Özet

Genetik olarak kodlanmış sensörler biyolojik süreçlerin aydınlatılması için güçlü araçlardır. Bu tür sistemler otofloresans proteinlerin ilgili analiti tanıma ve bağlama yeteneğine sahip olan biyoloji makromoleküller ile genetik olarak birleştirilmesi ile kolaylıkla geliştirilebilirler. Bu tez kapsamında genetik olarak kodlama yöntemi ile çeşitli ligand bağlayıcı proteinler ve fotoproteinlerden oluşan biyosensör sistemleri tasarlandı. Biyosensörlerin tasarımı için, floresans proteinlerin ligand bağlayıcı proteinlere doğrudan füzyonu ve biyoluminesans rezonans enerji transferi (BRET) stratejileri kullanıldı. Maltoz bağlayıcı protein, aspartat dehidrogenaz ve alloz bağlayıcı proteinin tanıma elemanı olarak kullanıldığı biyosensörler ile maltoz, L-aspartat, ve D-alloz analizleri gerçekleştirildi. Genetik olarak kodlanmış biyosensörlerin en önemli özelliklerinden birisi in vivo uygulama potansiyelidir. Bu tez kapsamında in vitro karakterizasyonları gerçekleştirilmiş olan maltoz, aspartat ve alloz biosensörleri, in vivo uygulamalar için de model sistemlerdir.

Genetically encoded sensors are powerful tools for elucidation of biological processes. These systems can be simply developed by genetically fusing autofluorescent proteins to biological macromolecules, which are capable of recognizing and binding the analyte of interest. In this thesis, biosensor systems consisting of photoproteins and ligand binding proteins have been designed via various genetically encoding strategies. Fusion proteins of fluorescent proteins and ligand binding proteins were developed by using either bioluminescent resonance energy transfer (BRET) strategy or a direct fusion approach. Maltose binding protein, aspartate dehydrogenase and allose binding protein were used as recognition elements in those biosensors. Analysis of maltose, L-aspartate and D-allose was achieved by the developed biosensors. One of the most important features of the genetically encoded sensors is their potential for in vivo applications. Therefore, maltose, L-aspartate and D-allose biosensors, which have been characterized in vitro could be considered as model systems for in vivo applications.
