Özet

Astrogliosis after spinal cord injury is proportional to lesion severity, regulates serotonergic axonal sprouting and correlates with motor recovery. Elucidating the reasons for motor impairments (weakness and abnormal involuntary reflex activity, called spasticity or spasms) after spinal cord injury (SCI) is crucial for understanding the mechanisms of functional recovery. In the past few decades, accumulating data has suggested that specific neuromodulatory agents, which mimic or facilitate the actions of the monoamines, including serotonin (5HT), can initiate or augment walking behaviors in animal models of SCI. In the present study we looked for correlations between objective functional (single-frame video motion analysis) and morphological (lesion scar volume intensity of the GFAP and 5HT fibers sprouting) parameters after SC compression injury in rats. Adult female Wistar rats were subjected to graded compression of the spinal cord at midthoracic level (Th8). Recovery of locomotion was analyzed using video recordings of beam walking and inclined ladder climbing. Four functional parameters were used: the foot-stepping angle (FSA), the rump-height index (RHl) estimating paw placement and body weight support, respectively, the number of correct Iadder steps (CLS) assessing skilled hind limb movements and the locomotor rating score of Basso, Beattie and Bresnahan (BBB). These parameters correlated with the scar volume (measured in Cresyl Violet-stained longitudinal sections through the lesion site) and with the intensity of CY3-fluorescence after immunostaining (rabbit anti-5HT, 1:2000, ImmunoStar, Cat. Nr. 20080 or polyclonal anti-GFAP from guinea pig, Progen, GP52, 1:4,000) of serotonergic fibers and astrocytes in 25 ?m thick transverse cryostat sections through the lumbar intumescence showing significant differences between moderately and severely injured rats at 1-9 weeks after SCI. The use of these objective functional and morphological measures provides a time- and cost-efficient opportunity for versatile and reliable functional evaluations in both severely and moderately impaired rats combining clinical assessment with precise numerical measures. Our morphological results indicate that the proliferation of astrocytes below the lesion site corresponded to the severity of lesion: there was significantly less astrogliosis following moderate compression than after severe compression of the spinal cord. This, in turn, allowed an increased amount of serotonergic axonal sprouts to regrow across the lesion site.
