Özet
Günümüzde tüketicilerin fonksiyonel ve sağlıklı beslenmeye olan ilgisinin artmasıyla birlikte, biyoaktif bileşenler açısından zengin fonksiyonel gıdalara olan talep hızla artmaktadır. Domates (Solanum lycopersicum) içeriğinde birçok faydalı biyoaktif madde içermesi sebebiyle oldukça önemlidir.Taze domates ve endüstriyel işlemeden elde edilen yan ürünleri de yüksek likopen, fenolik asit ve flavonoid içeriği nedeniyle son derece önemli bir besindir. Domates tazeliğini uzun süre koruyamamaktadır. Bu sebeple raf ömrünü uzetmak için farklı proseslerden geçerek domatesuy, püresi, salça, domates kurusu gibi yan ürünleri elde edilir.Epidemiyolojik çalışmalar, domates tüketiminin içerdiği biyoaktif bileşikler sayesinde kardiyovasküler hastalıklar, bazı kanser türleri ve oksidatif strese bağlı kronik hastalık riskini azalttığını göstermektedir. Bununla birlikte, bu biyoaktif bileşiklerin sağlık üzerindeki faydalı etkilerinin gözlemlenmesi, yalnızca gıdadaki miktarlarına değil, aynı zamanda sindirim sistemi boyunca yapılarını koruyabilme ve bağırsaklardan emilebilir hale gelme yeteneklerine, yani biyoyararlanımlarına ve biyoerişilebilirliklerine de bağlıdır. Sindirim sistemi boyunca yaşanan pH değişiklikleri, enzim aktiviteleri ve gıda matrisinin yapısı, fenolik bileşiklerin stabilitesini olumsuz etkileyebilir ve biyoerişebilirliğini azaltabilir. Bu bağlamda, çeşitli uygulamalar yoluyla gıdaların biyoerişebilirliğini olumlu yönde etkilemek için çalışmalar yapılmaktadır. Son yıllarda, antioksidan ve anti-inflamatuar özellikleriyle dikkat çeken ve çeşitli gıda uygulamadalrında kullanılan molekğler hidrojen oldukça önem kazanmıştır. Bununla birlikte moleküler hidrojen uygulamalarından biri olan Hidrojenle Zenginleştirlmiş Su (HZS), çeşitli alanlarda olarak önem kazanmıştır. Moleküler hidrojenin, biyolojik sistemlerde seçici bir antioksidan görevi gördüğü ve zararlı reaktif oksijen türlerini nötralize ettiği bilinmektedir. HZS üretimi için elektroliz, magnezyum kulanılarak ve doğrudan hidrojen gazı enjeksiyonu gibi farklı yöntemler kullanılmaktadır. Bu tezin temel amacı, saf su, HZS ve Magnezyumlu su (MS) ile tüketilen farklı fiziksel formlardaki domates ürünlerinin (domates suyu, domates püresi ve kurutulmuş domates) fenolik bileşik içeriğinin, antioksidan kapasitesinin ve biyoerişebilirliğinin in vitro ortak sindirim simülasyonu sırasında nasıl değiştiğini belirlemektir. Bu çalışma, farklı su türlerinin gıda matrisiyle etkileşimini ve sindirim sırasındaki koruyucu rollerini aydınlatarak literatüre benzersiz bir katkı sağlamaktadır. Bu çalışmada, malzeme olarak üç farklı endüstriyel olarak üretilmiş domates ürünü kullanılmıştır: domates suyu (sıvı matris), domates püresi (yarı katı matris) ve kurutulmuş domates (katı matris). Bu üç domates ürünü, başlangıç örneği olarak metil ile ekstrakte edildi. HZS üretimi için iki yöntem kullanıldı: elektroliz kullanan mekanik bir detoks hidrojenli su üretim makinesi ve magnezyum bazlı bir üretim yöntemi. Saf su, HZS ve MS domates örnekleri, in vitro sindirim modeline göre laboratuvar koşullarında işlendi. Hazırlanan örneklerin toplam fenolik içeriği, toplam flavonoid içeriği, toplam antioksidan kapasitesi ve in vitro biyoerişebilirliği analiz edildi. Toplam fenolik içeriği Folin-Ciocalteu yöntemiyle ölçülürken, antioksidan kapasitesi CUPRAC ve DPPH yöntemleriyle değerlendirildi. Analizler, ekstraksiyondan sonra ve sindirim sürecinin mide ve bağırsak aşamalarında gerçekleştirildi. Tüm analizler, in vitro sindirim modeline göre laboratuvar koşullarında yapıldı. Benzer şekilde, örneklerdeki fenolik profil belirlendi ve Yüksek Performanslı Sıvı Kromatografisi (HPLC) kullanılarak hesaplandı. Daha sonra sonuçlar doğrultusunda biyoerişebilirliği hesaplandı. Bu çalışma, farklı su tipleriyle (saf su, HZS, MS) in vitro ortak sindirimin domates suyu, püresi ve kurutulmuş domates örneklerinin Toplam fenolik miktarı içeriği ve