Özet

Amaç: Bu çalışmada farelerde indometazin ile oluşturulmuş gastrik ülser modelinde bir vazopeptidaz/anjiyotensin dönüştürücü enzim inhibitörü olan omapatrilatın gastroprotektif etkisinin araştırılması amaçlanmıştır. Hedefimiz, gastrik ülser hasarı oluşturulmuş farelerde omapatrilatın midede meydana getireceği farmakolojik ve biyokimyasal farklılıkları ortaya koymaktır. Materyal ve Metot: Erkek Balb C fareler 7 adet gruba ayrılmış, 24 saat torlu kafeslerde aç bırakılmış, sonrasında indometazin, omapatrilat ve famotidin oral yoldan verilmiştir. İndometazin uygulaması omapatrilat ve famotidin verildikten 10 dakika sonra yapılmıştır. İndometazin verildikten sonra fareler tekrar 6 saat aç bırakılmıştır. Bu sürenin sonunda mideler çıkarılmış, yıkanmış ve makroskobik olarak incelenmiştir. Mide dokuları gerekli incelemeler için uygun şartlarda saklanmıştır. Fare gruplarında SOD, ADE, Neprilisin aktiviteleri ile GSH ve MDA değerleri biyokimyasal yöntemlerle çalışılmıştır. Bulgular: Yapılan incelemelerde ülser grubundaki hasarın omapatrilat gruplarında azaldığı görülmüştür. En iyi antiülser etki gösteren doz 40 mg/kg omapatrilat dozu olmuştur. Ülser oluşturulan omapatrilat gruplarında doz artışına paralel bir şekilde SOD, GSH ve MDA değerleri sağlıklı gruba yakın ölçülmüştür. Yine sağlıklı gruba göre, omapatrilat uygulanan ülserli gruplarda doz arttıkça ADE ve Neprilisin aktivitelerinde de azalma gözlenmiştir. Pozitif kontrol grubu olan famotidin almış ülserli grubun sonuçlarının, 40 mg/kg omapatrilat alan ülserli grupun sonuçlarına en yakın ölçüldüğü belirlenmiştir. Sonuç: Omapatrilatın indometazinle oluşturulmuş ülsere karşı koruyuculuğu için gerekli dozun optimum 40 mg/kg olması gerektiği anlaşılmıştır. Bu dozda uygulandığında omapatrilat grubunun hem makroskobik ülser bulguları, hem de GSH, SOD, MDA, ADE ve Neprisilin bulguları açısından en iyi koruyuculuğu sağlamaktadır.

Objective: In this study, it is aimed to investigate the gastroprotective effect of omapatrilat, a vasopeptidase / angiotensin converting enzyme inhibitor, in a gastric ulcer model induced by indomethacin in mice. Our goal is to reveal the pharmacological and biochemical differences that omapatrilat will cause in the stomach in mice with gastric ulcer damage. Materials and Methods: Male Balb C mice were divided into 7 groups and fasted for 24 hours in cages. Afterwards indomethacin, omapatrilat, and famotidine were administered orally. Indomethacin was administered 10 minutes after omapatrilat and famotidine administration. After administration of indomethacin, the mice were fasted for another 6 hours. At the end of this period, the stomachs were removed, washed and examined macroscopically. Stomach tissues were stored under suitable conditions for necessary investigations. SOD, ADE, Neprilisin activities, GSH and MDA values were studied by biochemical methods in mouse groups. Results: In the examinations, it was observed that the damage in the ulcer group decreased in the omapatrilat groups. The dose that showed the best antiulcer effect was a dose of 40 mg / kg omapatrilat. Parallel to the dose increase in the omapatrilat groups with ulcers, SOD, GSH and MDA values were measured close to the healthy group. Again, compared to the healthy group, a decrease in ACE and Neprilisin activities was observed in the ulcer groups treated with omapatrilat as the dose increased. It was determined that the results of the positive control group, the group with ulcers that received famotidine, were measured closest to the results of the ulcer group receiving 40 mg / kg omapatrilat. Conclusion: It is understood that the optimum dose of 40 mg / kg should be required for the protection of omapatrilat against indomethacin-induced ulcers. When administered at this dose, it provides the best protection of the omapatrilat group in terms of both macroscopic ulcer findings and findings of GSH, SOD, MDA, ACE and Neprisilin.
