-53- SUMMARY o(..-Acid glycoprotein (AAG) is an important sialoglycoprotein for the binding of drugs in plasma. It contains a very high amount of carbohydrates compared with other glycoproteins and the sialic acids occupy the major part of this carbohydrate moiety. The studies concerned with the binding of drugs to AAG have been focused on the role of sialic acid residues in this binding. This study was planned to examine the alterations in the levels of AAG, total sialic acid and the AAG-bound sialic acids induced by the administration of heterocyclic drugs chosen from different psycho- pharmacological groups. The alterations in the activities of GGT and its izoforms, which are structurally sialoglycoproteins, were also examined. Forty male Wistar rats aged 3-3.5 months were used. Chlorpromazine (largactil), amitriptyline (laroxyl) and diazepam (diazem) were administered intraperitoneally and the control group was injected physiological saline solution. The experimental period lasted for 30 days. Then the animals were sacrified and biochemical measurements were carried on the sera and liver tissues.-54- Serum AAG levels were found to be increased in the amitrlptyline group t but the total and AAG-bound sialic acid levels in serum and the sialic acid levels in liver were found to be unchanged. We suggested that the role of sialic acid residues in the binding of amitriptyline to AAG is minor and the increased levels of AAG were due to the highest selectivity of this drug. Serum AAG levels remained the same in the chlorpromazine group but the total and AAG-bound sialic acid levels were both found to be increased. A discriminating observation in this group was the elevated levels of sialic acid in the liver tissue. In the group of animals which received diazepam, all the measurements related to AAG and sialic acids in sera gave significantly high values. On the other hand, serum GGT activity appeared to be decreased significantly although no difference was observed in the activity of the enzyme in liver tissue. It was postulated that the disturbance in the balance between the protein fractions and the increase in sialic acids in serum may cause an inhibition of the enzyme.
