Özet

6. ÖZET Meme Kanserli Hastalarda p27/Kipl Gen Mutasyonlarının Araştırılması P27/kipl proteini siklin bağlı kinaz inhibitor proteinleri ailesine ait olup, hücre döngüsünün negatif kontrolünde rol almaktadır. Son araştırmalar düşük p27 ekspresyonunun hastalarda kötü sağkalım ve ciddi malignansi ile ilişkili olduğunu düşündürmektedir. Meme kanseri olan 24 hastada PCR-SSCP uygulanarak, bu gende genetik anormallikler tespit edildi. P27 değişimlerinin meme kanseri gelişimi ile ilgisi olduğu belirlendi. P27/kipl geni iki ekzon ve bir introndan oluşmaktadır. Bu nedenle bu bölgeler üç set primer kullanılarak çoğaltıldı. 11 (%45.8) tümör örneğinde farklı bant bulunduğu gözlendi. Bu mutasyonların iki tanesi ekzon 1-5' bölgesinde, sekiz tanesi ekzon 1-3' bölgesinde, bir tanesi de ekzon 2 bölgesinde saptandı. Genin 3' bölgesindeki değişimlerin, 109. kodonun polimorfizmleri olduğu daha önce gösterilmiştir. Bu polimorfîzmler önceden tanımlanmış olup, Bgl I restriksiyon enzim kesim bölgesi içerdikleri bilinmektedir. Kodon 109'daki polimorfizmi kesin olarak doğrular şekilde, farklı banda sahip PZR ürünleri Bgll ile kesilmişlerdir.

6.SUMMARY P27/Kipl Gene Mutation Detection in Breast Cancer Patients The p27/Kipl protein belongs to family of cyclin-dependent kinase inhibitory proteins that are negative regulators of cell cycle progression. Recent observations suggest that low p27 expression may correlate with poor survival in patients with certain malignancies. To determine whether p27 alterations may be involved in breast cancer progression, we have analysed the presence of genetic abnormalities in this gene in 24 patients with breast carcinoma using PCR- SSCP. The coding region of he p27/Kipl gene consist of two exons and an intron. Therefore three sets of primers were used for the amplification of these regions. Aberrantly migrating bands were observed in 11 (%45.8) of the tumor samples. Two of these mutations were in the 5' region of exon 1, 8 were in the 3' region of exon 1 and one mutation was in exon 2. The mutations at the 3' end were polymorphism at codon 109 of the gene. These polymorphism have been described previously and are known to create a restriction site for Bgl I. To confirm the exact nature of this codon 109 polymorphism aberrantly migrating PCR products were also digested with Bgl I.
