Özet
Bütün dünyada ölüm sebeplerinin başlıcaları arasında kanser yer almaktadır. Kadınlarda en sık görülen jinekolojik malignite olan endometriyum kanseri, genç yaşlarda bile artan morbiditeye sahiptir. Bunun yanı sıra yayılma ve ilerleme mekanizması hala araştırılmaya muhtaçtır. Bu sebeple endometriyum kanserinin tanı ve tedavisinde yeni yaklaşımların geliştirilebilmesi amacıyla yapılan çalışmalar son yıllarda artış göstermekte ve elde edilen veriler önem kazanmaktadır. Son yıllarda yapılan çalışmalarda; bitkisel kaynaklı besinlerin ve bu besinlerin aktif bileşenlerinin, kanser tedavilerine ek olarak kullanımı ve kansere karşı etkileri üzerine yeni yaklaşım arayışları artmıştır. Epigallokateşin gallat anti-kanser, antioksidan vb. etkileri olan bir polifenoldür. Bu sebeple Epigallokateşin gallatın anti-kanser etkisinin moleküler mekanizmasının aydınlatılmasına yönelik yoğun çalışmalar yapılmaktadır. Epigallokateşin Gallat (EGCG)'ın Ishikawa hücreleri üzerindeki etkileri kapsamlı analizlerle değerlendirilmiştir. Epigallokateşin gallatın sitotoksik etkisinin değerlendirildiği MTT testi sonucunda; Epigallokateşin gallatın IC50 dozu belirlenmiş ve sonraki deneylerde bu doz kullanılmıştır. Epigallokateşin gallatın IC50 dozunda hücre canlılığında anlamlı bir azalma gözlenmiştir. İmmünohistokimyasal boyamada; IC50 dozu uygulanan grupta Bax ve P53 ekspresyonunda anlamlı bir artış, Bcl-2 ekspresyonunda ise anlamlı bir azalma olduğu görülmüştür. Bu durum; Epigallokateşin gallatın pro-apoptotik etkisini ortaya koymaktadır. DAPI ile yapılan immünofloresan boyamada; Epigallokateşin gallatın IC50 dozunun uygulandığı grupta boyanan hücre çekirdeklerinin sayısında, kontrol grubuna kıyasla istatistiksel olarak anlamlı bir azalma görülmüştür. Yara iyileşme (wound healing) testi sonuçları, Epigallokateşin gallatın hücre göçünü belirgin şekilde inhibe ettiğini ortaya koymuştur. TUNEL boyaması sonuçlarına göre; pozitif boyanan hücre sayısı, Epigallokateşin gallatın IC50 dozu grubunda anlamlı bir artış göstererek Epigallokateşin gallatın apoptoz indükleyici etkisini desteklemiştir. Biyokimyasal analizlerde; Epigallokateşin gallat uygulamasının Total Antioksidan Seviyesi (TAS)' nin kontrol grubuna kıyasla anlamlı şekilde artırdığı (p<0,05) görülmüş ancak sodyum, potasyum ve klorür gibi elektrolit seviyelerinde istatistiksel olarak anlamlı bir değişiklik gözlenmemiştir. Çalışmamızda; insan endometriyal hücre hattı olan Ishikawa hücrelerinde Epigallokateşin gallatın antioksidan etki gösterdiği, apoptozisi indüklediği, hücre proliferasyonunu baskıladığı ve hücre göçünü azalttığı görülmüştür. Bu sonuçlar; Epigallokateşin gallatın endometriyum kanser tedavisi için umut verici bir yaklaşım olma ihtimalini güçlendirmektedir. Epigallokateşin gallat ile daha fazla çalışma yapılması; endometriyum kanser tedavisinde yeni yaklaşımların elde edilebilmesinde ve klinik açıdan da anlamlı bilgi birikimi sağlanmasında etkili olacaktır.

Cancer is one of the leading causes of death worldwide. Endometrial cancer, the most common gynecologic malignancy in women, has increasing morbidity even in young individuals. Additionally, its mechanisms of spread and progression still require further research. Consequently, studies aimed at developing new approaches for the diagnosis and treatment of endometrial cancer have increased in recent years, and the data obtained are becoming increasingly important. Recent studies have explored new approaches to the use of plant-based foods and their active compounds as complementary treatments for cancer. Epigallocatechin gallate (EGCG) is a polyphenol with anti-cancer, antioxidant, and various other effects. Therefore, extensive research has been conducted to elucidate the molecular mechanism of EGCG's anti-cancer properties. The effects of Epigallocatechin Gallate (EGCG) on Ishikawa cells have been evaluated through comprehensive analyses. In the MTT assay, which assesses the cytotoxic effect of EGCG, the IC50 dose was determined, and subsequent experiments were conducted using this dose. The results indicated a significant decrease in cell viability at the IC50 dose. Immunohistochemical staining revealed a significant increase in Bax and P53 expression and a notable decrease in Bcl-2 expression in the IC50 dose group, demonstrating the pro-apoptotic effect of EGCG. In immunofluorescence staining with DAPI, a statistically significant decrease in the number of stained cell nuclei was observed in the IC50 dose group compared to the control group. The wound healing assay results showed that EGCG significantly inhibited cell migration. According to the TUNEL assay results, the number of positively stained cells showed a significant increase in the IC50 dose group, supporting the pro-apoptotic effects of Epigallocatechin gallate. Biochemical analyses revealed that EGCG treatment significantly increased the Total Antioxidant Status (TAS) compared to the control group (p<0.05), but no statistically significant changes were observed in sodium, potassium, or chloride levels. In this study, EGCG exhibited antioxidant properties, induced apoptosis, suppressed cell proliferation, and reduced cell migration in the human endometrial cancer cell line Ishikawa. These findings reinforce the potential of EGCG as a promising therapeutic approach for endometrial cancer treatment. Further research on EGCG could contribute to new treatment strategies and provide clinically meaningful insights into endometrial cancer therapy.
