Özet

Germinal centres of the“noduli lymphatici”of the cortex were enlarged and intensified by their cells. Lymphatic fol licles reached to a peak number on day eight, with the tingible body mac rophages (TBM) in their germinal centres plus the enlargment of me dullary cords. As conslusion, a high degree of stimulative action has been determined in the rat lymph nodes when treated with the salmonella paratyphi H an tigen with special reference to noduli lymphatici, leucocyte and macro phage proliferation and increase of the number of venules with high en dothelium. A prominent role could be attributed to the venules with high endothelium in the response to this antigen. However, further experiments should be done by immunolabelling and high-level electron microscopy techniques.

54 8. ÖZET (İNG.) ABSTRACT THE PRIMARY IMMUNE RESPONSE OF RAT LYMPH NODES TO SALMONELLA PARATYPHI ANTIGEN This research work has been done for being able to detect the response of lymphoid tissue of lymph nodes to a given antigen. A special reference has also been given to the venules with high endothelium. Twenty of the Wistar type rats were taken under the experiments. Eight of them were selected as controls, given parenteral 0.1 ml. NaCl solution from their cervical region. As for the stimulated group, sal monella paratyphi H antigen solution were injected into the subcutaneous tissue of the same region. Lymph nodes of mandibular and superficial cervical regions were removed from the control animals one, three, five days after the treatment of soduim chloride solution. The same lymph nodes have been taken from the rats already stimulated by salmonella paratyphi H antigen on time intervals as six hours, one, two, three, four, five and eight days after the antigen treatment. Tissues of the lymph nodes of control and antigenically stimulated groups were processed using rou tine paraffin techniques. Several staining methods were applied on par affin and semi-thin resin sections. Significant alterations were detected in the tissues of lymph nodes of antigen treated animals, namely in the cortical, paracortical, medully re gions and sinusoidal areas of the same. For the rats, stimulated by the antigen for six hours a prominent nom inal increase of neutrophil, eosinophil leucocytes and macrophages were clearly visible. Mast cells were crowded around the medullary sinus areas at the end of two day antigenic stimulation. Paracortex was enlarged. A highly increase of lymphocyte movements were found across the venules with high endotelium, reaching to a peak on day four of antigenic treat ment. On the third day of stimulation germinal centres of lymphatic nod ules started to react against the salmonella paratyphi H antigen. Leu cocytes and macrophages have synchronously get crowded in the medullary area. On the fourth day of antigenic stimulation plasma cells55 proliferated in the medullary cords and it continued during the fifth day. Parallel to the cellular proferations, venules with high endothelium have become increased in number.
