Özet

Son yıllarda tüketicilerin tüketime hazır durumdaki“taze gibi”ürünlere artan ilgisi, gıda endüstrisini, gıdaları işlemek için alternatif teknolojilere yönlendirmektedir. Bu teknolojilerden biri olan UV-C ışık teknolojisiyle gıdaların fiziksel ve duyusal özellikleri değişime uğratılmadan; yüzey dekontaminasyonun sağlanması amaçlanmaktadır. Üzümsü meyveler grubunda yer alan ahududu ve böğürtlen, kendilerine özgü renk, tat, koku ve yapısı ile taze tüketim için uygun olmasının yanında, gıda endüstrisinde de çok çeşitli alanlarda kullanılmaktadır. Ayrıca bu meyveler bazı bileşenler bakımından zengin olması nedeniyle insan sağlığına fayda sağlamaktadır. Ancak bu meyveler, hasat ve hasat sonrası koşullarının en iyi olduğu durumlarda dahi; mikrobiyal yükünden ve hasat sonrası yumuşamadan dolayı diğer meyvelere göre daha kısa bir depolama ömrüne sahiptir. Bu nedenden, çoğunlukla başka gıdalar için işlenmekte (reçel, marmelat, jöle) ya da dondurulmuş şekilde tüketilmektedir. Taze tüketim ömrünü uzatmak amacıyla yeni teknolojiler ile çalışmalar yapılmaktadır. Bu yüksek lisans tez çalışmasında öncelikle 8 lambalı UV-C sisteminde en uygun ışınlamanın belirlenmesi için dozimetre çalışması yapılmıştır. Daha sonra farklı doz (ahududu için; 33, 66 ve 100 kJ/m2, böğürtlen için; 50, 100 ve 150 kJ/m2 olmak üzere) ve doz hızlarında (2,3 ve 4,5 kJ/m2.min) UV-C ışığına maruz bırakılan ahududu ve böğürtlen meyvelerinin dekontaminasyonu ve kalite özelliklerinin değişimi araştırılmıştır. Meyvelerin fiziksel kalite olarak incelenen özellikleri; renk, sertlik ve pH, kimyasal kalite olarak incelenen özellikleri ise toplam fenolik, toplam flavonoid, toplam antosiyanin ve toplam antioksidan aktivitesidir. Bu özellikler 0. günde ve +4oC'de depolama sonrasında (ahududu için 2 gün, böğürtlen için 7 gün) incelenmiştir. UV-C teknolojisinin ahududu meyvelerinin doğal mikroflorası (toplam aerobik mezofilik bakteri ve toplam küf-maya) üzerine etkisi incelenmiştir. Ahududu meyvesinin başlangıç TAMB (toplam aerobik mezofilik bakteri) yükü; 8,31±0,11 log kob/g olarak belirlenmiştir ve UV-C uygulamasına bağlı olarak, 33, 66 ve 100 kJ/m2 doz uygulamalarında sırasıyla 1,24, 1,68 ve 1,41 log kob/g inaktivasyon sağlanmıştır (p<0,05). Ahududu meyvelerinin başlangıç toplam maya-küf sayısı ise 7,35±0,24 log kob/g olarak belirlenmiştir. UV-C uygulamasından sonra 0. günde bu sayıda istatistiksel olarak önemli inaktivasyon sağlanamazken, 2 günlük depolama sonrasında toplam maya-küf sayısındaki azalma kontrole göre; sırasıyla 33 kJ/m2 dozda 1,12; 66 kJ/m2 dozda 1,04 ve 100 kJ/m2 dozda ise 0,96 log kob/g olarak belirlenmiştir (p<0,05). Escherichia coli ve Staphylococcus aureus inoküle edilen böğürtlen meyvelere UV-C uygulanarak bakteri dekontaminasyonu incelenmiştir. İnokülasyon yapılan ve uygulama yapılmayan kontrol meyvelerinin E. coli yükü 4,16±0,26 log kob/g olarak belirlenmiştir. 0. günde 50, 100 ve 150 kJ/m2 doz uygulamasıyla böğürtlen örneklerinde kontrole göre E. coli inaktivasyonu sırasıyla 0,43, 0,80 ve 0,94 log kob/g olarak belirlenmiştir (p<0,05). S. aureus bakterisinde ise, inokülasyon sonrası kontrol meyvelerinin yükü 5,03±0,33 log kob/g olarak belirlenmiştir. 0. günde 50, 100 ve 150 kJ/m2 doz uygulanan örneklerin S. aureus yükü ise kontrole göre sırasıyla 1,78, 1,59 ve 1,83 log kob/g azalmıştır (p<0,05). S. aureus bakterisinin gram pozitif, E.coli'nin ise gram negatif olmasının inaktivasyon üzerine etkisi olabileceği düşünülmektedir. UV-C ışığın böğürtlen meyvesinin doğal toplam maya üzerine herhangi bir etkisi 0. günde ve 7. günde belirlenmemiştir. Doğal toplam küf sayısında ise, 7. günde 50, 100 ve 150 kJ/m2 doz uygulamalarında sırasıyla 0,80 log, 0,52 log ve 0,73 log toplam küf inaktivasyonu sağlanmıştır. Toplam doğal maya-küf sayımında 100 kJ/m2 doz uygulamasında kontrole göre 0,87 log inaktivasyon sağlanmıştır. 7 günlük depolama sonrasında böğürtlenlerdeki toplam maya-küf sayısındaki ortalama artış 1,70 log olarak belirlenmiştir (p<0,05). UV-C uygulaması yapılan ahududu ve böğürtlen meyvelerindeki renk değişimleri L, a, b, Chroma, hue açısı ve Delta E değerleri ile incelenmiştir. Özellikle düşük doz hızında meyveler 1 saati geçen sürelerde UV-C ışınlarına maruz kalmalarına rağmen, renk değerlerindeki değişimler istatistiksel açıdan önemli olsa da, sayısal anlamda çok büyük farklılık belirlenmemiştir. Ayrıca pH analizlerinde de benzer bir durumla istatistiksel farklılıkların sayısal anlamda çok büyük olmadığı ortaya konmuştur. Dolayısıyla UV-C uygulaması meyvede kararmaya ya da asitliğinin değişmesine neden olmamıştır. UV-C uygulamasının böğürtlen meyvesinin sertliğine etkisi incelendiğinde ise; uygulama dozuna ve doz hızına bağlı olarak 0. ya da 7. günde meyve sertliğinde değişim gözlenmemiştir (p>0,05), yalnızca 7 günlük depolamaya bağlı olarak meyvenin dokusunda yumuşama belirlenmiştir (p<0,05). UV-C uygulamasına bağlı olarak ahududu ve böğürtlen meyvelerinin toplam fenolik madde ve toplam antosiyanin miktarında önemli bir kayıp belirlenmemiştir. 