Özet

Yara iyileşmesi sürecinde enfeksiyon riski ve doku yenilenmesinin yavaş seyri, mevcut tedavi yöntemlerinin etkinliğini sınırlamaktadır. Bu soruna çözüm getirmek için hem iyileşmeyi hızlandıran hem de enfeksiyonu önleyen biyofonksiyonel yara örtülerine ihtiyaç duyulmaktadır. Bu çalışmada PVA (polivinil alkol) ve kitosan gibi biyolojik olarak parçalanabilen polimerler ile yara örtüsü oluşturma ve biyolojik uyumluluğu arttırma yolları izlenmiştir. Yara örtüsü çalışmaları için seçilen bitkiler doğal; antimikrobiyal ve iyileştirici özelliklere sahip bileşikler içermektedir. Bu bileşiklerin yara örtülerine entegrasyonu, iyileşme sürecine olumlu katkı sunmaktadır. Bu çalışmanın amacı, bitkisel kaynaklı antimikrobiyal bileşenler ve biyouyumlu polimerler kullanılarak, 3B baskı ve elektrosprey yöntemiyle çift katmanlı bir yara örtüsü geliştirmektir. Bu amaçla, PVA/Kitosan/Arap zamkı içeren solüsyonlar hazırlanmış ve 3B baskı gerçekleştirilmiş; ardından PCL (poli-ε-kaprolakton)/Kınakına (KK) ekstraktı içeren solüsyon elektrosprey yöntemiyle ilk katmanın üzerine uygulanarak çift katmanlı yapı oluşturulmuştur. Üretilen yara örtüleri SEM, FTIR, çekme, antioksidan, antibakteriyel, hücre canlılığı ve salım testleriyle karakterize edilmiştir. SEM analizi, yara örtülerinin homojen bir film yapısı oluşturduğunu göstermiştir. FTIR testi, bu doğal bileşenlerin biyopolimer matris içerisine başarılı şekilde entegre edildiğini doğrulamakta ve bileşenler arasında kimyasal etkileşimlerin oluşabileceğine işaret etmektedir. PVA/Kitosan/AZ/PCL/KK (çift katman) kompozit örnekleri E. coli'ye karşı 8.53 ± 0.38 mm ve S. aureus'a karşı 9.53 ± 0.65 mm çapında zon oluşturarak antimikrobiyal etki göstermiştir. Çift katman yapısı sayesinde toplam salımda kontrollü bir denge elde edilmiştir. Ayrıca KK'nın güçlü antioksidan aktivite gösterdiği belirlenmiştir. PCL/KK (%104,8 ± 6,1), PVA/Kitosan (%82,8 ± 5) ve PVA/Kitosan/AZ (%89,3 ± 6,5) formülasyonları, hücre canlılığını %70'in üzerinde tutarak ISO 10993-5 standardına göre sitotoksik olmayan davranışlar sergilemiştir. Buna karşılık, PVA/Kitosan/AZ/PCL/KK içeren çok bileşenli sistemde hücre canlılığının %56,6 ± 3,8'e düşmesi, ISO 10993-5 standardına göre potansiyel sitotoksik etkiye işaret etmektedir. Literatürdeki çalışmalar incelendiğinde 3B yazıcı teknolojisi kullanılarak kınakına ekstraktı içeren ve kitosan, Arap zamkı, PVA ve PCL polimerlerini bir arada bulunduran bir yara örtüsü sisteminin üretimine yönelik bir çalışmaya rastlanmamıştır. Genel olarak bu çalışma, doğadan elde edilen ve biyouyumlu malzemeler kullanılarak, yara iyileşmesini destekleyen ve enfeksiyon riskini azaltan yenilikçi bir yara örtüsü üretilebileceğini göstermektedir. Bu yaklaşım, biyoteknoloji ve biyomühendislik birleşimiyle daha etkili ve güvenli yara bakım ürünlerinin geliştirilmesine katkı sunmaktadır.

The risk of infection and the slow progression of tissue regeneration during the wound healing process limit the effectiveness of current treatment methods. To address this problem, biofunctional wound dressings that both accelerate healing and prevent infection are needed. This study investigated ways to create wound dressings using biodegradable polymers such as PVA and chitosan and to improve their biocompatibility. The plants selected for wound dressing studies contain natural compounds with antimicrobial and healing properties. The integration of these compounds into wound dressings contributes positively to the healing process. The aim of this study is to develop a double-layered wound dressing using plant-derived antimicrobial components and biocompatible polymers via 3D printing and electrospray. For this purpose, solutions containing PVA/chitosan/Gum arabic were prepared and 3D printed; then, a solution containing PCL/Cinchona extract was applied to the first layer using an electrospray method to create a double-layered structure. The produced wound dressings were characterized using SEM, FTIR, tensile strength, antioxidant, antibacterial, cell viability, and release tests. SEM analysis showed that the wound dressings formed a homogeneous film structure. The FTIR test confirmed that these natural components were successfully integrated into the biopolymer matrix and indicated that chemical interactions could occur between the components. PVA/Chitosan/AZ/PCL/KK (bilayer) composite samples exhibited antimicrobial activity by forming inhibition zones of 8.53 ± 0.38 mm against E. coli and 9.53 ± 0.65 mm against S. aureus. A controlled balance in total release was achieved thanks to the double-layered structure. Furthermore, KK was determined to exhibit strong antioxidant activity. PCL/KK (104,8 ± 6,1%), PVA/Chitosan (82,8 ± 5%) and PVA/Chitosan/AZ (89,3 ± 6,5%) formulations maintained cell viability above 70%, demonstrating non-cytotoxic behavior according to the ISO 10993-5 standard. In contrast, the multicomponent system containing PVA/Chitosan/AZ/PCL/KK exhibited a reduced cell viability of 56,6 ± 3,8%, indicating a potential cytotoxic effect according to ISO 10993-5 standard. A review of the literature revealed no previous study reporting the fabrication of a wound dressing system containing quinine extract combined with chitosan, Gum arabic, PVA, and PCL polymers using 3D printing technology. Overall, this study demonstrates that an innovative wound dressing that promotes wound healing and reduces the risk of infection can be produced using biocompatible materials derived from nature. This approach contributes to the development of more effective and safer wound care products through the combination of biotechnology and bioengineering.
