The Tenebrio molitor larvae, also known as the mealworm, have various bioactive compounds with anti-obesity, anticancer and antioxidant properties. This study investigated and compared the effects of the fermented Tenebrio molitorlarvae extract (TMP) with casein (CA) or soy protein (SP), as a dietary protein source in C57BLKS/J-db/db mice. The glycemic control of the proteins was compared with metformin (Met) (0.5% in diet), a hypoglycemic drug. Mice were fed CA, SP, and TMP, respectively, comprising 20% of the AIN-76 diet for six weeks. Food intake was significantly higher in the TMP group than in the CA and SP groups; however, the body weight change in the TMP group was not significantly different from the others. Met significantly improved fasting blood glucose and oral glucose tolerance in the mice, but SP and TMP had no effect when compared with the CA group. TMP marginally lowered serum insulin and leptin levels compared with CA. vi TMP and SP significantly lowered hepatic triglyceride levels and free fatty acid content as well as lipid droplets compared to the Met group. The improvement in these parameters in the TMP and SP groups were marginal as compared to the CA group. TMP lowered serum aspartate transaminase (AST) and tended to lower alanine transaminase (ALT) levels compared to the CA and SP groups. Proteomic analysis showed that 14 proteins (H4C1, CYB5A, AnxA5, RGN, 40S ribosomal protein, Atp2, PDI, GRP78, RDX, GCH1, LDHA, GNMT, ACAA2, and UROC1) were up-regulated and 8 proteins (UQCRC1, Txndc5, SBP2, HSPA8, PHB, TF, PC, and PK) were down-regulated in the CA group compared to the nondiabetic control group. However, TMP significantly down-regulated AnxA5, RGN, 40S ribosomal protein, Atp2, PDI, RDX, LDHA, ACAA2, and UROC1, and up-regulated UQCRC1, TF, and PK. These proteins are related to glycolysis, lactate production, fatty acid β-oxidation, oxidative stress, and endoplasmic reticulum stress. These results suggest that TMP is a potential protein source that can replace CA or SP in type 2 diabetes.
