Özet
Bu çalışmada, fluopyram ve tebukonazol kombinasyonlu Luna Experience SC 400 fungusitinin sıçan karaciğer ve kan dokularında genotoksik etkileri ve oksidatif hasar potansiyeli araştırılmıştır. Çalışmada; genotoksik etkiyi belirlemek için Mikronukleus yöntemi ve oksidatif hasara bağlı DNA hasarını belirlemek için tek hücre jel elektroforez yöntemi kullanılmıştır. Oksidatif hasar potansiyelini belirlemek için antioksidan enzimlerden biri olan katalaz (CAT) düzeyi ölçülmüştür. Lipit peroksidasyonunu belirlemek için ise malondialdehit (MDA) düzeyi ölçülmüştür. Sıçanlara verilecek olan dozlar, 5 mg/kg, 10 mg/kg ve 20 mg/kg olarak belirlenmiştir. Komet ve MN test yönteminde pozitif kontrol gruplarına 2 mg/kg Mitomycin-C (MMC) uygulanmıştır. Komet testi sonucunda elde edilen hasarlı hücre yüzdesi (HHY) ve genetik hasar indeksi (GHI) ve MN sonuçları istatiksel açıdan doz artışına bağlı olarak anlamlı bir artış göstermektedir. CAT ve MDA değerlerinde istatiksel açıdan anlamlı bir değişim gözlenmemiştir. Yapılan bu çalışmada elde edilen veriler Luna Experience SC 400 fungusitinin sıçan karaciğer ve kan dokularında genotoksik etkiye sebep olduğunu, CAT'a bağlı oksidatif hasar potansiyeli üzerinde bir etkisi olduğu, ancak MDA'ya bağlı oksidatif hasar potansiyeli üzerinde bir etkisi olmadığını göstermektedir.

In this study, potential genotoxic effects and oxidative damage of Luna Experience SC 400 fungicide which is combinated Fluopyram and Tebuconazole in rat liver and blood tissues was investigated. In this study; to determine the genotoxic effect, micronucleus assay was used, to determine DNA damage due to oxidative stress, single cell gel electrophoresis (comet assay) was used. To determine the oxidative stress potential of the Luna Experience SC 400, catalase level (CAT) which is one of the antioxidant enzymes were measured. To determine the lipid peroxidation, malondialdehyde (MDA) levels were measured. Doses of Luna Experience SC 400 which were given to rats were determined as 5 mg/kg, 10 mg/kg and 20 mg/kg. In the positive control groups for comet and MN assay, 2 mg/kg MMC was used. As a result of comet and MN assay, percentage of damage cells and arbitary unit was statistically increased depend on doses. CAT and MDA levels were not changed statistically. The results of this study show that Luna Experience SC 400 fungicide has genotoxic effect and on CAT-dependent oxidative stress potential, but doesn't have MDA-dependent oxidative stress potential in rat liver and blood tissues
