Özet

ÖZET Oksidatif stres diyabetes mellitusun patogenezinde önemli bir rol oynar. Diyabetes mellitus gerek periferal ve gerekse santral sinir sisteminin her ikisini de etkileyebilir. Diyabetin santral sinir sisteminde meydana getirdiği zararlı etkiler periferdeki etkilerine göre daha az çalışılmıştır. Bu çalışmanın amacı diyabetes mellitusun uzun dönemde santral sinir sisteminde meydana getirdiği değişiklikleri araştırmaktır. Çalışmada Mus musculus türü fareler kontrol grubu, alloksanla uyarılmış diyabet grubu (DM), alloksanla uyarılmış diyabet + melatonin grubu (DM+MLT) ve alloksanla uyarılmış diyabet + pentoksifilin grubu (DM+PTX) olmak üzere 4 gruba ayrıldı. Çalışma diyabet oluşturulduktan sonra 8 hafta sürdürüldü. Çalışmanın sonunda fareler eter anestezisi altında dekapite edildi ve tüm gruplarda beyin korteks, subkorteks ve serebellum katmanlarına ayrıldı. Her üç tabakada malondialdehid (MDA), nitrit ve nitrat düzeyleri ölçüldü. Kontrol, DM+MLT ve DM+PTX gruplarında bulunan farelerde çalışmanın sonunda ağırlık artışı tespit edilirken (p<0,01) diyabet grubunda yer alan farelerin ağırlık ölçümlerinde değişiklik yoktu (p>0,05). DM, DM+MLT ve DM+PTX gruplarının açlık kan şekeri (AKŞ) düzeyleri çalışmanın sonunda kontrol grubuna göre yüksek olarak ölçüldü (p<0,001). Çalışma sonunda kontrol grubuna ait korteks, subkorteks ve serebellum MDA düzeyleri DM, DM+PTX ve DM+MLT grubu ile karşılaştırıldığında anlamlı bir fark olmadığı gözlendi (p>0,05). DM+MLT ve DM+PTX gruplarının her üç beyin tabakasındaki MDA düzeyleri DM grubu ile karşılaştırıldığında anlamlı olarak düşük bulundu (p<0,05). Her dört grubun korteks, subkorteks ve serebellumlarındaki nitrit ve nitrat değerleri arasında istatistiksel olarak fark tespit edilmedi (p>0,05). Bu çalışmanın sonuçlan, beynin kendisini diyabetik hiperglisemiye bağlı oksidatif strese karşı kan-beyin bariyerinde bulunan özel glukoz taşıyıcılarının down-regüle olması, beyinde yer alan antioksidan savunma sistemlerinin artışı gibi çeşitli adaptif mekanizmalarla koruduğunu düşündürmektedir. Ayrıca melatonin ve pentoksifilin uygulamasının diyabetin oluşturacağı beyin dokusu lipit peroksidasyonunu önleyebileceğini göstermektedir. VII

SUMMARY The oxidative stress plays an important role in pathogenesis of diabetes mellitus. Diabetes mellitus could effect both central arid peripheral nervous system. Detrimental effects of diabetes mellitus on the central nervous system is less well studied compared to the effects on peripheral nervous system. The aim of this study is to search the changes in central nervous system induced by diabetes mellitus in long term period. In this study, mice of Mus musculus Swiss albino species were separated into four groups: alloxan-induced diabetes mellitus (DM), alloxan-induced diabetes with melatonin supplementation (DM+MLT), alloxan-induced diabetes with pentoxifylline administration (DM+PTX), and the control group. The study was carried on for eight weeks after the induction of diabetes. Mice have been decapitated under ether anesthesia at the end of the study and the brains were dissected into cortex, subcortex and cerebellum parts. Malondialdehyde (MDA), nitrite and nitrate levels were measured in each of these parts. While the body weights were increased in the controls, DM+MLT and DM+PTX groups (p<0,01), there was no change in DM group (p>0,05). At the end of this study, levels of fasting glucose in DM, DM+MLT and DM+PTX groups were measured higher compared to the control group (p<0,001). There were no significant differences in MDA levels of cortex, subcortex and cerebellum of the control group compared to the DM. DM+PTX, and DM+MLT groups (p>0.05). MDA levels in each of the three brain layers of DM+MLT and DM+PTX groups were found significantly lower than the DM group (p<0,05). There weren't any significant differences among the nitrite and nitrate levels in cortex, subcortex and cerebellums of all of the 4 groups (p>0,05). As a result of this study, it was concluded that various adaptive defence mechanisms such as the down regulation of specialized glucose transporters of the blood brain barrier and the increase of antioxidant defence systems of the brain play an important role to protect the brain against the oxidative stress caused by diabetic hyperglycemia. Besides this it was indicated that administration of melatonin and pentoxifylline could prevent the lipid peroxidation of the brain tissue caused by diabetes. VHI