biyoerişebilirliği üzerindeki etkilerini araştırmıştır. Bulgulara göre, matris yapısı sonuçlarda belirleyici bir faktördü; sıvı matrisli domates suyunda, HZS kullanımı bağırsak fazında en yüksek toplam fenolik miktarı değerini koruyarak %113,51 ile en yüksek biyoerişebilirliği sağlamıştır. Domates püresinde sular arasında biyoerişebilirliğinde istatistiksel olarak anlamlı bir fark gözlenmezken, katı matrisli kurutulmuş domates örneklerinde, HZS ve MS uygulamaları saf suya (%86,65) kıyasla biyoerişebilirliği %110'a kadar önemli ölçüde artırmıştır (p<0,05). Bu sonuçlar, fonksiyonel suların, özellikle sıvı ve kurutulmuş ürünlerde, sindirim koşulları altında bağlı fenolikleri serbest bırakarak biyoerişebilirliği olumlu yönde etkilediğini göstermektedir. Toplam analizlerinde, su türünün etkisi ürünün fiziksel formuna bağlı olarak değişmiştir. Domates suyu ve püresinde HZS ile birlikte sindirim, bağırsak fazında en yüksek flavonoid geri kazanımını ve biyoerişebilirliğini sağlamıştır. Özellikle domates püresinde, HZS (%65,40) ve MS (%54,33) uygulamaları, flavonoidleri oksidasyondan koruyarak saf suya (%47,31) kıyasla biyoerişebilirliği önemli ölçüde artırmıştır (p<0,05). Kurutulmuş domates örneklerinde, su türleri arasında anlamlı bir fark gözlenmezken, muhtemelen yüksek kabuk içeriği nedeniyle tüm gruplarda en yüksek biyoerişebilirlik oranları (%95-101) kaydedilmiştir. Genel olarak, HZS ve MS'nin flavonoidlerin çözünürlüğünü artırarak ve yapısal oksidasyonu önleyerek sıvı ve yarı katı matrislerde biyoerişebilirliği desteklediği sonucuna varılmıştır. CUPRAC yöntemiyle belirlenen antioksidan kapasite sonuçları incelendiğinde, su türlerinin etkisi gıda matrisinin fiziksel yapısına göre değişmiştir. Sıvı matrisli domates suyunda su türleri arasında istatistiksel olarak anlamlı bir fark gözlenmezken (%63-69 biyoerişebilirlik aralığı), fonksiyonel sular yarı katı (püre) ve katı (kurutulmuş domates) matrislerde önemli bir üstünlük göstermiştir. Özellikle, kurutulmuş domates örneklerinde, MS uygulaması %105,93'lük bir biyoerişebilirlik değeri elde etmiş, bağırsak fazında en yüksek antioksidan aktiviteyi korumuş ve saf suya (%69,67) kıyasla istatistiksel olarak anlamlı bir fark yaratmıştır (p<0,05). Benzer şekilde, domates püresinde, HZS ve MS, saf suya kıyasla daha yüksek biyoerişebilirlik sağlamıştır. Bu veriler, magnezyumca zenginleştirilmiş suyun, katı gıda matrislerinde antioksidan bileşenlerin salınımını ve stabilitesini artırmada elektrolizle indüklenen HZS ve saf sudan daha etkili bir strateji olduğunu ortaya koymaktadır. DPPH radikal temizleme aktivitesi sonuçları, matris içindeki moleküler hidrojenin difüzyon yeteneğinin önemini vurgulamaktadır. Minimum difüzyon bariyerlerine sahip domates suyu örneklerinde, HZS uygulaması ince bağırsak fazında biyoerişebilirliği önemli ölçüde artırarak olağanüstü bir %203,75'e ulaşmıştır. Bu, sıvı ortamda HZS'deki hidrojen moleküllerinin serbest radikal temizleme aktivitesinin maksimize edilmesiyle açıklanmaktadır. Buna karşılık, daha yoğun bir matrise sahip domates püresi ve kurutulmuş domates örneklerinde sindirim sırasında genel bir kapasite kaybı yaşanmasına rağmen, MS her iki üründe de saf suya kıyasla daha yüksek biyoerişebilirlik ( %33-35 aralığında) sunmuştur. Sonuç olarak, DPPH bulguları, HZS'nin sıvı ürünlerde benzersiz bir antioksidan potansiyeli sunduğunu, MS'nin ise posalı ve kurutulmuş ürünlerde antioksidan kapasitesini korumada daha istikrarlı bir performans sergilediğini göstermektedir. Çalışmanın çarpıcı bulgularından biri, saf su sindirimde tamamen parçalanan epikateşinin, HZS ve MS ortamlarında biyoerişebilirliğini korumasıdır. Dahası, kurutulmuş domates örneklerinde, MS işlemi, saf suya kıyasla klorojenik asit (%96,71) ve kateşinin (%296,48) biyoerişebilirliğinde önemli artışlara neden olmuştur. Bağırsak fazında başlangıçta tespit edilemeyen klorojenik asidin ortaya çıkması, fonksiyonel suların sindirim