Özet
Kanserin dünya genelinde giderek artan yaygınlığı ve mevcut kemoterapötik ajanların agresif, çoğu zaman kontrolsüz hücresel yanıtlar oluşturması, daha güvenilir ve sürdürülebilir ilaç taşıma stratejilerinin geliştirilmesini zorunlu kılmaktadır. Bu bağlamda nano taşıyıcı sistemlerde çok katmanlı mimarilerin tasarlanması hem ilaç stabilitesini iyileştirmek hem de hedef hücrelerle kurulan etkileşimleri daha kontrollü hâle getirmek açısından önemli bir araştırma alanı olarak öne çıkmaktadır. Bu çalışma, halloysit nanotüp (HNT) temelli yapılara polidopamin (PDA) modifikasyonu ve 5-fluorourasil (5-FU) konjugasyonun entegre edildiği; ardından PEG ve Dekstran gibi biyouyumlu polimerlerle enkapsülasyonları gerçekleştirilerek çok bileşenli bir platformun tasarlanıp kapsamlı bir şekilde değerlendirilmesini temel almıştır. Doğal nanotüp mimarisi, biyomimetik PDA yüzey kimyası ve polimerik üst fazların bir arada kullanılması, serbest 5-FU'nun agresif sitotoksisitesini azaltan ve biyolojik ortamda daha öngörülebilir bir davranış sunan bir nano taşıyıcı yaklaşımının geliştirilmesini mümkün kılmıştır. Karakterizasyon analizleri, geliştirilen yapıların çok katmanlı ve stabil bir mimariye sahip olduğunu göstermiştir. FTIR, XPS TEM ve DSC verileri, PDA'nın HNT yüzeyine başarıyla modifiye olduğunu, 5-FU'nun ise PDA matrisi içinde kovalent bağ ve hidrojen bağı gibi sekonder etkileşimler aracılığıyla amorf bir fazda tutulduğunu doğrulamıştır. ¹H-NMR ve ¹⁹F-NMR analizleri, HNT-PDA@5-FU ve PDA@5-FU konjuge yapılarında ilacın kimyasal ortamının belirgin biçimde değiştiğini ve PDA yapısı içerisine gömülü, serbest formundan farklı bir konumda yer aldığını göstermiştir. PEG ve Dekstran enkapsülasyonları, konjuge yapının dış yüzeyine farklı özellikler kazandırmış; PEG düzensiz temasları sınırlayan kompakt bir koruyucu tabaka, Dekstran ise hidrofilik ve elastik bir faz oluşturarak daha biyouyumlu bir ortam sunmuştur. Hücre sitotoksisite değerlendirmeleri, serbest 5-FU'nun keskin sitotoksik etkisine karşın PDA ve HNT varlığının ilacın hücreyle temasını düzenleyerek daha kontrollü bir yanıt sağladığını ortaya koymuştur. MDA-MB-231 hücrelerinin dopamin reseptörlerin varlığı nedeniyle sadece PDA varlığı ile konjuge ve enkapsüle edilmiş sistemlerde PDA'nın varlığının hücre membranı çevresinde kurduğu temas belirli düzeyde hücreler üzerinde stres oluşturabilse de HNT'li sistemlerde HNT'nin varlığının hidrojen bağı etkileri ve özellikle PEG ve Dekstran enkapsüle sistemde kendisini göstermiştir. Elde edilen bulgular, HNT-PDA@5-FU ve PDA@5-FU merkezli çok katmanlı nano taşıyıcıların serbest 5-FU'ya kıyasla daha stabil, daha kontrollü ve daha biyouyumlu bir profil sunduğunu göstermiştir. Polimer enkapsülasyon stratejileri, hem ilacın PDA matrisi içindeki stabilitesini hem de yüzey etkileşimlerinin hücre üzerindeki etkisini optimize ederek sistemin genel biyolojik performansını güçlendirmiştir. Bu çalışma, doğal, biyomimetik ve polimerik fazların bütünleşik kullanımıyla gelecekte hedeflenebilir ve çok bileşenli ilaç taşıyıcı tasarımlarına güçlü bir temel sunmaktadır.

The global rise in cancer incidence and the aggressive, often uncontrolled cellular responses elicited by conventional chemotherapeutic agents underscore the need for safer and more sustainable drug-delivery strategies. In this context, the design of multilayered architectures within nanocarrier systems has emerged as a critical research direction, offering opportunities to enhance drug stability and to regulate interactions with target cells in a more predictable manner. This study focuses on the development and comprehensive evaluation of a multicomponent platform built upon halloysite nanotubes (HNTs), integrating polydopamine (PDA) modification and 5-fluorouracil (5-FU) conjugation, followed by encapsulation within biocompatible polymers such as PEG and Dextran. The combination of the natural nanotubular structure, biomimetic PDA surface chemistry, and polymeric outer phases enables a nanocarrier approach capable of attenuating the aggressive cytotoxicity of free 5-FU while promoting a more controlled behavior in biological environments. Characterization analyses confirmed that the engineered structures exhibit stable and well-defined multilayered architecture. FTIR, XPS, TEM, and DSC results verified successful PDA modification on the HNT surface and demonstrated that 5-FU is retained within the PDA matrix in an amorphous state through covalent interactions and secondary forces such as hydrogen bonding. Both ¹H-NMR and ¹⁹F-NMR further revealed distinct changes in the chemical environment of the drug in HNT-PDA@5-FU and PDA@5-FU conjugates, indicating its embedded and chemically altered state compared with the free form. PEG and Dextran encapsulation imparted additional surface properties; PEG provided a compact protective layer restricting nonspecific interactions, while Dextran offered a hydrophilic and elastic outer phase that enhanced the biocompatibility of the system. Cell cytotoxicity assessments demonstrated that, in contrast to the sharp cytotoxic effect of free 5-FU, the presence of PDA and HNT modulated the drug–cell interaction, resulting in a more controlled cellular response. Owing to the expression of dopamine receptors in MDA-MB-231 cells, PDA-conjugated and PDA-encapsulated systems induced a certain level of cellular stress through PDA-mediated interactions at the cell membrane. However, in HNT-containing systems, the incorporation of HNT contributed to the regulation of these interactions via hydrogen-bonding effects, a phenomenon that became particularly pronounced in PEG- and dextran-encapsulated formulations. Overall, the findings indicate that multilayered nanocarriers centered on HNT-PDA@5-FU and PDA@5-FU exhibit greater stability, enhanced control over drug release and cell interactions, and improved biocompatibility compared with free 5-FU. Polymer encapsulation strategies strengthened both the stability of the drug within the PDA matrix and the modulation of surface interactions, collectively enhancing the overall biological performance of the system. This work provides a solid foundation for future targeted and multicomponent drug-delivery designs through the integrated use of natural, biomimetic, and polymeric phases.
