Neonectria fuckeliana is becoming an important issue as it causes significant damage on spruce species, mainly Picea sitchensis (Sitka spruce) and Picea abies (Norway spruce), two of the most important commercial species in the UK and in Nordic countries. Infected trees lose commercial value and the fungus may cause mortality of substantial numbers of trees. The taxonomy of the fungus has changed several times. Neonectria fuckeliana is a complex of species more recently placed in the genus Corinectria, which includes Corinectria fuckeliana, Corinectria constricta and Corinectria tsugae. The purpose of this study was to characterise different N. fuckeliana isolates from the UK, analysing their morphological traits and molecular phylogeny to determine their true identity, and to examine the susceptibility of a range of different spruce species to infection by this pathogen. Isolates were incubated at different temperatures and, in a comparison of growth rates, clustered them into four clades. Sequences of four nuclear DNA regions, the internal transcribed spacer (ITS), translation elongation factor 1 (EF), actin and ß-tubulin were obtained, and all the isolates were identified as N. fuckeliana. These results suggest that in axenic culture, C. fuckeliana shows a variety of morphologies and growth rates. In the pathogenicity tests, six spruce species (Sitka spruce, Norway spruce, Picea. omorika (Serbian spruce) and 3 spruce hybrids (hybrid 1: Picea x lutzii from selected stand, Canadian; hybrid 2: Picea x lutzii from a registered seed orchard, Canadian; hybrid 3: PLU emblings, raised in tissue culture by private nursery) were inoculated with N. fuckeliana. Sitka spruce was the most affected species with the highest probability of mortality and bigger lesion sizes.
