated surfaces compared to the control group (p < 0.0001). Conclusion The type and concentration of 2D carbon-based materials induce distinct biological responses in HGF-1 cells. N-GO at 0.5 mg/ml appears to be a promising coating material for the transmucosal surfaces of titanium implants. The findings of this study suggest that further advanced in vivo studies are needed to support the potential clinical implications.
