Özet

This descriptive research was done for searching dyadic adjustment at women who had hysterectomy at Ege University The Hospital of Faculty of Medicine, Obstetrics and Gynecology Department and policlinic between the dates of May 22 ? August 14 2006. In 2005, 600 women had hysterectomy at this gynaecology department. This research sampling which calculated by repeating of realizing event method is 82 women. ?Dyadic Adjustment Scale? developed by Spanier (1976) which has 32 item and validity and reliable was done by Fısıloglu and Demir (2000) was used for collective of data. According to scale high point means that the quality of relation. Womens average pre and post hysterectomy points was found statistically meaningfull but average Dyadic Adjustment scale points and demographic characteristic's relation was not found statistically meaningfull. Finally, women who have hysterectomy and different demographic characteristics dyadic adjustment effect from hysterectomy.

In this thesis, were investigated and developed liquid suppositories composed of two different series poloxamer (P407/P188), ketoprofen and different bioadhesive gelled at physiological temperature and was mucoadhesive to the rectal tissues without leakage after the dose. First of all were made standardization and purity controls of the drug that would be used in the formulations. Using cold process were prepared verious formulastions composed of only poloxamer mixtures, poloxamer mixtures and ketoprofen, poloxamer mixtures, ketoprofen and different bioadhesive polymers with different concantration. In a low-temperature thermostat water bath, with a digital thermosensor were measured the gelation temperature of formulations. 4%/20%/2.5%-P407/P188/ketoprofen formulation was choosed more suitable mixture as bases of liquid suppositorys, which gelled at 37.1 ºC. Then were prepared thermaly gelling suppositories contained 4%/20%/2.5%-P407/P188/ketoprofen and 0.2, 0.4, 0.6, 0.8% Carbopol, HPMC, CMC and PVP. Mechanical properties of formulations were measured using Texture Analyzer. The viscosities of liquid suppositories at room temperature and 37°C were measured using a Brookfield digital viscometer-III. In vitro release of ketoprofen from liquid suppository formulations was made by the USP XXII rotating basket method using a semipermeable membrane. In vitro release of ketoprofen from conventional suppository formulation was made by the USP XXII rotating basket method. After completing the in vitro release rate tests release kinetics were determined. The mechanism of release of ketoprofen from liquid suppositories was analyzed using Peppas equation. Two liquid suppositories composed of 4%/20%/2.5%/0.8%-P407/P188/ketoprofen/karbopol and 6%/20%/2.5%/0.8%-P407/P188/ketoprofen/karbopol and each one containing 0.1% blue lake were administrered into the rectum of rabbits 4 cm above the anuse using a stomach sondle needle. After 30 min administration, the rabbits were sacrified and the rectums were sectioned and the localization of poloxamer gels in the rectum were indetified by the blue color.
