Özet
Amaç: Bu çalışmanın amacı meme kanseri alt tiplerinde hedef peptit olan P18-4'e bağlanma gösteren ortak bir reseptörün varlığının CRISPR/Cas9 sistemi ile belirlenmesidir. Gereç ve Yöntem: Bu çalışmada meme kanseri alt tiplerine mensup olan dört, sağlıklı meme tipine sahip bir hücre hattı; T-47D, MDA-MB-231, AU-565, BT-474 ve MCF-10A ilk olarak akış sitometrisi yardımı ile peptit doygunluğunun belirlenmesinde kullanılmış ardından belirlenen Kd değeri ile rekabet bağlanma deneyi yapılmıştır. Bunun için projenin önceki iş paketlerinde ortak olarak bulunan reseptör hedeflenmiştir. Bu reseptörün doğruluğu qPZR ve western emdiriminin yanı sıra, in silico analizlerle doğrulanmıştır. Belirlenen bu reseptör daha sonra CRISPR/Cas9 sistemi ile susturulmuş ve hücrelerin susturma kalitesi western emdirimi ve ICC ile kontrol edilmiş, devamında P18-4'ün susturulmuş hücrelerde bağlanma aktivitesi gözlemlenmiştir. Bulgular: In silico analizler ve tüm hücre hatlarıyla yapılan qPZR ve western emdirimi deneyleri sonucunda PEBP1 reseptörünün hedef olarak değerlendirilebileceği gösterilmiştir. Ardından T-47D, MDA-MB-231, AU-565, BT-474 ve MCF-10A hücre hatları için P18-4'ün Kd değeri sırasıyla 2,5, 20, 3,8, 17 ve 74,22M olarak bulunmuştur. PEBP1 ile yapılan rekabet deneyinde AU-565 ve T-47D hücrelerinde anlamlı sonuçlar alınmıştır. CRISPR/Cas9 susturmasının ardından floresans görüntülerinde GFP ışımaları gözlemlenmiş olup, özellike AU-565 ve T-47D hücrelerinde western emdirimi ve ICC yardımı ile susturma kalitesi kanıtlanmıştır. Son olarak susturulan hücrelerde P18-4'ün bağlanma etkinliğinde T-47D ve AU-565 hücre hatlarında anlamlı bir azalma gözlemlenmiştir. Sonuç: Sonuç olarak PEBP1'ın özellikle AU-565 ve T-47D hücre hatlarında P18-4 ile bağlanan ortak bir reseptör olduğu belirlenmiştir.

Aim: The aim of this study is to find a common receptor that binds P18-4 in breast cancer subtypes by the use of CRISPR/Cas9 system. Materials and Methods: In this study 4 breast cancer and 1 healthy breast cell types; T-47D, MDA-MB-231, AU-565, BT-474 and MCF-10A were used. First flow cytometry was used to determine peptide saturation and calculated Kd concentration was used for competitive binding assays. For this, the common receptor that was found in the previous studies was targeted. Acceptibility of this receptor was confirmed by both qPCR and western blot as well as in silico analysis. Determined receptor then knocked out with CRISPR/Cas9 system and the effect of the knock-out were checked by western blot and ICC. After that, the binding activity of the P18-4 on knocked-out cells were observed. Results: In silico analysis and the qPCR and western blot experiments for all cell lines showed PEBP1's possibility as a common receptor. Later on, Kd values for T-47D, MDA-MB-231, AU-565, BT-474 and MCF-10A were calculated as 2,5, 20, 3,8, 17 ve 74,22M respectively. Competitive binding assay with PEBP1 results showed significance on AU-565 and T-47D cell lines. After CRISPR/Cas9 knock-out, GFP florescences were observed and western blot and ICC showed effective knock-out especially on AU-565 and T-47D cell lines. Lastly, from the knock-out cells T-47D and AU-565 showed significant decrease in P18-4 binding ability. Conclusion: It has been concluded that PEBP1 can be considered as a common receptor that binds P18-4, especially on T-47D and AU-565 cell lines.
