Begomoviruses belong to the family of Geminiviridae. Begomoviruses are usually transmitted by whitefly. Mechanical transmission of begomoviruses is rare. Mechanical transmissibility has been shown to be associated with the movement protein (MP) of begomovirus DNA-B in the tomato leaf curl New Delhi virus (ToLCNDV). A ToLCNDV-OM isolate isolated from diseased oriental melon can be mechanically transmitted, however, a ToLCNDV-CB isolate isolated from diseased cucumber cannot. Transgenic plants expressing the movement protein of ToLCNDV-OM or ToLCNDVCB under the control of an estrogen inducible promoter were developed. Plasmid constructs built in a binary vector were independently transformed into Nicotiana benthamiana plants by agrobacterium-mediated transformation. MP was expressed either in cytoplasm or nucleus to study how MP affects mechanical transmission. Movement protein-coding sequences in transgenic plants were confirmed by PCR, and movement protein expression was confirmed by enzyme-linked immunosorbent assay (ELISA). Expressing the ToLCNDV-OM movement protein in plants inoculated with the ToLCNDV-CB isolate by mechanical transmission showed infection occurred in nucleus, but no ToLCNDV-CB infection was seen in transgenic plants expressing ToLCNDV-CB movement protein. The result reveals that the expression of the ToLCNDV-OM movement protein in the nucleus helped ToLCNDV-CB become mechanically transmissible.
