Özet

Kemoterapide sıklıkla kullanılan Paklitaksel (PTX)'in karaciğer dokusunda oluşturduğu toksik etki klinik uygulamasını sınırlandırmaktadır.

The toxic effect of paclitaxel (PTX) on liver tissue, frequently used in chemotherapy, limits its clinical application. In this thesis, it is aimed to investigate the protective effect of parthenolide (PTL) against hepatotoxicity caused by PTX exposure at gene and protein levels. In this thesis 48 male rats were divided into 6 groups control, PTX, sham, and 3 different treatment groups treated with PTL at different doses. After two weeks of PTX exposure, 1, 2, and 4 mg/kg PTL were administered to the treatment groups for two weeks. Liver tissues from all groups were collected. Oxidative damage markers, DNA damage markers, inflammation markers, and changes in antioxidant system components at gene and protein levels were examined. For this purpose, spectroscopic, histochemical, immunohistochemical, and Real Time PCR methods were used. While the levels of GSH and MDA, which are markers of oxidative stress, 8-OhDG level, which is a marker of DNA damage, and the expressions of Tnfa, Il-6, Foxo1, Foxo3, NFkB, Cox-2, which are known as inflammation markers genes, were significantly altered by PTX exposure, It reached the same level as the control group with PTL treatment as the dose-dependent manner. As a result of histopathological analysis, it was determined that severe necrotic and degenerative hepatocytes due to PTX turned into mildly necrotic and degenerative hepatocytes in the 4 mg/kg PTL group. While the gene expression levels and enzymatic activities of SOD, CAT, GPX, GR, and GST, which are antioxidant system components, were significantly increased/decreased with PTX, the changes reached the control group levels with PTL treatment in a dose-dependent manner. In conclusion, it was observed that the oxidative stress induced in the rat by PTX treatment was eliminated by PTL treatment in a dose-dependent manner and protected the rat against hepatotoxicity. The interesting part here is that the point of action of PTL was seen at the protein level rather than the gene level, especially in the 4 mg/kg treatment groups.
