Özet

Glioblastoma multiforme (GBM), yetişkinlerde en sık görülen ve en agresif birincil beyin tümörlerinden biridir. Standart tedavi protokolü cerrahi rezeksiyon, radyoterapi ve temozolomid kemoterapisini içermektedir; ancak tümörün yaygın infiltratif yapısı, kan-beyin bariyeri ve tedaviye dirençli hücre popülasyonları nedeniyle sağkalım oranları halen düşüktür ve hastalık yüksek oranda nüks etmektedir. Bu nedenle, GBM tedavisinde yeni ve etkili moleküler terapötik ajanların geliştirilmesi kritik önem taşımaktadır. Son yıllarda ftalosiyanin türevleri, fotodinamik tedavi ajanları olarak yaygın şekilde incelenmekle birlikte, bu moleküllerin ışık aktivasyonu olmadan da antikanser etkiler gösterebileceği hipotezi giderek artan ilgi görmektedir. Ftalosiyaninlerin yapısal özellikleri, suda çözünürlükleri ve biyolojik aktiviteleri, onları GBM gibi tedaviye dirençli tümörlerde potansiyel kemoterapötik ajanlar haline getirmektedir. Bu tez çalışmasında, U87 glioblastoma hücre hattı kullanılarak immünsüprese nude farelerde ortotopik ksenogreft modeli oluşturulmuştur. Modelde, aday silisyum ftalosiyanin bileşikleri sistemik yolla uygulanmış ve tümör progresyonu kapsamlı şekilde değerlendirilmiştir. Sonuçlar, ftalosiyanin türevlerinin tümör büyümesini belirgin şekilde azalttığını göstermiştir. Bu tez çalışması, ftalosiyanin türevlerinin glioblastoma tedavisinde etkili ve umut vadeden bir kemoterapötik ajan olduğunu ortaya koymaktadır. Preklinik modellerde elde edilen bulgular, bu bileşiklerin klinik uygulamalara taşınması için önemli bir dayanak sağlamaktadır. Gelecek araştırmalar, ftalosiyaninlerin etki mekanizmalarının derinlemesine incelenmesiyle, GBM tedavisinde daha etkili ve yenilikçi stratejilerin geliştirilmesine katkıda bulunacaktır. Glioblastoma multiforme (GBM) is one of the most common and aggressive primary brain tumors in adults. The standard treatment protocol includes surgical resection, radiotherapy, and temozolomide chemotherapy; however, survival rates remain low due to the tumor's diffuse infiltrative nature, the blood-brain barrier, and therapy-resistant cell populations, and the disease has a high recurrence rate. Therefore, the development of new and effective molecular therapeutic agents is of critical importance in GBM treatment. In recent years, phthalocyanine derivatives have been extensively studied as photodynamic therapy agents; however, the hypothesis that these molecules may also exhibit anticancer effects without light activation has been gaining increasing interest. The structural properties, water solubility, and biological activities of phthalocyanines make them potential chemotherapeutic agents for treatment-resistant tumors such as GBM. In this thesis study, an orthotopic xenograft model was established using the U87 glioblastoma cell line in immunosuppressed nude mice. Candidate silicon phthalocyanine compounds were systemically administered in the model, and tumor progression was comprehensively evaluated. The results demonstrated that phthalocyanine derivatives significantly reduced tumor growth. This thesis demonstrates that phthalocyanine derivatives are effective and promising chemotherapeutic agents for glioblastoma treatment. The findings obtained from preclinical models provide an important foundation for the clinical translation of these compounds. Future studies focusing on elucidating the mechanisms of action of phthalocyanines are expected to contribute to the development of more effective and innovative strategies in GBM therapy.
