.u Hilal fl(XtrM*-m*(ıL- uınalucloy Anabil üaU ? ^'2_ SUMMARY 1- Ih ThTs sTTüdy, tB^7“roXe”“öt''”vaT“tıra5'_Hi”“anı^”“”if«“”“”~«tıtTtgtnr-i-st xm histamine (3xİ0-T-10-° M) 4-methylhistamine (3xi0-*-10-« M) induced positive chronotropic effect has been investigated in spontaneously beating right atria of mice. 2- Histamine and 4-methylhistamine caused a concentration related increase in the rate of isolated right atria of mice. 3- Me py ramine (10~s, 10~a, 10~TM) as Ha. receptor blocker, famotidin (10-*, 10~T, 10-*M), cimetidine (10~s, 10-T, 10-“8M) and lupitidin (SKF 93579) (10”*, 10~a, 1Q-«M) as Ha receptor blockers and ikotidin (10_a, 10-T, 10_SM) as a nonspecific Hi and Ha receptor blockers were used to assess affinity constants for their respective receptors. 4- Beside the specific histamine receptor blockers, a 0-blocker propranolol (10~s, 10_T, 10~eM) was used for indicating whether noradrenaline release occured in this tissue or not. 5- An uptake i blocker cocaine (10~TM) also used to reverse an amine uptake. 6- For estimating of the apparent pAB values, Schild regression was used as an indicator of simple competitive inhibition. 7- Since the slope and linearity of a Schild regression are sensitive indicators of non-equilibrium conditions in an isolated tissue, in the present study, deviations of the slopes from unity evaluated from the Schild regressions were used as a tool to define the possible mechanisms which were involved in HA- induced responses.8- Results were indicated that isolated right atrium of mice has heterogenous histamine receptor distribution (Hi and He receptors). Both receptors were mediated by HA- induced chronotropic effect with an involvement of noradrenaline release which was potentiated, the chronotropy. It has also been found that, there was a significant amine uptake in this t i s s ue.
