Özet
Acanthamoeba türleri doğada yaygın olarak bulunan amiplerdir. Bu fırsatçı patojenler, Acanthamoeba Keratitine, Granülomatöz Amibik Ensefalite, Acanthamoeba pnömonisine ve kutanöz acanthamoebiasisine neden olabilir. Acanthamoeba türleri, 18S ribozomal RNA gen dizisi kullanılarak sınıflandırılmış ve günümüze kadar T1-T23 olarak adlandırılan 23 genotipe ayrılmıştır. Bu çalışmada amaç, T4, T5, T11 ve T12 genotipine sahip Acanthamoeba suşlarında genotip grupları arasında, aynı genotipe sahip türler içerisinde ve çevresel ve keratit olgularından izole edilen Acanthamoeba suşlarının osmotolerans ve termotolerans özelliklerini karşılaştırarak patojenlikle ilişkisini değerlendirmektir. Bu çalışmada, klinik ve çevresel örneklerden izole edilmiş T4 (Neff, A, B, D, E), T5, T11 ve T12 genotipine sahip, 22 Acanthamoeba suşunun aksenik kültürleri yapıldıktan sonra termotolerans (37, 39 ve 41 °C) ve osmotolerans (0,5-1 M) testleri yapılmıştır. Sonuç olarak; bütün suşlar hem 37 °C sıcaklıkta hem de 0,5 M osmolaritede üreme yeteneği göstermiştir. Acanthamoeba keratitli hastalardan izole edilen 5 suşun tamamında 37 °C'de ve 0,5 M osmolaritede üreme yeteneği gösterirken, aynı genotipe sahip (T4B ve T4E) klinik suşların üçünde 39-41 °C ve 1 M osmolaritede üreme saptanmamıştır. T4 genotipine sahip Acanthamoeba suşları genel olarak değerlendirildiğinde, 39 °C'de suşların %62,5'inde, 41 °C'de ise %6,25'inde üreme saptanmıştır. Osmotolerans testlerinde ise 1 M osmolaritede %56,25'inde üreme görülmüştür. T5 genotipindeki suşlar 39 °C'de ve 0,5 M mannitolde üreme tespit edilmiştir. T11 genotipine ait 1 suş, bütün sıcaklık derecelerinde ürerken yalnızca 0,5 M osmolaritede üremiştir. T12 genotipindeki suş ise 41 °C'de ve 1 M osmolaritede ürememiştir. Araştırma sonuçlarımıza göre, Acanthamoeba türlerinin özellikle 39 °C ve 1 M mannitole toleransının patojenite göstergesi olmadığı kanısındayız. Bu konunun açıklığa kavuşabilmesi için her genotip grubuna ait daha fazla sayıda Acanthamoeba suşu ile tolerans testlerinin yapılması daha güvenilir sonuçların elde edilmesini sağlayacaktır.

Acanthamoeba species are amoebae commonly found in nature. These opportunistic pathogens can cause Acanthamoeba Keratitis, Granulomatous Amoebic Encephalitis, Acanthamoeba pneumonia and cutaneous acanthamoebiasis. Acanthamoeba species were classified using the 18S ribosomal RNA gene sequence and have been divided into 23 genotypes called T1-T23 to date. The aim of this study is to compare the osmotolerance and thermotolerance characteristics of Acanthamoeba strains isolated from environmental and keratitis cases among genotype groups, within species with the same genotype, and to evaluate the relationship between pathogenicity in Acanthamoeba strains with T4, T5, T11 and T12 genotypes. In this study, thermotolerance (37, 39 and 41°C) and osmotolerance (0,5-1 M) tests were performed after axenic cultures of 22 Acanthamoeba strains with T4 (Neff, A, B, D, E), T5, T11 and T12 genotypes isolated from clinical and environmental samples. As a result; All strains showed growth ability at both 37 °C and 0,5 M osmolarity. While all of the 5 strains isolated from patients with Acanthamoeba keratitis showed growth ability at 37 °C and 0,5 M osmolarity, no growth was detected at 39-41 °C and 1 M osmolarity in three of the clinical strains with the same genotype (T4B and T4E). When Acanthamoeba strains with T4 genotype were evaluated in general, growth was found in 62,5% of the strains at 39 °C and 6,25% at 41 °C. In osmotolerance tests, growth was observed in 56,25% of them at 1 M osmolarity. T5 genotype strains were found to grow at 39 °C and 0,5 M mannitol. A strain of the T11 genotype grew at all temperatures, but only at 0,5 M osmolarity. T12 genotype strain did not grow at 41 °C and 1 M osmolarity. According to our research results, we believe that the tolerance of Acanthamoeba species especially to 39 °C and 1 M mannitole is not an indicator of pathogenicity. In order to clarify this issue, performing tolerance tests with more Acanthamoeba strains belonging to each genotype group will provide more reliable results.
