Özet
104 ÖZET Broylerlerde Doğal ve Sentetik Antioksidanların Bazı İz Elementler Üzerine Etkisi. Bu çalışmada, etlik piliçlerin kan serumunda enzimatik antioksidanların ko-faktörü olarak görev yapan ve birbirileriyle etkileşimleri sonucunda miktarlan değişen demir (Fe), bakır (Cu), çinko (Zn), mangan (Mn) ve selenyum (Se) değerleri ölçülmüştür. Çalışmada her grup için 25 adet piliç olmak üzere kontrol, vitamin A, C, E ve BHT destekli antioksidan gruplarında toplam 125 adet broyler civcivi kullanılmıştır. Ross PM-101 genotipli erkek broyler civcivlerin birinci gününden başlamak üzere altı hafta oluncuya kadar her hafta canlı ağırlık daralan alınmış ve 42 günün sonunda kesime sevk edilmiştir. Kesim sırasında her piliçten 10 mL kan alınmış ve laboratuvarda serumları ayrıldıktan sonra analizleri geciktirilmeden yapılmıştır. Mikro mineral maddelerden demir (Fe), bakır (Cu), çinko (Zn) tayini için Perkin-Elmer Flame Atomik Absorpsiyon Spektrofotometriden (F-AAS) yararlanılmıştır. Mangan (Mn) ve selenyum (Se) tayini için grafit firm kullamlmıştır. Analizler sonucunda etlik piliçlerin ortalama kan serumu iz element değerleri bulunmuş ve istatistik değerlendirmede, kontrol grubu ile diğer antioksidan destekli gruplar arası tek yönlü varyans analizi ile farkın önemliliği için Duncan çoklu karşılaştınlmasından faydanılmıştır. Kan serumu ortalama Fe değerleri kontrol grubunda 4.00 ± 0.84 ug/mL; vitamin A grubunda 3.95 ± 0.10 ug/mL; vitamin C grubunda 4.95 ± 0.08 ug/mL; vitamin E grubunda 1.45 ± 0.07 ug/mL ve BHT grubunda 1.96 ± 0.29 ug/mL olarak seviyeleri tespit edilmiştir. Kontrol ve vitamin A grupları Fe düzeyleri arasında fark bulunmazken, bu gruplarla hem vitamin C grubu hem de vitamin E ve BHT grupları arasındaki farklılıklar belirgin ve istatistik olarak da anlamlıdır (pO.001). Kontrol ve vitamin A grubu, vitamin C grubundan istatistik olarak önemli düzeyde (p<0.001) daha düşük, vitamin E ve BHT gruplarından da istatistik olarak önemli düzeyde (p<0.001) daha yüksek serum Fe değerlerine erişmişlerdir. Antioksidan ilave edilen gruplardaki piliçlerin serum Cu düzeyleri ile kontrol grubu piliçler arasında önemli farklılıklar saptanmış ve parametreler arasında anlamlı bir istatistiksel ilişki hesaplanmıştır. Tespit edilen serum ortalama Cu düzeyleri kontrol grubunda 0.97 ± 0.01 ug/mL; vitamin A grubunda 0.63 ± 0.02 ug/mL; vitamin C grubunda 0.12 ± 0.00 ug/mL; vitamin E grubunda 0.25 ± 0.02 ug/mL; BHT grubunda 0.18 ± 0.01 ug/mL olarak bulunmuştur. Kontrol grubunun serum Cu düzeyi tüm gruplardan p<0.001 istatistik önemlilikte daha yüksek ve vitamin C grubunun serum Cu düzeyi ise tüm diğer gruplardan istatistik önemlilik düzeyinde daha düşüktür (pO.001). Tüm gruplar arasında gözlenen serum Cu düzeylerindeki farklılıklar belirgin olup tek yönlü varyans analizi ve Duncan testi ile gerçekleştirilen gruplar arası farklılıklar pO.001 düzeyinde önemlilik göstermiştir. Yemlere eklenen antioksidanlardan vitamin A alan broylerlerin serum Cu düzeyleri diğer doğal ve sentetik antioksidan ilave edilen gruplardan daha