enzimatik aktivitesi yoluyla bağlı fenolik maddeleri serbest bırakma potansiyelini göstermektedir; HRW'nin düşük redoks potansiyeli ise oksidasyona duyarlı bileşenlerin kimyasal stabilitesinin korunmasını sağlar. Bu tez çalışmasından elde edilen veriler, domates ürünlerinin HZS ve MS ile birlikte sindirilmesi stratejisinin, fenolik bileşenlerin biyoerişebilirliğini artırmak için etkili bir yöntem olduğunu ortaya koymaktadır. HZS örneklerinde, moleküler hidrojenin, sindirim sistemindeki pH değişiklikleri ve enzimatik reaksiyonlar sırasında ortaya çıkan serbest radikalleri temizleyerek oksidatif stresi azalttığı düşünülmektedir. Bu şekilde, oksidasyona duyarlı fenolik bileşikler, parçalanmadan emilim noktası olan bağırsaklara ulaşabilir. Bu etki, difüzyonun hızlı olduğu sıvı matrislerde (domates suyu) daha belirgindir. MS örneklerinde, magnezyum iyonları, özellikle kateşinler olmak üzere flavonoidlerin yapısındaki hidroksil gruplarıyla kompleksler oluşturarak kimyasal stabilite sağlayabilir. Ek olarak, magnezyum, mikro düzeyde bağırsak ortamındaki pH'ı tamponlayarak asidik veya alkali bozunmayı önleyebilir. Bu etki, kompleks matrisli kurutulmuş domateslerde daha net gözlemlenmiştir. Çalışma sonuçları, fonksiyonel suların sadece içecek olarak değil, aynı zamanda besin emilimini artıran bir gıda matrisi modülatörü olarak da kullanılabileceğini göstermektedir. Bu suların farklı polifenol kaynaklarıyla etkileşimini araştırmak için gelecekteki çalışmalar önerilmektedir. Ayrıca, daha büyük ölçekli endüstriyel çalışmalar yapılabileceği de önerilmektedir. Ek olarak, tüketici etkisini gözlemlemek için duyusal analizler yapılabilir. Sonuç olarak, domates ürünlerinin yüksek yağlı veya orta yağlı gıdalarla birlikte tüketimi, diyetteki doğal antioksidanların faydalarını en üst düzeye çıkarmak için basit, uygulanabilir ve etkili bir beslenme stratejisi olarak değerlendirilmelidir.

With the increasing interest of consumers in functional and healthy nutrition today, the demand for functional foods rich in bioactive components is rapidly increasing. Tomatoes (Solanum lycopersicum) are particularly important because they contain many beneficial bioactive substances. Fresh tomatoes and their by-products from industrial processing are also extremely important foods due to their high lycopene, phenolic acid, and flavonoid content. Tomatoes cannot maintain their freshness for a long time. Therefore, to extend their shelf life, they undergo different processes to obtain by-products such as tomato juice, puree, tomato paste, and dried tomatoes. Epidemiological studies show that tomato consumption reduces the risk of cardiovascular diseases, some types of cancer, and chronic diseases related to oxidative stress, thanks to the bioactive compounds it contains. However, observing the beneficial effects of these bioactive compounds on health depends not only on their amounts in the food but also on their ability to maintain their structure throughout the digestive system and become absorbable from the intestines, i.e., their bioavailability and bioaccessibility. pH changes, enzyme activities, and the structure of the food matrix throughout the digestive system can negatively affect the stability of phenolic compounds and reduce their bioaccessibility. In this context, studies are being conducted to positively influence the bioaccessibility of foods through various applications. In recent years, molecular hydrogen, which attracts attention with its antioxidant and anti-inflammatory properties and is used in various food applications, has gained considerable importance. Hydrogen-Rich water (HRW), one of the applications of molecular hydrogen, has gained importance in various fields. Molecular hydrogen is known to act as a selective antioxidant in biological systems and neutralize harmful reactive oxygen species (ROS). Different methods, such as electrolysis, using magnesium, and direct