Özet
Amaç: Bu çalışmada insan eritrositlerinden elde edilen 6-fosfoglukonat dehidrogenaz (6PGD) ve glukoz-6-fosfat dehidrogenaz (G6PD) enzimleri üzerine 2-florofeniltiyoüre, 3-florofenil tiyoüre, 4-florofeniltiyoüre, 2,5-diflorofeniltiyoüre, 2,6-diflorofeniltiyoüre bileşiklerinin etkilerini incelemeyi amaçlanmıştır. Yöntem: Bu çalışmada 6-fosfoglukonat dehidrogenaz (6PGD) ve glukoz-6-fosfat dehidrogenaz (G6PD) enzimleri afitinite kromatografisi metoduyla saflaştırıldı. Saflaştırma sonrası enzimlerin aktivitesi spektrofotometrik yöntemle belirlendi. Bulgular: Yapılan çalışmalar sonucunda, insan eritrositlerinden saflaştırılan G6PD ve 6PGD enzimlerini 2-florofeniltiyoüre, 3-florofeniltiyoüre, 4-florofeniltiyoüre, 2,5-diflorofeniltiyoüre, 2,6-diflorofeniltiyoüre bileşiklerinin tamamı yarışmalı olarak inhibe ettiği gözlemlenmiştir. Sonuç: İnhibisyon çalışmalarının sonuçlarına göre, inhibisyon etkisi gözlemlenen bu bileşikler için % Aktivite-[I] grafikleri çizilerek IC50 değerleri bulundu. Ayrıca bu bileşikler için Lineweaver- Burk grafikleri kullanılarak Ki sabitleri ve inhibisyon tipleri belirlendi.

Purpose: In this study, 2-fluorophenyl thiourea, 3-fluorophenyl thiourea, 4-fluorophenyl thiourea, 2,5-difluorophenyl thiourea, 2, 6-difluorophenyl thiourea on 6-phosphogluconate dehydrogenase (6PGD) and glucose-6-phosphate dehydrogenase (G6PD) enzymes obtained from human erythrocytes. It is aimed to examine the effects of difluorophenyl thiourea compounds. Method: In this study,6PGD and G6PD enzymes were purified by affinity chromatography method. Enzymes activity after purification was determined by spectrophotometric method. Findings: As a result of the studies, it was observed that all of the 2-fluorophenyl thiourea, 3-fluorophenyl thiourea, 4-fluorophenyl thiourea, 2,5-difluorophenyl thiourea, 2, 6-difluorophenyl thiourea compounds competitively inhibited the G6PD and 6PGD enzymes purified from human erythrocytes. Results: According to the results of the inhibition studies, IC50 values were found by plotting % Activity-[I] graphs for these compounds whose inhibition effect was observed, and Ki constants and inhibition types were determined for these compounds by using Lineweaver-Burk graphs.
