Özet
Günümüzde gıda güvenliğini sağlamak için uygulanan minimal işleme teknikleri, gıda kaynaklı patojen bakterileri öldürücü olmayan (subletal) stres koşullarına maruz bırakmaktadır. Bu subletal stres, bakterilerin enerji harcayarak hasarı onarmasına ve gelecekteki tehditlere karşı daha dirençli olmasına neden olan adaptif yanıtları tetiklemektedir. Bu adaptasyon, gıda zincirinde daha dirençli mikroorganizmaların ortaya çıkması potansiyeli nedeniyle gıda güvenliği açısından kritik öneme sahiptir. Bu araştırma, Afyonkarahisar'daki entegre et işleme tesislerinden alınan 22 adet çiğ et örneğinden izole edilen Gram-pozitif (Listeria monocytogenes, Listeria ivanovii, Staphylococcus aureus, Bacillus cereus) ve Gram-negatif (Salmonella Enteritidis, Salmonella Typhi, Escherichia coli O157, Escherichia coli O157:H7) patojen bakterilerin biyolojik aktivitelerini soğuk stresi (-18 °C ve -80 °C'de 1 ve 2 saat), düşük basınç stresi (150 mbar ve 300 mbar'da 1 ve 2 saat), manyetik alan stresi (100 mT ve 200 mT'de 1 ve 2 saat), ultrasonik stres (30 kHz ve 60 kHz'de 1 ve 2 saat) olmak üzere farklı stres koşulları altında incelemeyi amaçlamıştır. Uygulanan bu stres işlemlerinin ardından bakterilerin enzimatik aktiviteleri (lipaz, üreaz, proteaz, katalaz, nitrat-redüktaz), hemolitik özellikleri, farklı çevresel koşullara (tuz ve pH) uyumları, karbonhidrat fermantasyon yetenekleri, antibiyotik dirençleri ve biyojen amin üretim potansiyelleri (dekarboksilaz aktivitesi) incelenmiştir. İncelenen 8 patojen bakterinin tamamı kontrol ve stres koşullarında lipolitik, üreaz ve proteolitik aktiviteyi sürdürme eğilimi göstermiştir. Bu durum, bu enzimlerin hücresel hayatta kalma ve metabolik esneklikteki kritik rolünü vurgulamaktadır. Lipaz ve üreaz aktivitelerinde bazı stres koşullarına bağlı istatistiksel olarak anlamlı farklılıklar (P <0.05) bulunmuştur. L. monocytogenes ve L. ivanovii gibi Gram-pozitif patojenler %6 NaCl'ye kadar yüksek tolerans gösterirken, ultrasonik stres bu toleransı artırarak bu bakterilerin %10 NaCl'de dahi gelişimini sağlamıştır. Genel olarak Gram-negatif patojenlerin yüksek tuz konsantrasyonlarına (%6 ve %10 NaCl) karşı daha hassas olduğu tespit edilmiştir. Tüm patojenler nötr pH'ta (pH 7) gelişim gösterirken, asidik (pH 4) ve alkali (pH 10) koşullara karşı değişken hassasiyet sergilemiştir. Özellikle E. coli O157 ve E. coli O157:H7 gibi Gram-negatif suşlar, soğuk ve ultrasonik stres sonrasında pH 4'te üreme yeteneğini artırmıştır. Tüm bakterilerde katalaz aktivitesinde önemli fenotipik değişiklikler gözlemlenmiş, bu da çevresel strese yanıt olarak oksidatif stresten korunma mekanizmalarının aktive olduğunu düşündürmektedir (P <0.05). Bazı stres koşullarında, özellikle ultrasonik ve manyetik alanda, nitrat-redüktaz aktivitesinin azaldığı veya kaybolduğu, bunun da azot metabolizması üzerindeki baskıyı yansıttığı tespit edilmiştir (P <0.05). Patojenlerin kan hücrelerini parçalama yeteneği (hemoliz), stres koşulları altında önemli fenotipik değişiklikler sergilemiştir. Kontrol grubunda beta-hemolitik olan L. monocytogenes, soğuk, düşük basınç ve manyetik alan stresinden sonra alfa-hemoliz yapmaya başlamıştır. Aynı şekilde beta-hemolitik olan E. coli O157, düşük basınç stresinden sonra alfa-hemoliz yapmaya başlamıştır. Beta-hemolitik olan B. cereus, ultrasonik stres ile alfa-hemolize dönüşmüş; düşük basınç ve 200 mT manyetik alana maruziyet sonrasında ise hemolitik özelliğini kaybetmiştir (Gama-hemoliz). Uygulanan stres işlemlerinin, bakterilerin test edilen 6 farklı antibiyotiğe karşı duyarlılığını azaltma eğilimi gösterdiği (direncin arttığı) belirlenmiştir. Bu, düşük basınç ve soğuk stresi altında en belirgin şekilde gözlemlenmiştir. Tüm stres koşulları ve antibiyotik türleri arasındaki etkileşim istatistiksel olarak anlamlı bulunmuştur (P <0.001). Stres koşulları, bakterilerin karbonhidrat fermantasyon yeteneklerinde belirgin değişikliklere yol açmıştır. Bazı stres uygulamaları (özellikle düşük basınç ve soğuk), belirli şekerlerin (sorbitol, glikoz) fermantasyon miktarını önemli ölçüde artırmıştır (P <0.001). L. monocytogenes, L. ivanovii, S. Enteritidis, S. Typhi ve B. cereus suşlarının bazılarının, stres altında histidin veya lizinden biyojen aminler (histamin ve kadaverin) ürettiği tespit edilmiştir. Bu aktivitenin çoğunlukla manyetik alan stresine maruz kalan bakterilerde gözlenmesi, stresin biyojen amin üretimini tetiklediğini düşündürmektedir (P <0.05). Bu çalışma, gıda işleme sırasında uygulanan fiziksel stres faktörlerinin (soğuk, düşük basınç, manyetik alan, ultrasonik ses), gıda kaynaklı patojen bakterilerin metabolik ve fizyolojik fenotiplerinde önemli değişikliklere yol açtığını göstermektedir. Elde edilen bulgular, bakterilerin stres toleransı ve çapraz koruma mekanizmalarını anlamak, daha güvenli ve etkili gıda koruma stratejileri geliştirmek için temel bir çerçeve sunmaktadır.

