Background: Food allergy prevalence is increasing all over the world. Recent epidemiologic 5 studies have shown the link between vitamin D3 insufficiency and food allergy. In this study, we 6 investigated the effect of cholecalciferol, a widely used supplement form of vitamin D3, on food 7 allergy and the possible pathway for the effect. 8 Methods: BALB/c mice were sensitized and challenged with ovalbumin (OVA) to generate an 9 experimental food allergy mouse model. The mice received cholecalciferol treatment in vivo. In 10 another set of experiment, cells of OVA-sensitized mice were treated with cholecalciferol and OVA 11 in vitro. In a different set of experiment, OVA-challenged mice received anti-CD69 mAb treatment 12 in vivo. Clinical food allergy symptoms, cellular and humoral immunological changes were 13 analyzed. 14 Results: Cholecalciferol reduced allergic diarrhea in food allergy mouse model. Cholecalciferol significantly decreased population of CD69+ CD4+ 15 T cells. Furthermore, anti-CD69 mAb treatment 16 reduced the allergic diarrhea symptom, suggesting of an inhibitory effect of cholecalciferol on CD69+ CD4+ 17 T cells in food allergy. 18 Conclusions: Cholecalciferol ameliorated food allergy symptom and affected immune reactions in a mouse model of food allergy by reducing CD69+ CD4 + 19 T cells. Anti-CD69 mAb treatment in our 20 food allergy mouse model also induced suppressive effects in the same tendency as our 21 cholecalciferol treatment mouse model. Therefore, we suggest that administration on cholecalciferol 22 can be useful to suppress symptomatic food allergy. 23 (224 words)
