Özet

The aim of this research is to determine the most appropriate nutrient media content in anther culture. Moreover, the effect of using maltose as carbohydrate source and silver nitrate in nutrient media was studied in this thesis. The anthers of four different pepper varieties (Asena, Athena, Naomi and Rain Nil) were cultured in six different nutrient culture media combination with different sugar and various concentration of AgNO3.Callus and embryo in all the genotypes were obtained but plant regeneration was observed only in Asena (capia type) and Rain Nil (bell type) genotypes. M10 and M15 nutrient media which include maltose as a carbohydrate source gave the highest result for callus, embryo, and plant regeneration. However the M0 nutrient media without silver nitrate was not succesfull for adrogenesis compare to the other media that include maltose. The sucrose as a carbohydrate source in nutrient media also gave succesfull results depending on the genoytpes and AgNO3 composition. As a result, Asena genotype gave the best adrogenesis performance in maltose media with silver nitrate (10mg/l 15mg/l). Results are evaluated in terms of variety-nutration media the Asena genotype in M10 (71,1%) and M15 (74,2%) cultured media gave the best interaction for callus regeneration. In terms of the culture media for embryo regeneration, M15 (66,2 %) cultured media gave the most succesfull results and in genotypes the best success was achieved from Asena (60,1) genotype. Plant regeneration were oblained from the Rain Nil and the Asena genotypes. M15 cultured media (12,7%) and the Rain Nil genotype (8,4%) showed success for oblained plants from the pepper anthers. However, the best interactions for plant regeneration was derived from M15 cultured media for Rain Nil genotype (28,5%).
