Özet
Bu tez çalışmasında, fruktozun indüklediği hücresel stres yanıtlarının ve kuersetinin bu süreçler üzerindeki düzenleyici etkilerinin HepG2 hücre hattında otofaji, endoplazmik retikulum (ER) stresi ve apoptozis düzeyinde değerlendirilmesi amaçlanmıştır. Kontrol, fruktoz, kuersetin ve fruktoz+kuersetin olmak üzere dört deney grubu oluşturularak, IC₅₀ konsantrasyonlarında 24 saatlik uygulamalar gerçekleştirilmiştir. Oksidatif stres parametreleri incelendiğinde, fruktoz grubunda TOS (42,85 ± 1,90 µmol H₂O₂ equiv./L) ve OSI (3564 ± 403) düzeyleri anlamlı şekilde artarken, TAS düzeyi düşmüştür. Kuersetin uygulaması TAS'ı artırıp OSI'yi düşürerek oksidatif dengeyi kısmen yeniden sağlamıştır. Otofaji oranları floresan mikroskopisiyle değerlendirilmiş; fruktoz grubunda yüksek otofaji (%68,51 ± 5,44) saptanırken, kuersetin grubu (%32,38 ± 4,97) bu yanıtı belirgin şekilde baskılamıştır. RT-qPCR analizlerinde fruktoz grubunda yalnızca CHOP ekspresyonunda artış gözlenmiş; kuersetin grubunda ise GRP78, ATF4, CHOP, Bax ve Bid genlerinde anlamlı ekspresyon artışları saptanmıştır. CHOP düzeyinin kombinasyon grubunda en yüksek seviyeye ulaşması (4,563), persistent ER stresine bağlı olarak apoptotik sürecin şiddetlendiğini göstermektedir. Tüm veriler GraphPad Prism 8.0.1 ile analiz edilmiş, istatistiksel değerlendirmelerde One-Way ANOVA ve Dunnett's çoklu karşılaştırma testleri uygulanmış ve p<0,05 anlamlılık düzeyi olarak kabul edilmiştir. Bu bulgular, fruktozun oksidatif stres, endoplazmik retikulum (ER) stresi ve otofaji aktivasyonunu tetiklerken; kuersetinin bu yanıtları modüle ettiğini ve tümör kökenli bir hücre hattı bağlamında apoptotik sinyallemeyi teşvik ettiğini göstermektedir. Bu sonuçlar, kuersetinin yalnızca bir antioksidan olmadığını, aynı zamanda bağlama bağlı olarak seçici stres yanıtlarını indükleyebilen bir düzenleyici olduğunu ortaya koymakta ve onu metabolik stres koşullarında potansiyel bir terapötik aday haline getirmektedir.

In this thesis, it was aimed to evaluate the cellular stress responses induced by fructose and the regulatory effects of quercetin on these processes in the HepG2 cell line, particularly focusing on autophagy, endoplasmic reticulum (ER) stress, and apoptosis. Four experimental groups—control, fructose, quercetin, and fructose+quercetin—were established, and 24-hour treatments were applied at IC₅₀ concentrations. In the analysis of oxidative stress parameters, the fructose group exhibited significantly increased TOS (42.85 ± 1.90 µmol H₂O₂ equiv./L) and OSI (3564 ± 403) levels, while TAS levels were reduced. Quercetin treatment increased TAS and decreased OSI, partially restoring oxidative balance. Autophagy ratios were assessed via fluorescence microscopy, revealing elevated autophagy in the fructose group (68.51 ± 5.44%), whereas the quercetin group (32.38 ± 4.97%) significantly suppressed this response. RT-qPCR analyses showed that fructose induced an increase only in CHOP expression, whereas quercetin treatment led to significant upregulation of GRP78, ATF4, CHOP, Bax, and Bid genes. The CHOP expression reached its highest level in the combination group (4.563), indicating that persistent ER stress intensified the apoptotic response. All data were analyzed using GraphPad Prism 8.0.1. Statistical evaluations were performed with one-way ANOVA and Dunnett's multiple comparison tests, with p<0.05 considered statistically significant. Overall, the findings suggest that while fructose triggers oxidative stress, ER stress, and autophagy activation, quercetin modulates these responses and, in the context of a tumor-derived cell line, promotes apoptotic signaling. These results highlight quercetin not only as an antioxidant but also as a context-dependent modulator capable of inducing selective stress responses, making it a promising therapeutic candidate under metabolic stress conditions.
