Özet
52 ÖZET Koçlarda Sinaptonema Kompleksi' nin Histomorfolojisi Üzerinde Araştırmalar Bu çalışmanın amacı, koçların primer spermatositlerinde sinaptonema kompleksi analizi ile kromozomların eşleşme davranışını incelemektir. Primer spermatositler için Counce ve Meyer'e ait mikroyayma tekniğinin modifikasyonu uygulandı. Bu uygulama ile hücre membranlarının yapısı bozularak, sinaptonema kompleksi'nin, iki boyutlu olarak üç paralel iplik şeklinde görülmesi sağlandı. Protein yapısındaki bu üçlü kompleks, bivalanlar boyunca şekillenmişti. Fosfotungstik asit boyamasından sonra, lateral elementler yoğun olarak boyandı ve bunların arasında daha zayıf boyanan sentral element ayırt edildi. Gümüş boyamasından sonra ise sadece lateral elementler görüldü. Bu çalışmada, iki profaz aşaması- zigoten ve pakiten- tanımlandı. Otozomal kromozomlar, zigotende uçlarından sinapsise başladı; en küçük sinaptonema komplekslerinin birkaçı sinapsisi tamamlamıştı. Seks kromozomları geç zigotende eşleşmeye başladı; bu alt aşamada eşleşmemiş segmentler farklılaşma göstermedi. Otozomal kromozomlar, sinapsisi pakitende tamamladı. Bunlar tipik sinaptonema kompleksi yapısını gösterdi. Kinetokor, eksenler üzerinde yoğun boyanmış bir yapı olarak tanımlandı.

53 SUMMARY Investigations on the Histo morphology of Synaptonemal Complex in the Ram The principal aim of this study was to investigate chromosome pairing behaviour by synaptonemal complex analysis in the Ram's primer spermatocytes. A modification of the Counce and Meyer microspreading technique for primary spermatocytes was applied. This process was disrupted the cell membranes; the synaptonemal complex was appeared as three parallel threads in a two dimensional image. A tripartite proteinaceous complex had formed along the bivalents. After phosphotungstic acid staining, the lateral elements were stained densely and between them the weaker central element was distinguished. After silver staining, the lateral elements was stained, only. In this study, two prophase stages - zygotene and pachytene - were identified. The autosomes initiated synapsis terminally at zygotene, some of the smaller synaptonemal complexes had completed synapsis. The sex chromosomes started pairing at late zygotene; at this substage the free segments of the sex chromosomes did not show differentiation. The autosomes completed synapsis at pachytene. They showed a typical synaptonemal complex structure. The kinetochore was identified as a densely stained structure on the axes.