hydrogen gas injection, are used for HRW production. The main objective of this thesis is to determine how the phenolic compound content, antioxidant capacity (AC), and bioaccessibility of different physical forms of tomato products (tomato juice, tomato puree, and dried tomatoes) consumed with distilled water, HRW, and Magnesium Water (MW) change during in vitro co-digestion simulation. This study provides a unique contribution to the literature by elucidating the interaction of different water types with the food matrix and their protective roles during digestion. In this study, three different industrially produced tomato products were used as materials: tomato juice (liquid matrix), tomato puree (semi-solid matrix), and dried tomatoes (solid matrix). These three tomato products were extracted with methyl as a starting sample. Two methods were used for HRW production: a mechanical detox hydrogen water production machine using electrolysis and a magnesium-based production method. Distilled water, HRW, and MW tomato samples were processed in laboratory conditions according to an in vitro digestion model. The total phenolic content, total flavonoid content, total antioxidant capacity, and in vitro bioaccessibility of the prepared samples were analyzed. Total phenolic content was measured using the Folin-Ciocalteu method, while antioxidant capacity was evaluated using the CUPRAC and DPPH methods. Analyses were performed after extraction and during the gastric and intestinal stages of the digestion process. All analyses were performed under laboratory conditions according to an in vitro digestion model. Similarly, the phenolic profile in the samples was determined and bioaccessibility was calculated using High-Performance Liquid Chromatography (HPLC). This study investigated the effects of in vitro co-digestion with different water types (distilled water, HRW, MW) on the Total Phenolic Compound (TPC) content and bioaccessibility of tomato juice, puree, and dried tomato samples. According to the findings, matrix structure was a determining factor in the results; in tomato juice with a liquid matrix, the use of HRW maintained the highest TPC value in the intestinal phase, resulting in the highest bioaccessibility at 113.51%. While no statistically significant difference in bioaccessibility was observed between the waters in tomato puree, in dried tomato samples with a solid matrix, HRW and MW applications significantly increased bioaccessibility to 110% compared to distilled water (86.65%) (p<0.05). These results indicate that functional waters positively affect bioaccessibility, especially in liquid and dried products, by releasing bound phenolics under digestion conditions. In Total Flavonoid Compound (TFC) analyses, the effect of water type varied depending on the physical form of the product. Co-digestion with HRW in tomato juice and puree resulted in the highest flavonoid recovery and bioaccessibility in the intestinal phase. Specifically, in tomato puree, HRW (65.40%) and MW (54.33%) applications significantly increased bioaccessibility by protecting flavonoids from oxidation compared to distilled water (47.31%) (p<0.05). In dried tomato samples, while no significant difference was observed between water types, the highest bioaccessibility rates (95-101%) were recorded across all groups, likely due to high peel content. In general, it was concluded that HRW and MW support bioaccessibility in liquid and semi-solid matrices by increasing the solubility of flavonoids and preventing structural oxidation. When the antioxidant capacity results determined by the CUPRAC method were examined, the effect of water types varied according to the physical structure of the food matrix. While no statistically significant difference was observed between water types in tomato juice with a liquid matrix (63-69% bioaccessibility range), functional waters showed a