0. günde ahududuların flavonoid değerlerinde önemli herhangi bir uygulama etkisi belirlenmezken, böğürtlende ise tüm uygulamalarda toplam flavanoid miktarında kontrole göre artış kaydedilmiştir (p<0,05). Tüm antioksidan analiz yöntemleriyle doz, doz hızı ve depolama etkileri farklı değerler ve eğilimler göstermiştir. Bu spektroskobik analizlerde meyvelerin içerdiği biyoaktif bileşenler ile farklı metotlardaki aktif bileşiklerin etkileşimi sonucunda değerler belirlendiğinden, bu tür farklılar raporlanabilmektedir. Sonuç olarak, ahududu ve böğürtlende UV-C uygulamalarında meyvenin pH, sertlik, renk gibi kalite indikatörü özellikleri ile, toplam antosiyanin, toplam fenolik, toplam flavonoid ve toplam antioksidan kapasitesi gibi besinsel öğelerinde kayıplar olmadan; TAMB, küf-maya, E. coli ve S. aureus yüklerinde önemli ölçüde inaktivasyon sağlanmıştır.

Recently, there has been a significant increase in demand for ready to eat and“fresh-like”food products. Therefore, studies on novel non-thermal technologies have increased to processing foods with minimal change in quality parameters such as nutritional value and sensory properties. One of these technologies, UV-C light treatment, has been successfully applied to reduce the microbial load on various food surfaces, liquids, and air environments. Berry fruits have a wide variety of uses in the food industry as well as fresh consumption because of its unique flavour, structure, and smell. Also, they are good source of essential vitamins and minerals, and have diverse phytochemical compositions that related to consumer health. However, even in the case of the best harvesting conditions; berry fruits have a shorter storage life than other fruits due to excessive softening and attack of microorganisms. For this reason, berry fruits are often processed for other foods like jam and marmalade or consumed frozen. Therefore, studies are being carried out with novel technologies in order to extend the fresh consumption life. In this master thesis, different UV-C treatment doses (33, 66 and 100 kJ/m2 for raspberry; 50, 100 and 150 kJ/m2 for blackberry) and different UV-C dose rates (2.3 and 4.5 kJ/m2.min) were applied to raspberries and blackberries. The effects of UV-C light treatment on decontamination and quality characteristics on raspberries and blackberries were investigated. Color, hardness and pH properties of the fruits as physical qualities; total phenolic, total flavonoid, total anthocyanin and total antioxidant activity of the fruits as chemical qualities and the decontamination levels of fruits were evaluated. The effect of UV-C treatment on natural microflora of raspberries (total aerobic mesophilic bacteria and total mold-yeast counts) was investigated. Initial TAMB counts of raspberries were found as 8.31±0.11 log cfu/g and the decontamination levels were; 1.24, 1.68 and 1.41 log cfu/g, respectively, in 33, 66 and 100 kJ/m2 doses UV-C treated raspberries, on day 0 (p<0.05). Initial mold/yeast counts of raspberries were found as 8.31±0.11 log cfu/g. Although there was no significant decontamination on day 0, after 2 days of refrigerated storage, the inactivation levels of mold/yeast counts on 33, 66 and 100 kJ/m2 UV-C treated fruits were; 1.12, 1.04 and 0.96 log cfu/g respectively (p<0.05). The effects of UV-C light treatment on decontamination of Escherichia coli and Staphylococcus aureus inoculated blackberries were evaluated. Initial E. coli load of inoculated control blackberries was 4.16±0.26 log cfu/g and the decontamination levels of E.coli on blackberries were; 0.43, 0.80 ve 0.94 log cfu/g respectively on 50, 100 and 150 kJ/m2 UV-C treated fruits, on day 0 (p<0.05). Initial S. aureus load of inoculated blackberries was 5.03±0.33 log cfu/g and the decontamination levels of S. aureus on blackberries were; 1.78, 1.59 ve 1.83 log cfu/g respectively after 50, 100 and 150 kJ/m2 UV-C treatment, on day 0 (p<0.05). It can be explained that, inactivation levels of UV-C light treatment may vary depending on the bacterial species. The UV-C light did not have any significant effects on the natural total yeast of blackberries on both day 0 and day 7. Inactivation of natural total mold counts were; 1.78, 1.59 ve 1.83 log cfu/g respectively on 50, 100 and 150 kJ/m2 UV-C treatred blackberries (p<0.05). 