Özet

GİRİŞ-AMAÇ: Bitki polifenollerinin nörodejeneratif hastalıklar üzerinde olumlu etkileri olduğu birçok çalışmada gösterilmiştir. Epigallokateşin-3-gallat (EGCG) ve kurkumin antioksidan polifenollerdir. Bu çalışmada SH-SY5Y hücre hattında hidrojen peroksit (H2O2) hasarına karşı EGCG ve kurkumin ön uygulamasının koruyucu etkilerinin gösterilmesi amaçlanmıştır. YÖNTEMLER: Çalışmamızda SH-SY5Y hücre hattına çeşitli dozlarda, 1 saat süre ile H2O2 uygulandı. H2O2'nin LD50 dozu 100 µM olarak belirlendi. EGCG ve kurkuminin koruyucu etkilerini saptamak amacıyla 0,1 µM EGCG, 1 µM EGCG, 10 µM EGCG, 10 nM Kurkumin, 50 nM Kurkumin, 100 nM Kurkumin dozları ve bunların kombinasyonları, 24 saat süre ile ön tedavi olarak uygulandı. Ardından hücreler 1 saat süre ile, 100 µM H2O2'ye maruz bırakıldı. Uygulamalar sonucunda hücre canlılığı MTS testi ile, koloni oluşturma yetenekleri klonojenik test ile, proliferasyon kapasitesi ise Ki-67 testiyle belirlendi. Ayrıca hücre ölüm tipini değerlendirmek için Kaspaz-3 ELISA testi uygulandı. İstatistiksel analiz için one-way ANOVA kullanıldı. BULGULAR: Hücre hattında 1 saatlik H2O2 uygulamasının hücre popülasyonunun yaklaşık yarısını öldürdüğü görülmüştür (p<0,0001). Bir saat H2O2 (100 µM) uygulaması öncesinde 24 saat süreyle 0,1 µM EGCG (p<0,0001), 10 nM kurkumin (p<0,0001), 0,1 µM EGCG + 10 nM Kurkumin (p<0.0001), 1 µM EGCG + 10 nM kurkumin (p < 0.001) ve 0,1 µM EGCG + 50 nM kurkumin (p < 0.001) ön tedavisi uygulandığında hücre sağ kalımının arttığı gözlenmiştir. 100 µM H2O2'nin koloni oluşturma yeteneğini anlamlı olarak azalttığı, ayrıca 0,1 µM EGCG (p<0,0001), 10 nm kurkumin (p<0,005) ve 0,1 µM EGCG + 10 nM kurkumin (p<0,0001) ön tedavisinin koloni oluşturma yeteneğini anlamlı olarak arttığı görülmüştür. EGCG ön tedavisinin 0,1 µM ve 1 µM dozlarında, Ki-67 ile bağlanma yüzdelerini anlamlı olarak arttığı bulunmuştur (p<0.001). Kurkumin ise 50 nm uygulandığında, Ki-67 ile bağlanma yüzdelerini artırmıştır (p<0.001). Kombinasyon tedavilerinin (0,1 µM EGCG + 10 nM kurkumin, 1 µM EGCG + 10 nM kurkumin, 0,1 µM EGCG + 50 nM kurkumin ve 1 µM EGCG + 50 nM kurkumin), Ki-67 ile bağlanma yüzdelerini arttığı (p<0.001) ve dolayısıyla hücre proliferasyonu artırdığı bulunmuştur. EGCG ön tedavisinin 0,1 µM (p <0.0001) ve 1 µM (p<0.005) dozlarında aktif kaspaz-3 protein miktarını toksisite uygulanan gruplara göre anlamlı olarak azalttığı bulunmuştur. Kurkuminin ise 10 nm (p <0.0001) ve 50 nm (p<0.005) dozlarında aktif kaspaz-3 protein miktarını azalttığı bulunmuştur. Kombinasyon tedavilerinden 0,1 µM EGCG + 10 nM kurkumin (p<0.0001), 0,1 µM EGCG + 50 nM kurkumin, 1 µM EGCG + 10 nM kurkumin ve 1 µM EGCG + 50 nM kurkumin (p<0.005) uygulamalarının aktif kaspaz-3 protein miktarını azalttığı ve dolayısıyla apopitoz oranını azaltıcı etkileri olduğu saptanmıştır. SONUÇ: Çalışmamız, H2O2'nin apoptotik hücre ölümünü arttırarak hücre canlılığını azalttığını ortaya koymuştur. 0,1 µM EGCG, 10 nM Kurkumin dozları ve bunların kombinasyonları H2O2'nin sebep olduğu nöronal hasarı azaltmıştır. 0,1 µM EGCG, 1 µM EGCG, 10 nM Kurkumin, 50 nM Kurkumin dozları ve bunların kombinasyonları ise kaspaz-3 düzeylerini azaltmıştır. Ayrıca 0,1 µM EGCG, 1 µM EGCG, 50 nm kurkumin, 0,1 µM EGCG + 10 nM kurkumin, 1 µM EGCG + 10 nM kurkumin, 0,1 µM EGCG + 50 nM kurkumin ve 1 µM EGCG + 50 nM kurkumin uygulamasının hücre proliferasyonunu artırdığı görülmüştür. Sonuçlarımız bu ajanların nörodejenerasyona karşı koruyucu ajanlar olarak kullanılabileceğini göstermektedir. Ancak epigallokateşin gallat ve kurkumin kombinasyonlarının tek başlarına uygulandığından daha güçlü etki göstermediği belirlenmiştir.

