Özet
Plastik kirliliği, gıda zincirine dahil olan mikroplastikler (MP) yoluyla insan sağlığı için potansiyel bir risk oluşturan küresel bir sorundur. Gıdalar gibi kompleks matrislerden MP'lerin doğru bir şekilde tespiti, standart ve etkin izolasyon yöntemlerinin eksikliği nedeniyle önemli bir bilimsel zorluk teşkil etmektedir. Bu tez çalışması, gıdalardaki polistiren (PS) mikroplastiklerinin izolasyonu ve UV ışık altında optik mikroskop ile tespiti için etkin ve hızlı bir kimyasal sindirim protokolü geliştirmeyi amaçlamıştır. Çalışma kapsamında, farklı kimyasalların (KOH, HCl), konsantrasyonların, sıcaklıkların ve sürelerin, normal (NM) ve yüksek yağlı (YM) gıda modelleri üzerindeki sindirim performansı sistematik olarak incelenmiştir. Geliştirilen protokollerin hedef PS partiküllerinin morfolojisine ve Nil Kırmızısı (NK) ile boyanma performansına olan etkileri de değerlendirilmiştir. Sonuçlar, literatürde yaygın olarak kullanılan 24-48 saatlik sindirim sürelerinin, 85°C gibi yüksek sıcaklıklarda KOH kullanılarak 2-4 saate kadar düşürülebildiğini ve bu esnada %95'in üzerinde sindirim verimine ulaşıldığını göstermiştir. Hızlandırılmış bu koşulların, 40-560 µm boyut aralığındaki PS partiküllerinde mikroskopla tespit edilebilir bir hasara yol açmadığı doğrulanmıştır. Çalışmanın kritik bulgularından biri, asit (HCl) sindiriminin PS partiküllerinin yüzey kimyasını değiştirerek NK boyası ile etkileşimini bozması ve bu nedenle bu analitik iş akışı için uygun olmadığının tespit edilmesidir. Optimize edilen KOH bazlı protokoller ile yapılan doğrulama çalışmalarında, normal model için yaklaşık %97 ve yüksek yağlı model için yaklaşık %108 geri kazanım oranları elde edilmiştir. Bu tez, PS mikroplastiklerinin gıda matrislerinden analizi için hem hızlı hem de yüksek geri kazanım verimine sahip bir ön işlem metodu sunmaktadır. Çalışma, yüksek sıcaklıkta sindirimin etkinliğini ortaya koyması ve asit sindiriminin belirli bir polimer-boya sistemiyle uyumsuzluğunu göstermesiyle literatüre özgün bir katkı sağlamaktadır.

Plastic pollution is a global problem that poses a potential risk to human health through microplastics (MPs) that enter the food chain. The accurate detection of MPs from complex matrices such as foods constitutes a significant scientific challenge due to the lack of standard and effective isolation methods. This thesis study aimed to develop an effective and rapid chemical digestion protocol for the isolation and detection of polystyrene (PS) microplastics in foods via optical microscopy under UV light. Within the scope of the study, the digestion performance of different chemicals (KOH, HCl), concentrations, temperatures, and durations was systematically investigated on normal (NM) and high-fat (YM) food models. The effects of the developed protocols on the morphology of the target PS particles and on the Nile Red (NR) staining performance were also evaluated. The results have shown that the 24-48 hour digestion times commonly used in the literature could be reduced to 2-4 hours by using KOH at a high temperature of 85°C, while achieving a digestion efficiency of over 95%. It was confirmed that these accelerated conditions did not cause any microscopically detectable damage to the PS particles in the 40-560 µm size range. One of the critical findings of the study was the determination that acid (HCl) digestion is unsuitable for this analytical workflow, as it alters the surface chemistry of the PS particles, thereby impairing their interaction with the NR dye. In the validation studies conducted with the optimized KOH-based protocols, recovery rates of approximately 97% for the normal model and approximately 108% for the high-fat model were obtained. This thesis presents a pre-treatment method for the analysis of PS microplastics from food matrices that is both rapid and has a high recovery efficiency. The study provides an original contribution to the literature by demonstrating the effectiveness of high-temperature digestion and showing the incompatibility of acid digestion with a specific polymer-dye system.
