Purpose: Pigments are bioactive metabolites with different colors obtained from bacteria, fungi, algae, and plants. In this study, the chemical composition and some biological activities of a pigment extract obtained from the bacterium Micrococcus terreus SK34 (GenBank: PX765938) isolated from pine forest soil were investigated. Method: The identification of this isolate was performed according to the sequence analysis of the 16S rRNA gene. Pigment production from this bacterium was carried out in a shaking Erlenmeyer flask culture in liquid medium. Pigments were extracted from the cell biomass with an acetone-methanol mixture, and the composition of the pigment extract was determined by LC-MS. The antioxidant potential of the pigment extract was evaluated in vitro against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals. The antibacterial activity of the pigment was tested against four gram-positive (Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778, Staphylococcus epidermidis, and Listeria monocytogenes) and four gram-negative (Escherichia coli ATCC 43894, Klebsiella pneumonia ATCC 13883, Salmonella typhimurium, and Pseudomonas aeruginosa) bacteria. The cytotoxic properties of the pigment were evaluated on five different cancer cell lines (breast cancer, lung cancer, neuroblastoma, liver cancer, and uterine cancer) and one healthy cell line (human dermal fibroblasts). Antibacterial activity was analyzed using the agar well diffusion method, and cell viability was analyzed using the WST-1 test. Findings: The pigment extract of M. terreus SK34 was determined to consist of carotenoid-type pigments (phytoene, zeaxanthin, beta-cryptoxanthin, beta-carotene, astaxanthin, alpha-carotene, lycopene, and sarcinaxanthin). This extract has been found to possess in vitro antioxidant (moderate against DPPH and ABTS radicals) and antibacterial (against all test bacteria except S. typhimurium) activities, and also exhibit cytotoxic properties against all cancer cell lines without causing toxic effects on healthy cell line. Results: Due to its non-toxicity and strong biological activity, it is thought that the pigment extract of this bacterium may find applications in the nutraceutical, pharmaceutical, and food industries.
