Colorectal cancer (CRC) ranks third among all cancers in terms of global incidence. Biomarkers used for the diagnosis of CRC, which causes the death of approximately 900 thousand people annually, are insufficient due to the heterogeneous structure of the disease and complex development processes. Therefore, identifying the key molecular mechanisms in colorectal cancer and defining prognostic biomarkers that allow rapid diagnosis will contribute to both increasing the response of patients to treatment and minimizing the economic loss caused by incorrect treatment strategies. Competitive endogenous RNAs (ceRNAs) share microRNA response elements (MREs). Thus, they can inhibit microRNAs (miRNAs) through common MREs and regulate each other's expression. RNAs that code (messenger RNA-mRNA) and do not code as ceRNAs act as“miRNA sponges”. ceRNAs and miRNAs form complex regulatory networks called the competitive endogenous RNA network (ceRNET). Studies on cancer, in particular, yield results supporting the use of ceRNAs as independent biomarkers. In this context, despite having many unknowns, the discovery of endogenous miRNA sponges and ceRNET guides the functionalization of a large part of the transcriptome. It also has the potential to enable the development of new and exciting treatment strategies targeting key nodes in ceRNETs to be added to miRNA therapy approaches. Within the scope of the thesis, in addition to the validation of the four candidate biomarkers (THBS2, FNI, COL1A1 and COL5A1) at the RNA and protein levels, it is aimed to reveal the ceRNET in oncogenesis and metastasis processes in colorectal cancer, to identify potential molecules that play a key role in pathogenesis, and to question their usability as biomarkers. For this purpose, a prospective sample of 180 patients was created. Protein and mRNA levels of related genes were analyzed in this prospective sample. At the same time, RNA sequencing was performed from RNA obtained from fresh tissue and ceRNET regulators involved in metastatic processes were identified. As a result, it was shown that COL1A1, COL5A1 and THBS2 expression levels were significantly increased in primary and metastatic tumor tissues compared to normal tissues. In addition, a significant increase in COL1A1 and COL5A1 expression levels in metastatic tumor tissue compared to primary tumor tissue revealed results supporting their prognostic biomarker potential in relation to metastasis. In protein analyzes, it was shown that COL1A1, COL5A1 and THBS2 protein levels were significantly increased in metastatic CRC patients compared to non-metastatic CRC patients. This situation revealed results supporting the prognostic biomarker potential of COL1A1, COL5A1 and THBS2 protein levels in relation to metastasis.
