Özet
Faba bean is a plant that plays a crucial role in human and animal nutrition, as well as the soil nitrogen content. Therefore, most of the faba bean breeding studies focus on increasing the yield. The dwarf gene has become common to increase the harvest index and the yield. In this research, it was aimed to find the genetic sequence of the dwarf gene in the faba bean. To determine that, as a genetic source NV153_1DW_S3A2 which has dwarf faba bean genome, normal flower colour and, dark hilum colour and NV644_1_KASZTEL (tall, white flower, pale hilum colour) inbred lines were crossed. To evaluate phenotypic traits, which are, flower colour, hilum colour, plant height in the F2 generation were recorded for phenotypic evaluation. The segregation ratio of F2 populations was tested with Chi-square tests for 3:1 single recessive gene inheritance, and the frequency distribution of the traits was founded. Moreover, in order to observe growth differences in dwarf and tall plants, the internode distance was recorded in the F2 generation during the cultivation process. To create a genetic map based on phenotypic traits young flowers' leaves were taken and analysed with SNP based PCR method with KASP assay. Then from 179 SNPs markers 112 individuals the genetic map was created by MapDisto 2.1. Two more maps were created both for the quantitative trait and qualitative traits. From the phenotypic traits map, the synteny was explored, the interval of the gene that was responsible for dwarfism, flower colour, and hilum colour was founded according to collinearity between the faba bean chromosome with Medicago. It was concluded with the Medtr3g092830 gene responsible for the colour of the flowers. While Vf_Mt2g012470_001 and Vf_Mt2g013690_001 was assumed as a candidate gene for hilum colour, for dwarfism the previous gene of Vf_Mt2g104110_001 was evaluated to as candidates. 13 However, according to the QTL map, it was founded that the linkage group two had a“tall”QTL peak in the vicinity of the markers that were already identified as flanking the dwarfing gene.
