Özet
Nipah virüsünün, insanlarda ve hayvanlarda yüksek mortalite oranlarına sahip, ciddi klinik tablolara neden olabilen zoonotik bir etken olarak dikkat çekmektedir. Nipah virüsü (NiV)'nün Dünya Sağlık Örgütü (DSÖ)'nün 'Gelecekte pandemilere neden olma potansiyeline sahip yüksek öncelikli patojenler' listesinde bulunması halk arasaında ciddi bir tehdit oluşturabileceğinin sinyallerini vermiştir. Günümüzde bu virüse karşı onaylanmış, spesifik bir aşının bulunmaması, yeni nesil aşı stratejilerine olan ihtiyacı artırmaktadır. Bu doğrultuda, Nipah virüsüne karşı peptit temelli aşı adaylarının belirlenmesine yönelik in silico tabanlı biyoinformatik yöntemlerden yararlanılarak viral hücreye giriş, replikasyon ve yapısal organizasyon gibi temel süreçlerde görev alan füzyon (F), tutunma (G) ve matriks (M) proteinleri, çalışmanın hedef proteinleri olarak seçilmiştir. Çalışma iki temel fazdan oluşmaktadır. İlk aşamada, seçilen yapı proteinlerinin biyofiziksel ve biyokimyasal özellikleri detaylı bir şekilde analiz edilerek doğrudan aşı adayı olup olamayacakları değerlendirilmiştir. Bu analizler sonucunda, F, G ve M proteinlerinin fiziko-kimyasal nitelikleri açısından doğrudan aşı hedefi olarak uygun olmadıkları saptanmıştır. Takip eden ikinci aşamada ise, bu proteinlerden türetilen olası epitop dizileri, çeşitli hesaplamalı araçlar yardımıyla immunolojik uygunlukları açısından değerlendirilmiştir. Yapılan analizler sonucunda; Glikoproteinden türetilen MMTRLAVKPK epitopu, öne çıkan potansiyel aşı adayı olarak belirlenmiştir. Bu epitopun yüksek antijenite skorlaruna (1.4367) sahip olması, suda çözünürlüğünün yüksek olması, toksik ve alerjenik özellik taşımaması, uygun IC50 değerine ve immünolojik olarak anlamlı proteaz kesim bölgeleri içeriyor olması aşı adayı olarak belirlenmesinde etkili olmuştur. Öte yandan, antijenite skoru en yüksek olan (2.0045) FLLKNKIWCI peptidi, pozitif hidrofobiklik değeri (0.07) nedeniyle yapısal analizlere dahil edilse de değerlendirilmeye alınmamıştır. Bu çalışma, Nipah virüsüne karşı peptit bazlı aşı tasarımında kullanılabilecek bağışıklık yanıtı oluşturma potansiyeline sahip hedeflerin in silico ortamda başarılı bir şekilde tespit edilebileceğini göstermektedir. Ancak elde edilen bulgular deneysel doğrulamalardan bağımsız olarak hesaplamalı tahminlere dayandığı için, bu epitopların gerçek bağışıklık yanıtı oluşturma kapasitelerinin ortaya konabilmesi adına laboratuvar düzeyinde deneysel çalışmalara ihtiyaç duyulmaktadır.

Nipah virus is a zoonotic agent known for its high mortality rates and the ability to cause severe infections in both humans and animals. Its inclusion on the World Health Organization's (WHO) list of priority pathogens with pandemic potential underscores the urgent need for effective preventive strategies. Currently, no licensed or specific vaccine is available against Nipah virus (NiV), which reinforces the importance of developing novel vaccine approaches. This study focused on identifying potential peptide-based vaccine candidates using in silico bioinformatics tools. The fusion (F), attachment (G), and matrix (M) proteins of the virus, essential for viral entry, replication, and assembly, were selected as key targets. The research was carried out in two stages. The first stage involved a comprehensive analysis of the selected proteins to assess their biochemical and biophysical characteristics and their suitability as vaccine targets. The findings from this phase suggested that the F, G, and M proteins, due to certain unfavorable physicochemical features, are not ideal as direct vaccine candidates. In the subsequent second phase, the potential epitope sequences derived from these proteins were evaluated for their immunological suitability using various computational tools. As a result of the analyses conducted, the epitope MMTRLAVKPK, derived from the glycoprotein, was identified as a prominent potential vaccine candidate. This determination was supported by several factors, including its high antigenicity score (1.4367), high water solubility, non-toxic and non-allergenic nature, an appropriate IC50 value, and the presence of immunologically relevant protease cleavage sites. On the other hand, although the peptide FLLKNKIWCI, which exhibited the highest antigenicity score (2.0045), was included in the structural analyses despite its positive hydrophobicity value (0.07), it was not taken into consideration for further evaluation. Overall, the study highlights the potential of computational approaches to identify novel immunogenic targets for vaccine development against NiV. However, as the findings are based solely on in silico predictions, experimental validation through laboratory-based research is necessary to confirm the true immunogenic potential of the proposed epitopes.
