Özet
Amaç: Bu çalışmanın amacı, elajik asidin sirkadiyen ritim gen ekspresyonları üzerindeki düzenleyici etkilerini, karaciğer hastalıklarıyla ilişkili hücresel mekanizmalar çerçevesinde HepG2 hücre kültüründe in vitro olarak araştırmaktır. Gereç ve Yöntem: İnsan hepatokarsinom hücre hattı kullanılarak, elajik asit (0-100 μM) ve glukozamin (0-50.000 μM) uygulamaları yapılmıştır. Hücre canlılığı MTT testi ile değerlendirilmiş, gen ekspresyonları qRT-PCR yöntemiyle analiz edilmiştir. Reaktif oksijen türleri düzeyleri, mitokondriyal membran potansiyeli ve glikoz alımı belirlenmiştir. Veriler tek yönlü ANOVA ile analiz edilmiştir. Bulgular: Elajik asitin IC50 değeri 25 µM olarak belirlenmiştir. Glukozamin uygulaması, BMAL1 (p<0,0005), CLOCK (p<0,0001) ve GLUT4 (p<0,05) gen ekspresyonlarını anlamlı şekilde artırmıştır. Elajik asit, glukozaminin etkilerini tersine çevirerek BMAL1 (p<0,0005) ve GLUT4 (p<0,005) gen ekspresyonlarını anlamlı şekilde azaltmıştır. CLOCK gen ekspresyonunda ise elajik asit, glukozaminle karşılaştırıldığında anlamlı bir değişiklik göstermemiştir. Mitokondriyal membran potansiyelinde, uygulamalar arasında anlamlı bir fark gözlenmemiştir (p>0,05). Glukozamin, reaktif oksijen türleri seviyelerini artırmış (p<0,0001), elajik asit bu artışı doza bağlı olarak baskılamıştır (p<0,0001). Glikoz alımında doza bağlı olarak artış görülmüştür (p<0,0001). Glukozaminle oluşturulan insülin dirençli hücrelerde ise 25 µM ve 50 µM elajik asit kombinasyonları glikoz alımını anlamlı şekilde azaltmıştır (p< 0,0001). Sonuç: Elajik asit, glukozamin ile oluşturulan metabolik stres koşullarında sirkadiyen ritimle ilişkili gen ekspresyonlarını düzenleyici bir etki göstermiştir. Reaktif oksijen türleri seviyelerindeki düşüş ve mitokondriyal membran potansiyelinde stabilite, elajik asidin hücresel fonksiyonlar üzerindeki koruyucu etkilerini desteklemektedir. Bu bulgular, elajik asidin metabolik bozuklukların tedavisinde potansiyel bir terapötik ajan olabileceğini göstermektedir.

Aim: The objective of this study was to examine the mechanisms linking glycolipid metabolism in hepatocytes with circadian rhythm-related processes, focusing on the cellular mechanisms associated with liver diseases. Materials and Methods: Human hepatocellular carcinoma (HepG2) cells were utilized for the study. Various concentrations of ellagic acid (0-100 μM) and glucosamine (0-50,000 μM) were applied to the cells. Cell viability was assessed using the MTT assay, and gene expression was analyzed through qRT-PCR. Levels of reactive oxygen species, mitochondrial membrane potential, and glucose uptake were measured. The data were analyzed using one-way ANOVA. Results: The IC50 value of ellagic acid was determined to be 25 µM. Glucosamine treatment led to significant increases in the expression of BMAL1 (p<0.0005), CLOCK (p<0.0005), and GLUT4 (p<0.05). Ellagic acid reversed the effects of glucosamine, significantly decreasing the expression of BMAL1 (p<0.0005) and GLUT4 (p<0.005), but no significant change was observed in CLOCK gene expression. There were no significant differences in mitochondrial membrane potential (p>0.05). Glucosamine increased reactive oxygen species levels (p<0.0005), while ellagic acid suppressed this increase in a dose-dependent manner (p<0.0005). A dose-dependent increase in glucose uptake was observed (p<0.0005). In glucosamine-induced insulin-resistant cells, combinations of 25 µM and 50 µM ellagic acid significantly reduced glucose uptake (p<0.0005). Conclusion: Ellagic acid showed a regulatory effect on circadian rhythm-related gene expression in hepatocytes under glucosamine-induced metabolic stress conditions. The reduction in reactive oxygen species levels and the stability of mitochondrial membrane potential suggest protective effects of ellagic acid on cellular functions. These findings highlight the potential of ellagic acid as a therapeutic agent for managing metabolic disorders.
