Özet

Bu tez çalışmasının temel amacı, son yıllarda nanoteknolojinin biyoteknoloji alanındaki yenilikçi uygulamalarından biri olarak öne çıkan gen transferine yönelik, biyouyumlu ve biyoparçalanabilir özellikte nanopartiküler taşıyıcı sistemlerin sentezlenmesi ve karakterize edilmesidir. Çalışma kapsamında, bu sistemlerin yapısal özellikleri detaylı olarak aydınlatılacak ve gen aktarımındaki transfeksiyon verimlilikleri deneysel olarak değerlendirilecektir. Gen terapisi, kalıtsal ya da sonradan edinilmiş hastalıkların tedavisinde, terapötik genlerin vektör adı verilen taşıyıcı sistemler aracılığıyla hedef hücrelere aktarılmasına ve bu genlerin hücre içinde ekspresyonunun sağlanmasına dayanmaktadır. Ancak gen transfer süreci, vektörlerin hücre içi hedeflerine ulaşmasında karşılaşılan çeşitli hücresel bariyerler nedeniyle karmaşık bir hal alır. Bu süreçte kullanılan vektörlerin, minimal toksisite ile maksimum transfeksiyon etkinliği göstermesi büyük önem taşımaktadır. Bu doğrultuda, gen transferinde vektör seçimi kritik bir rol oynamaktadır. Gen aktarımı için temel olarak iki yaklaşım geliştirilmiştir: yüksek transfeksiyon etkinliğine sahip viral vektörler ve viral vektörlerin potansiyel immünojenik ve toksik etkileri nedeniyle tercih edilen viral olmayan taşıyıcı sistemler. Bu tez çalışması kapsamında, gen aktarımı için model olarak plazmid DNA (e-GFP) yüklenmiş, lipid-polimer esaslı hibrit non-viral nanopartiküler sistemlerin geliştirilmesi ve karakterizasyonu gerçekleştirilmiştir. Lipid-polimer hibrit nanopartiküller (LPHNP'ler), liposomlar ile polimerik nanopartiküllerin yapısal ve fonksiyonel avantajlarını bir araya getiren, çekirdek-kabuk mimarisine sahip yenilikçi ilaç taşıyıcı sistemlerdir. Bu hibrit yapılar, iç kısımda kontrollü salım sağlayan polimerik bir çekirdek ile dış yüzeyde biyomembran benzeri geçirgenlik özellikleri sunan lipid bir tabakadan oluşmaktadır. LPHNP'lerin geliştirilme amacı, liposomların sınırlı stabilite, içerik sızıntısı ve kısa dolaşım süresi gibi dezavantajları ile polimerik nanopartiküllerin hücre membranı geçişindeki yetersizliklerini ortadan kaldırmaktır. Bu nedenle, bu tez çalışmasında gen taşıma amacıyla kullanılabilecek yeni bir LPHNP formülasyonu geliştirilmiş; plazmid DNA ile kompleks oluşturularak bu sistemlerin transfeksiyon etkinliği değerlendirilmiştir. Formülasyonun hazırlanmasında, PEG-PCL (polietilen glikol–polikaprolakton) ve DOTAP içeren bileşenler kullanılmış ve modifiye su/yağ/su emülsiyon-temelli çözücü buharlaştırma yöntemi uygulanmıştır. Elde edilen nanopartiküller, yeşil floresan protein ekspresyon plazmidini taşıyacak şekilde yüklenmiş ve ardından sistemlerin karakterizasyonu gerçekleştirilmiştir. Bu kapsamda, partikül boyutu, boyut dağılımı (polidispersite indeksi), zeta potansiyeli ve enkapsülasyon etkinliği analiz edilmiştir. Devamında, optimize edilen formülasyonlarla in vitro ilaç salım profilleri belirlenmiş ve uygun fizikokimyasal özellikler gösteren sistemler kullanılarak HEK-293 hücre hattı üzerinde transfeksiyon etkinliği değerlendirilmiştir. Bu çalışmada geliştirilen nanopartiküllerin fizikokimyasal özellikleri değerlendirildiğinde, partikül boyutunun 200 nm'nin altında, polidispersite indeksinin (PDI) 0,3'ün altında ve zeta potansiyel değerlerinin ±10–30 mV aralığında olduğu tespit edilmiştir. Bu sonuçlar, hazırlanan nanopartiküllerin gen taşıma uygulamaları için uygun bir boyutsal ve yüzey yük dağılımına sahip olduğunu göstermektedir. Ayrıca, plazmid DNA'nın nanopartikül yapısı içerisinde yüksek verimle enkapsüle edildiği belirlenmiştir. Sonuç olarak, bu tez çalışmasında PEG-PCL ve DOTAP bileşenlerinden oluşan lipid-polimer hibrit nanopartiküllerin, HEK-293 hücre hattında transfeksiyon etkinliği gösteren, toksik olmayan ve viral olmayan gen taşıyıcı sistemler olarak etkili bir performans sergilediği belirlenmiştir. Yürütülen in vitro karakterizasyon analizleri ve hücre kültürü temelli biyolojik değerlendirmeler, geliştirilen sistemin hedeflenen kriterleri başarıyla karşıladığını ortaya koymuştur. Elde edilen veriler, bu hibrit nanopartikül sisteminin gen terapisine yönelik mevcut nanotaşıyıcı alternatifleri arasında potansiyel bir aday olduğunu ve gelecekteki terapötik uygulamalar için umut vadeden bir platform sunduğunu göstermektedir.