yüksek ölçülmüştür (pO.001).105 Kan serumu ortalama Zn değerleri kontrol grubunda 2.80 ± 0.13 ug/mL; vitamin A grubunda 2.48 ± 0.08 ug/mL; vitamin C grubunda 2.63 ± 0.07 ug/mL; vitamin E grubunda 2.73 ± 0.06 ug/mL ve BHT grubunda 2.83 ± 0.09 ug/mL olarak bulunmuştur. Değerlerden de anlaşılacağı üzere en yüksek serum Zn düzeyini kontrol grubu ile yemlerine sentetik antioksidan BHT ilave edilen gruptaki broyler göstermektedir. Ancak tüm grupların serum Zn düzeyleri birbirine yakın olup, gruplar arasındaki farklılıklar istatistik yönden önemli bulunmamıştır (p>0.05). Buna rağmen Vitamin A grubunun serum Zn düzeyi tüm gruplardan daha düşük gözükmektedir. Serum Mn düzeyleri kontrol grubunda 7.43 ± 0.39 ug/L'dır. Bu düzey ile kontrol grubu broylerleri en yüksek mangan düzeyine erişmişlerdir (p>0.001). Vitamin A grubunda 7.04 ± 0.32 ug/L, vitamin C grubunda 3.57 ± 0.13 ug/L, vitamin E grubunda 6.88 ± 0.32 ug/L ve BHT grubunda 4.78 ± 0.22 ug/L olarak belirlenmiştir. Vitamin C grubunun serum Mn düzeyi tüm gruplardan pO.001 istatistik önemlilikte dana düşüktür. Kontrol vitamin A ve vitamin E grupları Mn düzeyleri birbirine yakın tesbit edilmiş olup bu gruplar arasında istatistik fark bulunamamıştır. Ancak bu gruplarla hem vitamin C grubu hem de BHT grupları arasındaki farklılıklar bariz ve istatistik olarak da anlamhdır (pO.001). Vitamin C grubunun serum Mn düzeyi kontrol, vitamin A ve vitamin E gruplarının eriştiği serum Mn düzeyinin yansı kadardır (pO.001). Bulgular selenyum yönünden incelendiğinde, serum selenyum düzeyi kontrol grubundaki piliçlerde ortalama olarak 5.84 ± 0.20 ug/dL'dır. Bu düzey vitamin A grubunda 2.53 ±0.11 ug/dL, vitamin C grubunda 2.18 ± 0.10 ug/dL, vitamin E grubunda 2.87 ± 0.14 ug/dL ve BHT grubunda 3.25 ± 0.11 ug/dL olarak belirlenmiştir. En yüksek serum selenyum düzeyi kontrol grubundaki broylerlerde olup tüm gruplardan pO.001 istatistik önemlilikte daha yüksektir. Vitamin C grubu broylerlerin serum Se düzeyleri tüm gruplardan daha düşüktür (pO.001). Kontrol grubundan sonra en yüksek Se düzeyi BHT grubunda ölçülmüştür. Vitamin A grubu ile vitamin C ve vitamin E grupları Se düzeyleri arasındaki rakamsal farklılıkların istatistik önemi yoktur (p>0.05). Vitamin E grubu ile BHT grubu Se düzeyleri arasındaki fark da istatistiksel olarak önemsizdir (p>0.05). Sonuç olarak, kan serumunda vitamin C-Fe, vitamin A-Cu, vitamin A,E-Mn ve BHT-Se değerlerinde pO.001 düzeyinde istatistik önemlilikte artışa; ancak vitamin E-Fe, vitamin C-Cu ile Mn ve vitamin A,C,E-Se değerlerinde pO.001 düzeyinde istatistik önemlilikte düşüşe neden olmuştur. Ayrıca serum Zn düzeyleri ise birbirine yalan olup gruplar arasındaki farklılıkların istatistik önemi bulunmamıştır (p>0.05). Böylece broyler rasyonlanna ekstra olarak ilave edilen antioksidan vitaminlerin (vitamin A, vitamin C, vitamin E) ve sentetik bir antioksidan olan BHT'nin, önemli bazı iz elementlerin (Fe, Cu, Zn, Mn, Se) metabolizması üzerine beklenen sinerjist veya antagonist etkileri ile prooksidatif-antioksidatif etkileşimleri gerçekleşmiştir.

