Özet

Amaç: Bu çalışmanın amacı, kafeik asit fenetil ester (KAFE)'in konak yanıtını düzenleyip periodontal doku yıkımını ve kemik kaybını önleyebileceği veya azaltabileceği hipotezini test etmek ve deneysel periodontitis modelinde dişeti ve serum interlökin-1β (IL-1β) ve tümör nekroz faktör-α (TNF-α) seviyeleri üzerine etkisini biyokimyasal olarak incelemektir. Materyal ve Metot: Çalışmamızda 40 adet Sprague-Dawley sıçan rastgele 4 gruba ayrıldı. Çalışma grupları; Grup A: Kontrol (n=10), Grup B: Deneysel periodontitis, Grup C: Deneysel periodontitis+KAFE (5μmol/kg/gün), Grup D: Deneysel periodontitis+KAFE (10μmol/kg/gün) olarak belirlendi.

Aim: The aim of this study was to test the hypothesis that caffeic acid phenethyl ester (CAPE) may avoid or downregulate the periodontal tissuse distruction via host modulation and investigate the impact of this substancebiochemically on the levels of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in gingiva and serum in an experimental periodontitis model. Material and Method: 40 Sprague-Dawley rat were randomly divided into four groups. Group A: control, Group B: experimental periodontitis (EP), Group C: EP+CAPE (5μmol/kg/day), Group D: EP+CAPE (10μmol/kg/day) were designated as study groups. EP was created by injection of lipopolysaccharide (LPS) to the interproximal area of left maxillary first and second molars. Followed by the EP, CAPE was administered intraperitoneally (i.p.) to the Groups C and D in respect to the related dosages. Gingival and serum samples were investigated biochemically using enzym linked immuno assay (ELISA) kit peculiar to IL-1β and TNF-α. Alveolar bone loss was measured morphometrically. Results: The findings confirmed the bone loss around LPS injection sites. Serum levels were not statistically different between the groups (p>0,05). The levels of gingival tissue IL-1β and TNF-α in Group A were found significantly lesser than the periodontitis groups (Grup B, C and D) (P<0.001). The levels of gingival tissue levels of IL-1β and TNF-α in CAPE groups (Group C and D) were found statistically lesser than periodontitis group (Group B) (P<0.001). Conclusion: Within the limitations of this study, our results suggest that CAPE reveals antiinflammatory effect via host modulation. Long term clinical studies are needed to indicate CAPE as an antiinflammatory agent on periodontal tissues.
