Özet
Bu çalışmada hiperkolesterolemi oluşturulmuş farelerde Demir Dikeni (Tribulus Terrestris) ekstraktının oral gavaj yoluyla uygulanmasının farelerin testis dokusu üzerindeki etkilerinin histopatolojik olarak ve bu dokuda AQP1 ekspresyonu üzerine etkisinin immunohistokimyasal ve RT-PCR yöntemleri ile belirlenmesi amaçlanmıştır. Araştırma materyali olarak daha önceden çiftleşmemiş ve herhangi bir çalışmada kullanılmamış 43 adet / 40 günlük Balb-c cinsi erkek fare kullanıldı. Gruplar bir haftalık alıştırma süresi sonunda rastgele seçilen farelerden Kolesterol Grubu (n=8), Kolesterol ve Demir Dikeni Grubu (n=8), Demir Dikeni Grubu (n=8), Sham Grubu (n=8) ve Kontrol Grubu (n=8) olacak şekilde oluşturuldu. Kolesterol ve Kolesterol ve Demir Dikeni grubundaki farelere %3 w/w kolesterol intraperitonal (i.p) olarak 15 gün boyunca uygulandı. 15 günün sonunda Demir Dikeni ve Kolesterol ve Demir Dikeni gruplarına Demir Dikeni ekstraktı 6 mg/kg olacak şekilde oral gavaj yöntemi ile 7 gün boyunca verildi. Deney sonunda tüm gruplardaki fareler tartıldıktan sonra testis dokuları alındı. Hiperkolesterolemi oluşturulan farelerde Demir Dikeni ekstraktı uygulanması sonucunda farelerin canlı ağırlıklarının, testis ağırlıklarının ve serum total kolesterol düzeylerinin kontrol grubuna benzer düzeyde olduğu belirlendi. Kontrol, Sham, Demir Dikeni ve Kolesterol ve Demir Dikeni gruplarına ait farelerin testis dokularının normal histolojik yapıda olduğu ancak Kolesterol grubuna ait farelerin testis dokusunda seminifer tübüllerde hücre kayıpları ve hidropik dejenerasyon alanlarının olduğu görüldü. Sonuç olarak; Kolesterol grubuna göre Demir Dikeni ekstraktı uygulanan grupta AQP1 mRNA ekspresyonunun daha yüksek olduğu tespit edildi. AQP1 immunoreaktivite derecesinin ise diğer gruplara göre daha zayıf olan grubun hiperkolesterolemi oluşturulan grup olması testis dokusunda kolesterolün AQP1 üzerine inhibe edici bir etkiye sahip olduğunu düşündürmektedir.

In this study, it was aimed to determine the effects of administration of Tribulus Terrestris extract by oral gavage on mice testis tissue histopathologically as well as its effect on AQP1 expression with immunohistochemical and RT-PCR methods in mice with hypercholesterolemia. As research materials, 43, 40-day-old Balb-c male mice which had not been previously mated and used in any study were used. The groups were formed as Cholesterol Group (n=8), Cholesterol and Tribulus Terrestris Group (n=8), Tribulus Terrestris Group (n=8), Sham Group (n=8) and Control Group (n=8) from the mice randomly selected at the end of the one-week practice period. 3% w/w cholesterol were administered to the mice in the Cholesterol group, Cholesterol and Tribulus Terrestris group intraperitoneally (i.p) for 15 days. At the end of 15 days, 6 mg/kg Tribulus Terrestris extract were given to the Tribulus Terrestris and Cholesterol and Tribulus Terrestris groups by oral gavage for 7 days. At the end of the experiment, the mice in all groups were weighed and then their testicular tissues were taken. In consequence of the administration of the Tribulus Terrestris extract in mice with hypercholesterolemia, the mice's live weights, testis weights and serum total cholesterol levels were determined to be similar to the control groups. The testis tissues of the mice in the Control, Sham, Tribulus Terrestris and Cholesterol and Tribulus Terrestris groups were found to have normal histological structure, but in the testicular tissues of the mice belonging to the Cholesterol group, cell losses and hydrophilic degeneration areas were observed in the seminiferous tubules. As a result; it was found that the expression of AQP1 mRNA was higher in the group administered Tribulus Terrestris extract than the cholesterol group. The finding that AQP1 immunoreactivity level was weaker in the hypercholesterolemia group compared to other groups suggests that cholesterol in the testis tissue has an inhibitory effect on AQP1.
