Bu çalışmada, sıçan karaciğeri ve akciğeri karışık fonksiyonlu oksidazlan (anilin 4-hidroksilaz, AH; aminopirin N-demetilaz, AMND: 7-etoksiresorufîn O-de- etilaz, 7-ERDE; p-nitroanisol O-demetilaz, p-NAOD; sitokom P-450; sitokrom b

In this study, the influence of various treatments of cigarette smoke, benzo(a) pyrene, alcohol on rat liver and lung mixed function oxidases (aniline 4- hydroxylase, AH; aminopyrine N-demethylase, AMND; 7-ethoxyresorufine O-de- etthylase; p-nitsanisole O-demethylase, p-NAOD; cytochrome P-450; cytochrome D5), heme, lipid peroxidation (LP), reduced glutathione (GSH) and glutathione S- transferases (GST) toward several substrates (l,chloro-2, 4-dinitrobenzene, CDNB; 1,2 dichloro-4-nitrobenzene, DCNB; ethacrynic acid, EAA; l,2-epoky-3- (p-nitrophenoxy) propane, ENPP) were examined. 1) Time: Course of effect of cigarette smoke on mature male rat liver and lung xenobiotic metabolism. When the nature male rats were exposed to cigarette smoke for 3 days, the liver AMND, 7-ERDE, p-NAOD and EAA GST activitie and GSH level increased significantly whereas ENPP GST activity and LP level decreased significantly. No changes were observed in the other parameters examined. The lung AH, 7-ERDE and p-NAOD activities and cytochrome P-450, cytochrome b^, heme, LP and GSH levels were significantly elevated by cigarette smoke. ENPP GST activity, however, significantly decreased. The other parameters examined did hot alter. Upon exposur to cigarette smoke for 7 days, the rat liver 7-ERDE p- NAOD activities, and GSH level significantly increased while ENPP GST activity significantly decreased. No changes were noted in the other parameters examined. In the lung, 7-ERDE, p-NAOD, and EAA and GST activities, and cytochrome P- 450, cytochrome b$ and GSH levels significantly increased. The other parameters remained unaltered. After 15 days exposure to cigarette smoke the rat liver 7-ERDE, p-NAOD and ENPP GST activities, and GSH level significantly increased whereas the other parameters remained unchanged. In the lung, AH, 7-ERDE, p-NAOD and ENPP GST activities and cytochrome P-450, cytochrome b5, heme and GSH levels were observed to be increased significantly. No changes were noted in the other parameters.2) The effect of BaP on nature male rat liver and lung xenobiotic metabolism. BaP increased the rat liver 7-ERDE, p-NAOD and CDNB, DCNB and ENPP GST activities significantly. In addition, cytochrome P-450, cytochrome D5, heme and LP levels increased significantly. EAA GST activity, however, significantly decreased. No changes occured in the other parameters. In the lung, AH, 7-ERDE, p-NAOD, CDNB GST activities were increased significantly by BaP lung cytochrome P-450, cytochrome D5 and heme levels were also eleveted significantly by BaP. The other parameters were not influenced. 3) The effect of the smoke of foreign cigarette with low tar content on mature rat liver and lung xenobiotic metabolism. Three days of exposure to smoke of cigarette which contained low tar increased the mature male rat liver 7-ERDE and p-NAOD activities significantly. EAA and ENPP GST activities, however, were significantly depressed by the so me treatment. No changes were observed in the other parameters. Lung 7-ERDE and p-NAOD activities, and cytochrome P-450,cytochrome b^, heme and GSH le vel significantly increased. DCNB GST activity significantly decreased. The other parameters were not influenced. 4) The effect of cigarette smoke on mature female rat liver and lung xenobiotic metabolism. When the female matura rats were exposed to cigarette smoke for 3 days, the liver AMND, 7-ERDE, p-NAOD and DCNB GST activities; and GSH levels increased significantly. No changes were determined in the other parameters. In the lung, AMND, 7-ERDE, p-NAOD and CDNB and DCNB GST activies significantly increased. In addition, elevations were also observed in the cytochrome P-450, cytochrome b^, heme, LP and GSH levels upon exposure to cigarette smoke. The other parameters remained unchanged.5) Age-dependent alterations in the male rat liver and lung xenobiotic metabsolism by cigarette smoke. When the 20 days old male rats were exposed to cigarette smoke for 3 days liver AH, 7-ERDE and p-NAOD activities and cytochrome P-450, heme and LP levels increased significanty ENPP GST activity, however, decreased significantly. No changes were noted in the other parameters. In the lung, AH, 7- ERDE, p-NAOD and ENPP GST activities, and cytochrome P-450 and heme levels increased significantly. No alterations were observed in the other parameters. When the 360 days old male rats were exposed to cigarette smoke for 3 days, liver 7-ERDE and p-NAOD activities increased significantly on the other hand, AMND, and CDNB, DCNB and EAA GST activities significantly decreased. The other parameters remained unaltered. In the lung, although 7- ERDE activity and LP level increased significantly, CDNB, EAA and EPNN GST activities decreased significantly. The other parameters did not change. 6. The effect of interaction of cigarette smoke and alcohol on mature male rat liver and lung xenobiotic metabolism. When the alcohol was given to the animals in drinking water for 3 weeks, liver AH, AMND and 7-ERDE activities and cytochrome P-450, cytochrome b5, heme, LP and GSH levels increased significantly. The other parameters did not alter. In the lung, alcohol treatment increased CDNB and ENPP GST activities, and cytochrome P-450 and LP levels significantly. DCNB GST activity was inhi bited by alcohol treatment. No changes were noted in the other parameters. When the animals were exposed to alcohol concamitantly with cigarette smoke, liver AH, AMND, 7-ERDE and p-NAOD activities and cytochrome P- 450, cytochrome b^, heme, LP and GSH levels significantly increased. CDNB and ENPP GST activities significantly decreased. The other parameters remained unchanged. Combined treatment increased lung AH, AMND, 7-ERDE, p-NAOD, and CDNB, DCNB and ENPP GST activities significanty. In addition, cytochrome P-450 and LP levels were elevated significantly by the combined treatment. The other parameters were not altered by the combined treatment.
