This work had two objectives. The first one was to study the effect of oxidoreduction potential modified by gas (N2, N2-H2) on microbial quality, color retention and vitamin C stability of orange juice during storage. The second objective was to develop an apparatus for biological-energetic studies in controlled physico-chemical conditions. Results demonstrate the difference in the capability of intact and injured cells of spoilage microorganisms to spoil citrus juice and the different thermal resistance of cells. While Lactobacillus plantarum was more resistant to heat treatment than Saccharomyces cerevisiae growth recovery after pasteurization was faster for the latter microorganism. Whereas the oxidant conditions were the most effective for thermal destruction of L. plantarum and S. cerevisiae, the reducing conditions decreased recovery of heated cells of S. cerevisiae. In addition, gassing the juice with N2 or N2-H2 increased ascorbic acid retention and color stability. In the second part, we developed an apparatus that allow one to carry out the energetic and physiological experiments in controlled physico-chemical conditions. The method of ion selective electrodes was developed for intracellular pH measurement in lactic acid bacteria (L. plantarum and L. mesenteroides). Now, a single method can be proposed to study, simultaneously, the effect of environmental conditions on both components of the proton motive force ( and pH).
