Özet
Kuantum biyoloji, iyon taşınımı ve hücresel sinyal iletimi gibi bazı biyolojik süreçlerin tünelleme ve koherens gibi kuantum fenomenlerine dayanabileceğini öne sürmektedir. Ancak bu fikirlerin deneysel olarak kanıtlanması, canlı sistemlerde kuantum etkilerini izole etmenin zorluğu nedeniyle oldukça güçtür. Bu tez, Statik Manyetik Alanların (SMA) hücrelerdeki manyetik hassasiyeti araştırmak amacıyla kontrollü ve invaziv olmayan araçlar olarak kullanılması gibi kuantum-biyolojik etkileri araştırmak için olası bir yaklaşım sunmaktadır. Çalışmanın ilk bölümünde, düşük ve orta şiddetteki SMA'ların (0.5–3 militesla) glioblastoma ve kolorektal karsinom hücrelerinde etkisi incelenmiştir. İyon akışı ölçümleri, konfokal mikroskopi ve RNA dizileme analizleri, hücre tipine özgü dikkat çekici tepkiler ortaya koymuştur. Glioblastoma hücreleri, mitokondriyal membran potansiyelinin korunması, kontrollü potasyum iyon çıkışı ve membranla ilişkili genlerin düzenlenmiş ekspresyonu ile dirençli bir profil sergilemiştir. Buna karşın, kolorektal karsinom hücreleri mitokondriyal depolarizasyon, potasyum iyon alımı ve keratinizasyon, oksidatif fosforilasyon ve epigenetik yeniden yapılandırma yönünde transkripsiyonel değişimlerle karakterize edilen stres fenotipleri göstermiştir. Bu bulgular, SMA'ların hücresel fizyolojiyi farklı biçimlerde modüle edebileceğini ve potansiyel kuantum biyolojik etkileri deneysel olarak araştırmak için kullanılabileceğini desteklemektedir. Tezin ikinci bölümünde odak, potasyum kanallarına kaydırılmış ve bu kanalların klasik modellerle tam olarak açıklanamayan hız ve seçicilikle iyon iletimini kuantum tünelleme ile nasıl gerçekleştirebileceği araştırılmıştır. Moleküler dinamik simülasyonları ve kuantum tünelleme süresi hesaplamalarının birlikte kullanıldığı bu hibrit çalışmada, bir potasyum iyonunun kanalın seçicilik filtresi içinde bir kaç nanosaniye geçirdiği bulunmuştur. Bu süre, biyolojik gözlemlerle uyumlu olup iyon taşınımında kuantum davranışların etkili olabileceği fikrini desteklemektedir. Bu tez kapsamında elde edilen bulgular, SMA yanıtlarının ve kuantum iyon tünelleme dinamiklerinin incelenmesinin hücresel işlevlerin fiziksel temeline dair yeni anlayışlar sunabileceğini göstermektedir.

Quantum biology suggests that certain biological processes, such as ion transport and signaling, may rely on quantum phenomena such as tunneling and coherence. However, proving these ideas experimentally remains difficult due to the complexity of isolating quantum effects in living systems. This thesis explores one possible route: using static magnetic fields (SMFs) as controlled, non-invasive tools to probe magnetic sensitivity in cells. The first part of the study focuses on the effect of low to moderate intensity SMFs (0.5 to 3 millitesla) on two different human cell types: glioblastoma and colorectal carcinoma cells. A combination of ion flux assays, confocal microscopy, and RNA sequencing revealed strikingly cell-type-specific responses. Glioblastoma cells showed resilience, with stable mitochondrial potential, K⁺ efflux, and regulated expression of ion transport and membrane-associated genes. In contrast, colorectal carcinoma cells exhibited stress phenotypes including mitochondrial depolarization, potassium ion influx, and transcriptional shifts toward keratinization, oxidative phosphorylation, and epigenetic remodeling. These findings support the notion that SMFs can differentially modulate cell physiology and may serve as experimental probes for quantum-biological effects. The second chapter shifts focus to potassium channels and examines how they allow such rapid and selective ion movement that classical models still struggle to explain fully. Using a combination of molecular dynamics and quantum tunneling time calculations, we found that a single potassium ion spends about nanoseconds time range in the channel's selectivity filter. This timescale aligns with biological observations and supports the idea that quantum behavior plays a role in ion transport. Together, these findings suggest that studying SMF responses and quantum ion dynamics in parallel can offer new insights into the physical basis of cellular function.
