Özet

ÖZET Bu araştırma, broilerlerde M. Pectoralis Superficialis ve M. Biceps Femoris'den alınan örneklerin yeme ve gelişime bağlı olarak yapısal özelliklerinin histomorfolojik yöntemlerle incelenmesi amacıyla yapıldı. Araştırma'da, protein oranı %24,10 (I. grup), ve %16,54 (E. grup) olan iki grup rasyonîa beslenen broilerlerde 14., 28., 42., ve 56. günlerde alınan kas örnekleri incelendi. Histolojik incelemeler için, örneklerin bir kısmında kryostat'da kesitler alınarak ATPase ve Sudan HI boyaması uygulandı. Tamponlanmış formol ve formol- salin'de tesbit edilen diğer örneklerden rutin histolojik yöntemlerle parafin bloklar hazırlandı. 7uTuk kesitlere Triple, PAS ve Gümüşleme-Orcein-Anilin boyamaları uygulandı. Histometrik değerlendirmelerde M. Pectoralis Superficialis ve M. Biceps Femoris örneklerindeki primer kas demetindeki kırmızı, intermedier ve beyaz kas tellerinin çaplan ile sayılan ölçüldü ve demet içindeki % oranlan tesbit edildi. Elde edilen bulgulara göre; I. grup için M. Biceps Femoris'de beyaz, intermedier ve kırmızı kas ipliklerinin çap ortalamaları 14., 28., 42. ve 56. günlerde sırasıyla 41.23u-75.47u, 32.19u-58.96u ve 22.42u-47.05u arasında; II. grup için ise 35.46u-70.23u, 29.26u-54.89u ve 19.86u-41.39u arasmda değişen değerlerde tesbit edildi. M. Pectoralis Superficialis'de I. ve n. grup için beyaz, intermedier ve kırmızı kas iplMerinin çap ortalamalan 14., 28., 42. ve 56. günlerde sırasıyla 45.12u-75.72u ve 38.11u-67.53u, 34.15u-61.87u ve 27.64u-55.82u, 23.31u-55.96u ve 19.72u- 40.15u arasında olduğu saptandı. M. Biceps Femoris'de I. grup için beyaz, intermedier ve kırmızı kas ipliklerinin oranlan 14., 28., 42. ve 56. günlerde sırasıyla %15.09-%20.30, %13.41- %16.30 ve %63.58-%70.44; H grup için ise %15.18-%19.68, %17.88-%20.84 ve %60.24-%65.46 arasmda olduğu tesbit edildi. M. Pectoralis Superficialis'de I. grup için değişen büyüme dönemlerinde (14., 28., 42. ve 56. günler) beyaz, intermedier ve kırmızı kas iplMerinin oranlan sırasıyla %59.99-%69.09, %17.11-%22.93 ve %12.48-%Î7.08; II. grup için ise %60.75-%65.73, %19.23-%26.69 ve %14.56- %16.58 arasında değişen değerlerde olduğu saptandı. SUMMARY This study was performed for examination of the structural changes of M.Pectoralis Superficialis and M. Biceps Femoris due to diet and growth. The samples were taken from broilers and histological methods were used. The muscle samples were taken from two groups at 14th. 28th, 42th and 56th days. The first group and the second group were fed with diets which contain 24,10% and 16,54% proteins respectively. Some samples were sectioned with cryostat and treated ATPase and Sudan III stain. The other samples were fixed in buffered formaline and formaline-salin. Then they were embedded in paraffin with routine histological methods. The sections which are 7u thick, were stained with Triple PAS and Silver-Orcein- Anilin. The diameter, percentage and number of the red, intermediate and white muscle fibres were established in primary muscle bundle of M. Pectoralis Superficialis and M. Biceps Femoris. In the first group, average diameters of white, intermediate and red muscle fibres were 41.23u-75.47u, 32.19u-58.96u, 22.42u-47.05u in M. Biceps Femoris at 14th, 28th, 42th, 56th days respectively. In the second group, these diameters were 35.46u- 70.23 u, 29.26u-54.89u, 19.86u~41.39u. Diameter of white, intermediate and red muscle fibres in M. Pectoralis Superficialis for the I and II group were measured between following values: 45.12u-75.72u, 38.11u-67.53u, 34.15u-61.87u, 27.64u-55.82u, 23.31u-55.96u, 19.72u- 40. 15u at 14th, 28th, 42th, 56th days respectively. In the first group, percentage of white, intermediate and red muscle fibres in M. Biceps Femoris were 15.09%-20.30%, 13.41%-16.30%, 63.58%-70.44% at 14th, 28th, 42th, 56th days, respectively. These values were 15.18%-19.68%, 17.88%-20.84%, 60.24%-65.46% in the second group. In the M. Pectoralis Superficialis, percentage of white, intermediate and red muscle fibres were 59.99%-69.09%, 17.1 1%-22.93%, 12.48%- 17.08% in the different stage of growth (14th, 28th, 42th, 56th days). These values were 60.75%-65.73%, 19.23%-26.69%, 14.56%- 16.58% in the second group. The differences for the thickness of three types of muscle fibres in M. Biceps Femoris and M.