significant superiority in semi-solid (puree) and solid (dried tomato) matrices. In particular, in dried tomato samples, MW application achieved a bioaccessibility value of 105.93%, maintaining the highest antioxidant activity in the gut phase, and created a statistically significant difference compared to distilled water (69.67%) (p<0.05). Similarly, in tomato puree, HRW and MW provided higher bioaccessibility compared to distilled water. These data reveal that magnesium-enriched water is a more effective strategy than electrolysis-induced HRW and distilled water in increasing the release and stability of antioxidant components in solid food matrices. The DPPH radical scavenging activity results highlight the importance of the diffusion ability of molecular hydrogen within the matrix. In tomato juice samples with minimal diffusion barriers, HRW application dramatically increased bioaccessibility in the small intestine phase, reaching an extraordinary 203.75%. This is explained by the maximization of the free radical scavenging activity of hydrogen molecules in HRW in the liquid medium. In contrast, although a general loss of capacity was experienced during digestion in tomato puree and dried tomato samples with a denser matrix, MW offered higher bioaccessibility (in the range of 33-35%) compared to distilled water in both products. In conclusion, the DPPH findings demonstrate that HRW offers a unique antioxidant potential in liquid products, while MW exhibits a more stable performance in preserving antioxidant capacity in pulpy and dried products. One of the striking findings of the study is that epicatechin, which completely degrades in the control group (distilled water) digestion, remains bioavailable in HRW and MW media. Furthermore, in dried tomato samples, MW treatment resulted in dramatic increases in the bioaccessibility of chlorogenic acid (96.71%) and catechin (296.48%) compared to distilled water. The emergence of initially undetectable chlorogenic acid in the intestinal phase demonstrates the potential of functional waters to release bound phenolics through their digestive enzymatic activity; while the low redox potential of HRW ensures the preservation of the chemical stability of oxidation-sensitive components. The data obtained from this thesis study reveal that the strategy of co-digestion of tomato products with Hydrogen-Rich Water (HRW) and Magnesium Water (MW) is an effective method to increase the bioaccessibility of phenolic components. In HRW samples, molecular hydrogen is thought to reduce oxidative stress by scavenging free radicals (ROS) that arise during pH changes and enzymatic reactions in the digestive tract. In this way, oxidation-sensitive phenolic compounds can reach the intestines, the absorption point, without being broken down. This effect is more pronounced in liquid matrices (tomato juice) where diffusion is rapid. In M samples, magnesium ions can provide chemical stability by forming complexes with hydroxyl groups in the structure of flavonoids, especially catechins. In addition, magnesium may buffer the pH in the intestinal environment at the micro level, preventing acidic or alkaline degradation. This effect was observed more clearly in dried tomatoes with complex matrices. The study results show that functional waters can be used not only as a beverage but also as a food matrix modulator that enhances the absorption of nutrients. Future studies are recommended to investigate the interaction of these waters with different polyphenol sources. It is also suggested that larger-scale industrial studies can be conducted. In addition, sensory analyses can be performed to observe the consumer effect. In conclusion, the consumption of tomato products together with HRW or MW should be considered a simple, feasible, and effective nutritional strategy for maximizing the benefits of naturally occurring antioxidants in the diet.
