Rekombinant peptidlerin antifungal aktivitesi Candida glabrata (EUCC T806), Candida glabrata (EUCC G1), Trichosporon asahii (EUCC T3), Candida albicans (ATCC 10231), Candida albicans (ATCC 90029), Candida parapsilosis (EUCC P2), Candida tropicalis (NRRL Y-12968), Candida membranifaciens (EUCC M53), Candida zeylanoides (EUCC E11), Pichia kluyveri (NRRL Y-17774), Pichia fermentas (NRRL Y-11506) ve Yarrowia lipolytica (NRRL Y-1094) olmak üzere 12 maya suşuna karşı test edilmiştir.
Antifungal peptides (AFPs) are potential compounds for the treatment of fungal infections and control of fungal contamination of food spoilage. However, natural AFPs have disadvantages such as toxicity, poor solubility and low stability, which limit their application in various fields. In order to increase the efficacy of AFPs and reduce unwanted side effects, new alternative peptides to natural AFPs can be designed by modifying their physicochemical properties. Therefore, in this thesis, 2 antifungal peptide sequences were designed. For the design of the peptides, a design strategy based on the identification of motifs in peptides with high antifungal activity selected from the DBAASP database and the combination of these motifs to form new peptide sequences was applied. Production of recombinant peptides (rC40-6His and rM42-6His) was carried out in Pichia pastoris (Komagataella phaffi) GS115 expression system. The nucleotide sequences encoding rC40-6His and rM42-6His peptides were generated by recursive PCR technique. The target DNA sequences were cloned in Escherichia coli DH5α cells and transformed into P. pastoris GS115 cells after binding to pPICZαA, both cloning and expression vector. Recombinant P. pastoris GS115 transformants were induced with 1% methanol in shaking culture medium for 96 hours to produce rC40-6His and rM42-6His peptides. The concentration of rC40-6His and rM42-6His peptides purified after production was determined as 142.5 mg/L and 172.8 mg/L, respectively. The antifungal activities of rC40-6His and rM42-6His peptides were tested against 12 yeast strains including Candida glabrata (Type 806), Candida glabrata (EUCC G1), Trichosporon asahii (EUCC T3), Candida albicans (ATCC 10231), Candida albicans (ATCC 90029), Candida parapsilosis (EUCC P2), Candida tropicalis (NRRL Y-12968), Candida membranifaciens (EUCC M53), Candida zeylanoides (EUCC E11), Pichia kluyveri (NRRL Y-17774), Pichia fermentas (NRRL Y-11506) and Yarrowia lipolytica (NRRL Y-1094). Agar diffusion analysis showed inhibition zone formation in all yeast strains except C. albicans (ATCC 90029), C. parapsilosis (EUCC P2) and C. membranifaciens (EUCC M53). The minimum inhibition concentrations (MIC) ranged between 4-128 µg/ml for rC40-6His peptide and 4->128 µg/ml for rM42-6His peptide. The hemolytic activity of the recombinant peptides was tested against mouse erythrocytes. rC40-6His and rM42-6His peptides were found to have hemolytic activity at a concentration of 64 µg/ml with a hemolysis rate of 93.6% and 93.53%, respectively. In conclusion, new peptide sequences with antifungal activity were designed within the scope of the thesis study and the production of the designed peptide sequences was successfully realized in the P. pastoris expression system. It is thought that the peptides produced have the potential to be used in various fields, especially in the food industry. However, further studies are needed to reduce the hemolytic activity of the peptides in order to make their use possible.