106 ABSTRACT The Effect of Natural and Synthetic Antioxidants on Some Trace Elements in Broilers In this study, the levels of Ferro (Fe), Copper (Cu), Zinc (Zn), Mangan (Mn) and Selenium (Se), that the amount of which are changed following their interactions and are co-factors of enzymatic antioxidants in blood sera of broilers, were detected. As 25 broilers per each group, for control group and antioxidant groups supported by Vitamin A, C, E and BHT, total 125 broiler chicks were used. The live weight of male broiler chicks with Ross PM-101 genotype were detected from 1 day age to 6 week-age and at the end of the age of 42 days, they were slaughtered. After slaughter, 10 mL blood samples were collected from each broiler and following to the separation of sera the analyses were done without any delay. In micro-mineral analyses; by using 800 Analyst Perkin-Elmer Flame Atomic Absorption Spectrometry (F-AAS) for detection of levels of Fe, Cu and Zn was used. For the detection of Mn and Se, Furnace-Transverse Heated Graphite Atomizer (F-THGA) was used. At the end of analyses, the average trace element levels in the blood were detected and in statistical evaluation, variance analysis (One- Way ANOVA and Duncan Test) between control group and the groups supplemented with antioxidants was carried out. Average Fe levels of blood sera were detected to be 4.00 ± 0.84 ug/mL in control group, 3.95 ± 0.10 ug/mL in Vitamin A group, 4.95 ± 0.08 ug/mL in Vitamin C group, 1.45 ± 0.07 ug/mL in Vitamin E group and 1.96 ± 0.29 ug/mL in BHT group. While no statistically important difference (p>0.05) was found between the control group and Vitamin A group, the difference between control group and Vitamin C, E and BHT group was statistically very important (p<0.001). It was the same for Vitamin A and Vitamin C, E and BHT group (p<0.001). In another word, control group and Vitamin A group animals had statistically lower serum Fe levels compared with Vitamin C group animals. And Vitamin E and BHT group animals had statistically higher serum Fe levels compared with Vitamin C group animals. The differences between blood serum Cu levels of the broilers of the control group and the group supported with antioxidants were important and thus important relation could be established between the parameters. Detected average blood sera Cu levels were found to be 0.97 ± 0.01 ug/mL in the control group, 0.63 ± 0.02 ug/mL in Vitamin A group, 0.12 ± 0.00 ug/mL in Vitamin C group, 0.25 ± 0.02 ug/mL in Vitamin E group and 0.18 ± 0.01 ug/mL in BHT group. Serum Cu level of control group animals was higher which was statistically very important (pO.OOl) compared with those of the other group animals. Serum Cu level of Vitamin C group animals was lower compared with those of the other group animals. The difference between them was statistically very important (pO.OOl). The differences among all group animals' sera Cu levels were statistically very important (pO.OOl) by One- Way ANOVA and Duncan Test. Sera Cu levels of Vitamin A group animals were higher than those of the other group animals107 supported by the other natural and synthetic antioxidants and difference between them was statistically very important (p<0.001). Average blood sera Zn levels were found to be 2.80 ±0.13 ug/mL in the control group, 2.48 ± 0.08 ug/mL in Vitamin A group, 2.63 ± 0.07 ug/mL in Vitamin C group, 2.73 ± 0.06 u.g/mL in Vitamin E group and 2.83 ± 0.09 u.g/ml in BHT group. According to the results, serum Zn levels of control and BHT group animals were the highest compared with those of other groups animals. However, the differences between serum Zn levels of all group animals were statistically not important (p>0.05). In spite of this, serum Zn level of Vitamin A group animals was seen the lowest. Average blood sera Mn levels were found to be 7.43 ± 0.39 u,g/L in the control group, 7.04 ± 0.32 u.g/L in Vitamin A group, 3.57 ± 0.13 ug/L in Vitamin C group, 6.88 ± 0.32 ug/L in Vitamin E group and 4.78 ± 0.22 u.g/L in BHT group. According to the results, serum Mn levels of control group animals were the highest compared with those of other group animals. Contrarily, sera Mn levels of Vitamin C group animals were the lowest. The differences between Vitamin C group sera Mn levels and those of other group animals were statistically very important (p<0.001). However, the differences (p>0.05) were statistically not significant between the control, Vitamin A group and Vitamin E group animals sera Mn levels. The differences between sera Mn levels of Vitamin C group animals and other groups animals, and BHT group animals and other groups animals were statistically very important (pO.001). Average blood sera Se levels were found to be 5.84 ± 0.20 u.g/dL in the control group, 2.53 ± 0.11 ug/dL in Vitamin A group, 2.18 ± 0.10 ng/dL in Vitamin C group, 2.87 ± 0.14 u.g/dL in Vitamin E group and 3.25 ±0.11 ug/dL in BHT group animals. Sera Se levels of control group broiler were the highest compared with those of other group animals and differences between these were statistically very important (pO.001). Sera Se levels of Vitamin C group broiler were the lowest and difference with other group were statistically very important (pO.001). Following the control group animals, sera Se levels of BHT group broilers were highest. The results in Vitamin A, C and E group animals were statistically not important (p>0.05). The difference between Vitamin E and BHT group animal sera Se levels were also statistically not important (p>0.05). In conclusion, Vitamin C, A, A and E, and BHT caused statistically very important increase in sera Fe, Cu, Mn and Se levels, respectively (p<0.001). Contrarily, Vitamin E decreased sera Fe level, Vitamin C sera Cu and Mn level, and Vitamin A, C and E sera Se level (pO.001). However, the differences of sera Zn levels in all group animals were statistically not important (p>0.05). As a result, it was seen that natural antioxidant vitamins (Vitamin A, C, and E) and synthetic antioxidant BHT added to the ratios of the broilers as supplement showed synergist and antagonist effects with the pro-oxidative and anti-oxidative interactions that were expected.