100 kJ/m2 UV-C treatment resulted in 0.87 log total yeast/mold inactivation than control. After 7 days of storage, the average increase in total yeast-mold count in blackberries was determined as 1.70 log cfu/g (p<0.05) Effect of UV-C light on L, a, b, Chroma, hue angle and Delta E values of raspberries and blackberries were determined. Although, the treatment time was one hour long in low dose rate, there was a significant but slight changes occurred in colour values of fruits. Also pH value changes were determined by UV-C light treatment. All treated raspberries were found higher pH value than control, on day 0 (p<0.05). UV-C treatment at 150 kJ/m² resulted in significantly higher pH than 100 kJ/m² and the control on day 0. This changes were significant but slight, therefore did not lead to change in quality of fruits. UV-C applications did not have any significant effect on firmness of blackberries both day 0 and day 7 (p> 0.05), only softening of the tissue of blackberries were determined after 7 days storage (p <0.05). The total phenolic content of untreated raspberry and blackberry fruits was found 203.45±18.69 and 260.29±16.77 mg GAE/100 g fw, respectively. The total anthocyanin content of untreated raspberry and blackberry fruits were found 32.86±2.24 and 82.62±3.48 mg cyn-3-glu/100 g fw, respectively. No significant loss was determined on the total phenolic and total anthocyanin content of raspberries and blackberries by UV-C light treatment. The total flavonoid content of untreated raspberries and blackberries was found 52.72±4.13 and 65.94±1.95 mg CTE/100 g fw, respectively. While no significant effect was determined on the total flavonoid content of raspberries, all treatments resulted in higher total flavonoid content in blackberries, on day 0 (p<0.05). Antioxidant capacity of raspberries were determined 129.60±6.21, 143.33±4.16 and 260.35±19.48 µmol TE/g fw by DPPH, ABTS and CUPRAC methods, respectively. Antioxidant capacity of blackberries were determined 258.84±13.66, 230.00±15.95 and 263.07±10.01 µmol TE/g fw by DPPH, ABTS and CUPRAC methods, respectively. Antioxidant capacity of fruits showed different trends with treatment dose, dose rate and storage in all methods. In these spectroscopic analyzes, such differences can be reported because of the interaction between the bioactive components contained in the fruits and the active compounds in different methods. In this study, the average light intensity of fruits on each side is not the same in low dose rate applications because of the 2 lamps are used from below and above of the fruits. Therefore, it is thought that sufficient homogeneous application could not be achieved in these applications. As a result, TAMB, total mold-yeast, E. coli and S. aureus counts of the raspberries and blackberries treated with different UV-C treatments were reduced significantly, without any significant changes in the physical and chemical properties. In order to prevent this problem, the same type of apparatus can be created for low dose rate and it can be achieved with 8 lower intensity lamps. At the high dose rate, since the 8 lamps give light on each side, it can be said that the fruits get the light more homogenously, but it is seen that there was a variation. To prevent this situation, the number of lamps can be increased and placed more often. Similarly, a system in which the food rotated 360° or lamps rotated 360° around the food can be considered in further studies to provide a more homogeneous application. In addition to UV-C light related factors such as dose, dose rate and application specifications; the surface roughness, current microorganism load and contamination ways (natural microflora or inoculation) of the fruits are also significant factors to ensure sufficient decontamination. Therefore, the use of UV-C in decontamination of berry fruits may considered has a potential whose initial microbial load is not very high. Enzyme activities, vitamin C content and bioavailability can be used in further studies to determine the physiological response of the berry fruit to UV-C application. UV-C combine technologies with controlled atmosphere storage, modified atmosphere packaging, immersion in protective solution or edible coating methods can be investigate to find its effectiveness on fruit.
