Temozolomide bağlı gelişen nöronal toksisite modelinde oksitosinin etkisinin deneysel hayvan modelinde gösterilmesi

Özet
Santral sinir sistemi, yüksek metabolik gereksinimleri ve sınırlı yenilenme kapasitesi nedeniyle toksik ajanlara karşı oldukça duyarlıdır. Temozolomid (TMZ), glial tümörlerin tedavisinde etkin bir kemoterapötik ajan olmasına karşın, yan etkiler nedeniyle nörotoksisiteye neden olabilmektedir. Bu çalışmada, deneysel sıçan modelinde temozolomid ile indüklenen nörotoksisiteye karşı oksitosinin potansiyel koruyucu etkileri araştırıldı. Çalışmada, 48 adet dişi Wistar Albino sıçan rastgele dört gruba ayrıldı: Kontrol grubu, Oksitosin (OT) grubu, Temozolomid (TMZ) grubu ve Temozolomid + Oksitosin (TMZ+OT) grubu. Temozolomid, 4 hafta boyunca haftada iki kez 10 mg/kg dozunda oral gavaj yoluyla, toplam sekiz uygulama şeklinde verildi. Oksitosin ise 4 hafta süresince her gün 100 IU/kg dozunda intraperitoneal yolla uygulandı. Deney sonunda beyin dokularından Malondialdehit, Tümör Nekroz Faktörü-α, Beyin Kaynaklı Nörotrofik Faktör, Glial Hücre Kaynaklı Nörotrofik Faktör ve Glutamik Asit Dekarboksilaz-67 düzeyleri ölçüldü. Ayrıca, bilişsel ve davranışsal etkileri değerlendirmek amacıyla üç odalı sosyal tanıma testi, açık alan testi ve pasif kaçınma testi uygulandı. Histopatolojik incelemeler ise hipokampüs ve serebellum bölgelerinde gerçekleştirildi. Bulgular, temozolomid uygulamasının beyin dokusunda oksidatif stres ve inflamasyonu artırdığını, nörotrofik faktör düzeylerini azalttığını ve davranışsal testlerde bilişsel işlev bozukluklarına yol açtığını ortaya koydu. Histopatolojik analizlerde hipokampüs ve serebellumda belirgin nöronal kayıp, yapısal bütünlükte bozulmalar ve gliotik değişiklikler gözlendi. Oksitosin tedavisinin ise temozolomid kaynaklı biyokimyasal, davranışsal ve histopatolojik hasarları anlamlı düzeyde hafiflettiği; oksidatif stres belirteçlerini azalttığı, nörotrofik faktör düzeylerini koruduğu ve sinir dokusu bütünlüğünü desteklediği belirlendi. Sonuç olarak, bu çalışma oksitosinin temozolomid kaynaklı nörotoksisiteyi azaltmada potansiyel bir nöroprotektif ajan olabileceğini göstermekte ve kemoterapi sonrası gelişebilecek nörolojik yan etkilerin önlenmesi açısından umut verici bir molekül olduğunu düşündürmektedir.

The central nervous system is highly susceptible to toxic agents due to its high metabolic demands and limited regenerative capacity. Although temozolomide (TMZ) is an effective chemotherapeutic agent in the treatment of glial tumors, it can cause neurotoxicity as a side effect. In this study, the potential protective effects of oxytocin against temozolomide-induced neurotoxicity were investigated using an experimental rat model. A total of 48 female Wistar Albino rats were randomly divided into four groups: Control group, Oxytocin (OT) group, Temozolomide (TMZ) group, and Temozolomide + Oxytocin (TMZ+OT) group. Temozolomide was administered orally via gavage at a dose of 10 mg/kg twice weekly for 4 weeks (a total of eight administrations). Oxytocin was administered intraperitoneally at a dose of 100 IU/kg daily for 4 weeks. At the end of the experiment, brain tissues were collected for biochemical analyses, including measurements of malondialdehyde, tumor necrosis factor-alpha, brain-derived neurotrophic factor, glial cell line-derived neurotrophic factor, and glutamic decarboxylase-67 levels. In addition, behavioral and cognitive functions were evaluated using the three-chamber social recognition test, open field test, and passive avoidance test. Histopathological examinations were performed on the hippocampus and cerebellum regions. The findings revealed that temozolomide administration significantly increased oxidative stress and inflammation, reduced neurotrophic factor levels, and caused cognitive impairment in behavioral tests. Histopathological analyses demonstrated prominent neuronal loss, structural disruption, and gliotic changes in the hippocampus and cerebellum. Oxytocin treatment significantly alleviated the biochemical, behavioral, and histopathological alterations induced by temozolomide, reduced oxidative stress markers, preserved neurotrophic factor levels, and supported the integrity of neural tissue. In conclusion, this study demonstrates that oxytocin may serve as a potential neuroprotective agent in mitigating temozolomide-induced neurotoxicity, suggesting its promise in preventing neurological side effects associated with chemotherapy.
