Özet
Fusarium wilting, root and root-rot diseases are negatively affecting the agricultural production especially vegetable greenhouse growing, throughout our country. Fusarium wilt disease causal agent Fusarium oxysporum Schlecht. f. sp. melonis (Fom) causes economical yield losses in melon cultivation. In the present study, virulence of Fom collected from different geographical locations of Turkey were determined by pathogenicity experiments. Phylogenetic variations at TEF1-α, actin and β-tubulin gene regions of pathogenic isolates of Fom were also, investigated with sequence analyses and in silico restriction enzyme digestions. The virulence groups were determined through the percentage of the disease severity values of the pathogenic Foc isolates, and 8 of this isolates were highly virulent (76-100%), 13 of them were virulent (51-75%), 8 of them are moderate-virulent (26-50%) and 22 of them were determined to be low virulent (60-69%). Phylogenetic analyses were carried out with TEF-1α and actin ve β-tubulin gene regions of a selected subset of Fom isolates, along with other forma specialis (f. sp.) and non-patogen (NP) grouping. Sequence data for the three regions displayed limited variation among Fom isolates along with other forma specialis and NP grouping. Fom isolates exhibited the same profile with AfaI, Csp6I, CviQI, RsaI and RsaNI restriction enzymes while there were differences between the enzyme cut profiles of BseMII, BspCNI, BstDEI, DdeI and HpyF3I. Variations in the beta-tubulin gene region have the potential to contribute to the development of diagnostic markers for Fom races idetification in the future.