Minimal processing techniques currently used to ensure food safety expose foodborne pathogenic bacteria to sublethal stress conditions. This sublethal stress triggers adaptive responses, causing bacteria to expend energy repairing damage and becoming more resistant to future threats. This adaptation is critically important for food safety due to the potential for the emergence of more resistant microorganisms in the food chain. This study aimed to investigate the biological activities of Gram-positive (Listeria monocytogenes, Listeria ivanovii, Staphylococcus aureus, Bacillus cereus) and Gram-negative (Salmonella Enteritidis, Salmonella Typhi, Escherichia coli O157, Escherichia coli O157:H7) pathogenic bacteria isolated from 22 raw meat samples obtained from integrated meat processing facilities in Afyonkarahisar under different stress conditions, including cold stress (-18°C and -80°C for 1 and 2 hours), low pressure stress (150 mbar and 300 mbar for 1 and 2 hours), magnetic field stress (100 mT and 200 mT for 1 and 2 hours), and ultrasonic stress (30 kHz and 60 kHz for 1 and 2 hours). Following these applied stress treatments, the enzymatic activities of the bacteria (lipase, urease, protease, catalase, and nitrate reductase), hemolytic properties, adaptation to different environmental conditions (salt and pH), carbohydrate fermentation abilities, antibiotic resistances, and biogenic amine production potentials (decarboxylase activity) were examined. All 8 pathogenic bacteria examined showed a tendency to maintain lipolytic, urease, and proteolytic activity under control and stress conditions. This situation highlights the critical role of these enzymes in cellular survival and metabolic flexibility. Statistically significant differences (P < 0.05) were found in lipase and urease activities under certain stress conditions. Gram-positive pathogens such as L. monocytogenes and L. ivanovii exhibit high tolerance to up to 6% NaCl, while ultrasonic stress increases this tolerance, allowing these bacteria to grow even in 10% NaCl. It has generally been found that Gram-negative pathogens are more sensitive to high salt concentrations (6% and 10% NaCl). While all pathogens grow at a neutral pH (pH 7), they exhibit varying sensitivity to acidic (pH 4) and alkaline (pH 10) conditions. Specifically, Gram-negative strains such as E. coli O157 and E. coli O157:H7 have shown increased ability to grow at pH 4 after cold and ultrasonic stress. Significant phenotypic changes in catalase activity were observed in all bacteria, suggesting that mechanisms for protecting against oxidative stress are activated in response to environmental stress (P < 0.05). It has been found that under certain stress conditions, especially in ultrasonic and magnetic fields, nitrate reductase activity decreases or is lost, which reflects the pressure on nitrogen metabolism (P < 0.05). The ability of pathogens to break down blood cells (hemolysis) has shown significant phenotypic changes under stress conditions. L. monocytogenes, which was beta-hemolytic in the control group, began to exhibit alpha-hemolysis after cold, low pressure, and magnetic field stress. Similarly, E. coli O157, which is beta-hemolytic, began to exhibit alpha-hemolysis after low-pressure stress. B. cereus, which is beta-hemolytic, was converted to alpha-hemolytic by ultrasonic sound stress; after exposure to low pressure and a 200 mT magnetic field, it lost its hemolytic property (gamma-hemolysis). It has been determined that the applied stress treatments tend to decrease the sensitivity of the bacteria to the 6 different antibiotics tested (increasing resistance). This was most prominently observed under low pressure and cold stress. The interaction between all stress conditions and antibiotic types was found to be statistically significant (P < 0.001). Stress conditions have led to significant changes in the carbohydrate fermentation abilities of bacteria. Some stress applications (especially low pressure and cold) significantly increased the amount of fermentation of certain sugars (sorbitol, glucose) (P < 0.001). It has been found that some strains of L. monocytogenes, L. ivanovii, S. Enteritidis, S. Typhi and B. cereus produce biogenic amines (histamine and cadaverine) from histidine or lysine under stress. The observation of this activity mostly in bacteria exposed to magnetic field stress suggests that the stress triggers the production of biogenic amines (P < 0.05). This study demonstrates that physical stress factors applied during food processing (cold, low pressure, magnetic field, ultrasonic sound) lead to significant changes in the metabolic and physiological phenotypes of foodborne pathogenic bacteria. The findings provide a fundamental framework for understanding bacterial stress tolerance and cross-protection mechanisms and for developing safer and more effective food preservation strategies.