INTRODUCTION AND AIM: Many studies have shown that plant polyphenols have positive effects on neurodegenerative diseases. Epigallocatechin-3-gallate (EGCG) and Curcumin are antioxidant polyphenols. The aim of this study is to demonstrate the effects of EGCG and curcumin pre-treatment against hydrogen peroxide (H2O2) damage in SH-SY5Y cell line. METHODS: Various doses of H2O2 were administered to the SH-SY5Y cell line for 1 hour in the study. The LD50 dose of H2O2 was determined as 100 µM. In order to determine the protective effects of EGCG and curcumin, 0,1 µM EGCG, 1 µM EGCG, 10 nM curcumin, 50 nM curcumin doses and their combinations were administered as a 24-hour pretreatment. It was exposed to 100 µM H2O2 for 1 hour at the end of the application. As a result of the applications, cell viability was determined by MTS test, and colony formation abilities were determined by clonogenic test and proliferation capacity was determined by Ki-67 assay. Caspase-3 ELISA was performed to assess the type of cell death. One-way ANOVA was used for statistical analysis. RESULTS: Our results showed that 100 µM H2O2 manifested decrease approximately half of the cell population (p<0.0001). It was observed that cell survival increased when 0.1 µM EGCG (p<0.0001), 10 nM curcumin (p<0.0001), 0.1 µM EGCG + 10 nM curcumin (p<0.0001), 1 µM EGCG + 10 nM curcumin (p < 0.001) and 0.1 µM EGCG + 50 nM curcumin (p<0.001) pretreatment was applied for 24 hours before H2O2 (100 µM) application for one hour. It was observed that 100 µM H2O2 significantly reduced the colony formation ability, and also 0.1 µM EGCG (p<0.0001), 10 nm curcumin (p<0.005) and 0.1 µM EGCG + 10 nM curcumin (p<0.0001) pretreatment significantly increased the colony forming ability. EGCG pretreatment was found to increase significantly binding percentages with Ki-67 at 0,1 µM and 1 µM doses (p<0.0001) and thus increase cell proliferation. Curcumin, on the other hand, was found to increase the binding percentages with Ki-67 when 50 nm was applied (p<0.0001). Combination treatments (0.1 µM EGCG + 10 nM curcumin, 1 µM EGCG + 10 nM curcumin, 0.1 µM EGCG + 50 nM curcumin, and 1 µM EGCG + 50 nM curcumin) increased the binding percentages with Ki-67 (p< 0.001) and therefore was found to increase cell proliferation. It was found that EGCG pretreatment at doses of 0,1 µM (p <0.0001) and 1 µM (p <0.005) significantly reduced the amount of active caspase-3 protein compared to the only toxicity groups. It was found that pretreatment of curcumin at the dose of 10 nm (p < 0.0001) and 50 nm (p < 0.005) reduced the amount of active caspase-3 protein. Of the combination treatments, 0,1 µM EGCG + 10 nM curcumin (p<0.0001) and 0,1 µM EGCG + 50 nM curcumin, 1 µM EGCG + 10 nM curcumin and 1 µM EGCG + 50 nM curcumin (p<0.005) applications significantly decreased the active caspase-3 levels. Therefore, these treatments reduces the rate of apoptosis. CONCLUSION: Our study revealed that H2O2 decreases cell viability by increasing apoptotic cell death. Doses of 0,1 µM EGCG, 1 µM EGCG, 10 nM curcumin, 50 nM curcumin and their combinations reduced the neuronal damage caused by H2O2. Doses of 0.1 µM EGCG, 1 µM EGCG, 10 nM curcumin, 50 nM curcumin and their combinations reduced caspase-3 levels. Additionally, it was observed that the application of 0,1 µM EGCG, 1 µM EGCG, 50 nm curcumin, 0.1 µM EGCG + 10 nM curcumin, 1 µM EGCG + 10 nM curcumin, 0.1 µM EGCG + 50 nM curcumin, and 1 µM EGCG + 50 nM curcumin increased cell proliferation. Our results suggest that these agents can be used as protective agents against neurodegeneration. However, it has been determined that epigallocatechin gallate and curcumin combinations do not have a stronger effect than when applied alone.
