Özet
Bu tez çalışmasında, Türkiye florasının önemli tıbbi ve aromatik bitkileri arasında yer alan Salvia multicaulis, Salvia palaestina ve Salvia tomentosa türlerinin genotoksik ve antigenotoksik etkileri, model organizma olarak kullanılan Drosophila melanogaster üzerinde karşılaştırmalı olarak araştırılmıştır. Günümüzde artan çevresel kirlilik, kimyasal ajanlara maruziyet ve oksidatif stres kaynaklı DNA hasarları, mutasyon, kanser ve çeşitli dejeneratif hastalıkların oluşumunda önemli rol oynamaktadır. Bu nedenle doğal kaynaklı biyolojik koruyucu bileşiklerin belirlenmesi, modern toksikoloji ve farmakoloji araştırmalarında öncelikli çalışma alanlarından biri hâline gelmiştir. Özellikle Lamiaceae familyasına ait Salvia türleri, fenolik bileşikler, flavonoidler, diterpenler ve uçucu yağlar bakımından zengin içerikleri sayesinde antioksidan, antimikrobiyal, antiinflamatuvar ve antigenotoksik özellikleri ile dikkat çekmektedir. Bu kapsamda çalışmada, Anadolu florasında doğal yayılış gösteren üç farklı Salvia türünün DNA hasarına karşı koruyucu potansiyellerinin ortaya konulması amaçlanmıştır. Araştırmada deney materyali olarak S. multicaulis, S. palaestina ve S. tomentosa türlerinden elde edilen bitki ekstraktları kullanılmıştır. Genotoksisite ve antigenotoksisite analizleri, somatik mutasyon ve rekombinasyon testi (SMART) kullanılarak gerçekleştirilmiştir. Deney organizması olarak, kısa yaşam döngüsü, yüksek üreme kapasitesi, düşük maliyetle üretilebilmesi ve insan genomu ile önemli düzeyde genetik benzerlik göstermesi nedeniyle D. melanogaster tercih edilmiştir. Çalışmada standart çapraz yöntemi uygulanmış; transheterozigot larvalar farklı dozlarda hazırlanan Salvia özütlerine ve pozitif kontrol ajanı olarak kullanılan Mitomisin C (MMC)'ye maruz bırakılmıştır. Uygulama sonrasında ergin bireylerden elde edilen kanatlar stereomikroskop altında incelenmiş; küçük tekli benek, büyük tekli benek ve ikili benek frekansları değerlendirilerek genotoksik ve antigenotoksik etkiler istatistiksel olarak analiz edilmiştir. Elde edilen bulgular, MMC uygulanan pozitif kontrol grubunda belirgin düzeyde genetik hasar ve benek oluşumu meydana geldiğini göstermiştir. Buna karşın, tek başına uygulanan Salvia özütlerinin deney gruplarında anlamlı düzeyde genotoksik etki oluşturmadığı belirlenmiştir. Bu durum, incelenen bitki türlerinin belirlenen doz aralıklarında biyolojik açıdan güvenilir bir profil sergilediğini ortaya koymuştur. MMC ile birlikte uygulanan Salvia özütlerinin ise kanat benek frekanslarında azalmaya neden olduğu ve DNA hasarını baskılayıcı etki gösterdiği saptanmıştır. Özellikle S. multicaulis ve S. tomentosa uygulamalarında antigenotoksik etkinin daha belirgin olduğu gözlenmiş; bu etkinin bitkilerin yüksek fenolik madde ve flavonoid içerikleri ile ilişkili olabileceği değerlendirilmiştir. Elde edilen sonuçlar, Salvia türlerinin serbest radikal oluşumunu baskılayarak oksidatif DNA hasarını azaltabileceğini ve hücresel düzeyde koruyucu mekanizmaları destekleyebileceğini göstermektedir. Sonuç olarak, bu çalışma Türkiye florasında doğal yayılış gösteren bazı Salvia türlerinin genetik toksisiteye karşı koruyucu potansiyele sahip olduğunu ortaya koymuştur. Bulgular, doğal kaynaklı antigenotoksik ajanların geliştirilmesine katkı sağlaması açısından önem taşımaktadır. Ayrıca çalışma, Anadolu'nun zengin bitkisel biyoçeşitliliğinin yalnızca ekolojik değil, aynı zamanda farmakolojik ve biyoteknolojik açıdan da stratejik değere sahip olduğunu desteklemektedir. İncelenen türlerin ileri düzey moleküler analizler, fitokimyasal karakterizasyon çalışmaları ve farklı deney sistemleri ile desteklenmesi, gelecekte doğal ürün temelli koruyucu ve terapötik ajanların geliştirilmesine önemli katkılar sağlayacaktır.

In this thesis study, the genotoxic and antigenotoxic effects of Salvia multicaulis, Salvia palaestina, and Salvia tomentosa, which are among the important medicinal and aromatic plants of the Turkish flora, were comparatively investigated using Drosophila melanogaster as a model organism. Today, increasing environmental pollution, exposure to chemical agents, and oxidative stress-induced DNA damage play significant roles in the development of mutations, cancer, and various degenerative diseases. Therefore, the identification of natural bioactive protective compounds has become one of the major research areas in modern toxicology and pharmacology. Species belonging to the Lamiaceae family, particularly the genus Salvia, attract considerable attention due to their rich content of phenolic compounds, flavonoids, diterpenes, and essential oils, as well as their antioxidant, antimicrobial, anti-inflammatory, and antigenotoxic properties. In this context, the present study aimed to reveal the protective potential of three different Salvia species naturally distributed in the Anatolian flora against DNA damage. Plant extracts obtained from S. multicaulis, S. palaestina, and S. tomentosa were used as experimental materials. Genotoxicity and antigenotoxicity analyses were performed using the Somatic Mutation and Recombination Test (SMART). D. melanogaster was preferred as the experimental organism due to its short life cycle, high reproductive capacity, low maintenance cost, and considerable genetic similarity to the human genome. In the study, a standard cross system was applied, and trans-heterozygous larvae were exposed to different concentrations of Salvia extracts and Mitomycin C (MMC), which was used as the positive control agent. Following the treatments, wings obtained from adult individuals were examined under a stereomicroscope, and the frequencies of small single spots, large single spots, and twin spots were evaluated statistically to determine genotoxic and antigenotoxic effects. The findings demonstrated that significant genetic damage and spot formation occurred in the MMC-treated positive control group. In contrast, Salvia extracts administered alone did not produce a statistically significant genotoxic effect in the experimental groups. This result indicated that the investigated plant species exhibited a biologically safe profile within the tested dose ranges. Moreover, co-treatment with Salvia extracts reduced wing spot frequencies and showed suppressive effects against DNA damage. Antigenotoxic activity was found to be more pronounced particularly in S. multicaulis and S. tomentosa applications, and this effect may be associated with the high phenolic and flavonoid contents of these species. The obtained results suggest that Salvia species may reduce oxidative DNA damage by suppressing free radical formation and may support cellular protective mechanisms. In conclusion, this study revealed that some Salvia species naturally distributed in the Turkish flora possess protective potential against genetic toxicity. The findings are important in terms of contributing to the development of natural antigenotoxic agents. Furthermore, the study supports that the rich plant biodiversity of Anatolia has not only ecological but also pharmacological and biotechnological significance. Supporting the investigated species with advanced molecular analyses, phytochemical characterization studies, and different experimental systems may provide important contributions to the development of natural product-based protective and therapeutic agents in the future.