The main objective of this thesis is to synthesise and characterise biocompatible and biodegradable nanoparticulate carrier systems for gene transfer, which has been one of the innovative applications of nanotechnology in biotechnology in recent years. Within the scope of the study, the structural properties of these systems will be elucidated in detail and their transfection efficiency in gene transfer will be experimentally evaluated. Gene therapy, in the treatment of inherited or acquired diseases, is based on the transfer of therapeutic genes to target cells via DNA carrier systems called vectors and the expression of these genes within the cell. However, the gene transfer process is complicated by various cellular barriers encountered in the delivery of vectors to their intracellular targets. It is of great importance that the vectors used in this process show maximum transfection efficiency with minimal toxicity. Accordingly, vector selection plays a critical role in gene transfer. Basically, two approaches have been developed for gene transfer: viral vectors with high transfection efficiency and non-viral carrier systems, which are preferred due to the potential immunogenic and toxic effects of viral vectors. In this thesis, lipid-polymer based hybrid non-viral nanoparticular systems loaded with plasmid DNA (eGFP) as a model for gene transfer were developed and characterised. Lipid-polymer hybrid nanoparticles (LPHNPs) are innovative drug delivery systems with core-shell architecture that combine the structural and functional advantages of liposomes and polymeric nanoparticles. These hybrid structures consist of a polymeric core providing controlled release on the inside and a lipid layer offering biomembrane-like permeability properties on the outer surface. The aim of the development of LPHNPs is to overcome the disadvantages of liposomes such as limited stability, content leakage and short circulation time and the inadequacies of polymeric nanoparticles in cell membrane permeation. These hybrid systems are considered as an attractive alternative in drug delivery systems due to their high encapsulation efficiency and well-defined release profiles. While the polymer structure contributes to controlled and prolonged drug release, the lipid structure increases bioavailability by facilitating interaction with biological membranes and supports the orientation of the carrier to the target tissue. Therefore, in this thesis, a new LPHNP formulation that can be used for gene delivery was developed and the transfection efficiency of these systems was evaluated by complexing with plasmid DNA. In the preparation of the formulation, components containing PEG-PCL (polyethylene glycol-polycaprolactone) and DOTAP were used and modified water/oil/water emulsion-based solvent evaporation method was applied. The obtained nanoparticles were loaded to carry the green fluorescent protein expression plasmid and then the systems were characterised. In this context, particle size, size distribution (polydispersity index), zeta potential and encapsulation efficiency were analysed. Subsequently, in vitro drug release profiles were determined with the optimised formulations and transfection efficiency was evaluated on HEK293 cell line using systems with appropriate physicochemical properties. When the physicochemical properties of the NPs developed in this study were evaluated, it was found that the particle size was below 200 nm, the polydispersity index (PDI) was below 0.3 and the zeta potential values were in the range of ±10-30 mV. These results show that the prepared nanoparticles have a suitable dimensional and surface charge distribution for gene delivery applications. In addition, it was determined that plasmid DNA was encapsulated in the nanoparticle structure with high efficiency. In conclusion, in this thesis, lipid-polymer hybrid nanoparticles consisting of PEG-PCL and DOTAP components were determined to perform effectively as non-toxic and non-viral gene carrier systems with high transfection efficiency in HEK293 cell line. In vitro characterisation analyses and cell culture-based biological evaluations revealed that the developed system successfully met the targeted criteria. The data obtained show that this hybrid nanoparticle system is a potential candidate among the existing nanocarrier alternatives for gene therapy and offers a promising platform for future therapeutic applications.
